Transcriptome approach to understand the potential mechanisms of resistant and susceptible alfalfa(Medicago sativa L.) cultivars in response to aphid feeding

plant breeding for resistance to agricultural pests is an essential element in the development of integrated crop management systems, however, the molecular and genetic mechanisms underlying resistance are poorly understood. In this pilot study, we conducted a transcriptomic analysis of a resistant （R） and susceptible （S） variety of alfalfa, with （＋A） or without （-A） aphids （totally four treatments）. We used the resistant cultivar Zhongmu t and the susceptible cultivar Soca. A total of 3 549 mRNAs were differentially expressed, of which 1 738 up-regulated and 1 307 down-regulated genes were identified in S＋A/S-A plants, while 543 up-regulated and 331 down-regulated genes were identified in the R＋A/R-A plants. KEGG analysis mapped 112 and 546 differentially expressed genes to 8 and 17 substantially enriched pathways for Zhongmu 1 and Soca, respectively. Six shared pathways were linked to plant resistance traits, including phenylpropanoid biosynthesis associated with salicylic acid synthesis, and linoleic acid metabolism associated with both jasmonic acid and flavonoid biosynthesis. Ultimately, we proposed a preliminary regulatory mechanism of alfalfa cultivar resistance response to aphids feeding based on transcriptome analyses and published documents.

Molecular Taxonomy of Conogethes punctiferalis and Conogethes pinicolalis(Lepidoptera: Crambidae) Based on Mitochondrial DNA Sequences

Conogethes punctiferalis（Guenée）（Lepidoptera： Crambidae） was originally considered as one species with fruit-feeding type（FFT） and pinaceae-feeding type（PFT）, but it has subsequently been divided into two different species of Conogethes punctiferalis and Conogethes pinicolalis. The relationship between the two species was investigated by phylogenetic reconstruction using maximum-likelihood（ML） parameter estimations. The phylogenetic tree and network were constructed based upon sequence data from concatenation of three genes of mitochondrial cytochrome c oxidase subunits I, II and cytochrome b which were derived from 118 samples of C. punctiferalis and 24 samples of C. pinicolalis. The phylogenetic tree and network showed that conspecific sequences were clustering together despite intraspecific variability. Here we report the results of a combined analysis of mitochondrial DNA sequences from three genes and morphological data representing powerful evidence that C. pinicolalisand C. punctiferalis are significantly different.

Effects of Glutamate and Na^+ on the Development and Enzyme Activity of the Oriental Migratory Locust,Locusta migratoria manilensis(Meyen) in Successive Generations

Rapid and mass rearing ofLocusta migratoria manilensis is an urgent need to meet the increasing demand for food of people. In this study, the effects of four artificial feeds on the development, reproduction and the activities of detoxification and protective enzymes of L. migratoria manilensis in three successive generations were investigated. The results showed that sucrose and monosodium glutamate （MSG） significantly increased the net reproductive rate （R0） and the intrinsic growth rate （rm） ofL. migratoria manilensis, but sodium chloride （0.17%） suppressed this increase. Furthermore, the artificial feed with sucrose and monosodium glutamate increased the activities of esterase （EST）, acetylcholinesterase （ACHE）, glutathione-S- transferase （GST）, multi-function oxidase （MFO）, phenol oxidase （PO）, catalase （CAT） and peroxidase （POD）, but inhibited the activity of superoxide dismutase （SOD）. However, sodium chloride （0.17%） increased the activities of EST, ACHE, CAT and SOD, and inhibited the activities of MFO, GST, PO and POD. Correlation analysis found that the increasing of PO activity and the decreasing of SOD activities were significantly related with the increasing of the intrinsic growth rate （rm）. The above results indicated that sucrose and monosodium glutamate could promote the development and reproduction of L. migratoria manilensis, but Na＋ inhibit such promotion with the concentration above 0.2%. The activities of PO and SOD can be used as biochemical standard to assess the effect of artificial feed.

Nicotinic Acetylcholine Receptor Gene Family of the Pea Aphid,Acyrthosiphon pisum

The nicotinic acetylcholine receptors （nAchRs） are cholinergic receptors that form ligand-gated ion channels by ifve subunits in insect and vertebrate nervous systems. The insect nAChR is the molecular target of a class of insecticides, neonicotinoids. Here, we identiifed and cloned 11 candidate nAChR subunit genes in Acyrthosiphon pisum using genome-based bioinformatics combined modern molecular techniques. Most A. pisum nAChRs including α1, α2, α3, α4, α6, α8, and β1 show highly sequence identities with the counterparts of other insects examined. Expression proifles analysis showed that all subunit genes were expressed in adult head. At least two subunits have alternative splicing that obviously increase A. pisum nicotinic receptor diversity. This study will be invaluable for exploring the molecular mechanisms of neonicotinoid-like insecticides in sucking pests, and for ultimately establishing the screening platform of novel insecticides.

Genomic Analysis of the Natural Population of Wheat dwarf virus in Wheat from China and Hungary

During the last decade, the leafhopper transmitted Wheat dwarf virus （WDV） has become a serious problem both in northwestern China and Hungary. In order to study the molecular diversity and population structure of WDV in these two countries, 39 Chinese isolates and 16 Hungarian isolates were collected from different regions of China and Hungary, and their genomes were sequenced. all isolates belonged to the wheat strain of WDV and showed limited genetic diversity. The highest and lowest nucleotide sequence identities among isolates from China and Hungary were 99.9 and 90%, respectively. In all isolates, the lowest nucleotide sequence identity was 89.5% between MO10-1 and KP10-5, which were collected from Martonv^is^r and Kompolt, Hungary. Phylogenetic analyses showed the genome sequences of 55 WDV isolates belong to two big clades, but no clear correlation to geographical location. Population difference analyses indicated that the Chinese and Hungarian WDV populations have no significant difference. The regions in WDV genome with relatively low nucleotide diversities represented protein coding regions suggested that these regions evolved under negative selection, and might be one of the causes restricting the number of genetic variants.

Spectral sensitivity of the compound eyes of Anomala corpulenta motschulsky(Coleoptera:Scarabaeoidea)

The scarab beetle, Anomala corpulenta Motschulsky（Coleoptera： Scarabaeoidea）, is a widespread and destructive pest in China. Vision is one of the most important means of acquiring information about the external environment. In order to contribute to the understanding of the perception of visual stimuli in this species, the light sensitivity and spectral responses of the scarab beetle, A. corpulenta, were measured by using an electroretinogram（ERG） technique. In total, 14 monochromatic light intensities, between 340 and 605 nm, were applied to the compound eyes of A. corpulenta under varying levels of adaptation to dark and light conditions. The results showed that all light stimuli induced an ERG response, with varied amplitudes. The spectral sensitivity curve of dark-adapted eyes showed one major peak（~400 nm; near-ultraviolet）, a secondary peak（from 498 to 562 nm; yellow-green） and the third peakat 460 nm. By contrast, in light-adapted eyes, only a near-UV peak was observed. From these results, we conclude that the compound eye of A. corpulenta is likely to have at least three spectral types of photoreceptor. Significance of differences were also recorded in the responses of male and female compound eyes, as well as diurnally and nocturnally. The amplitude of ERG in response to white-light stimuli varied with the light intensity： The stronger the luminance, the higher the ERG value. This suggests that the compound eye of A. corpulenta adapts quickly to changing light conditions, enabling A. corpulenta to maintain nocturnal activities.

Characterization of two novel heat shock protein 70s and their transcriptional expression patterns in response to thermal stress in adult of Frankliniella occidentalis (Thysanoptera:Thripidae)

Heat shock protein 70（HSP70） is one of the most important members in the heat shock protein family, and plays important roles in the thermotolerance of insect. To explore the molecular mechanism of thermotolerance of Frankliniella occidentalis adults, the difference in the expression of HSP70s in F. occidentalis male or female adults under the thermal stress was studied under the laboratory conditions. Two full length c DNAs of HSP70s gene（Fohsc704 and Fohsc705） were cloned from F. occidentalis by using RT-PCR and RACE. The genomic sequence was demonstrated by genomic validation, and the position and size of the intron were analyzed by sequence analysis of c DNA. Real-time PCR was used to analyze the HSP70 expression patterns. The c DNA of Fohsc704 and Fohsc705 possessed 2 073 and 1 476 bp which encoded 690 and 491 amino acids（aa） with a calculated molecular weight of 75 and 54 k Da, respectively. Four introns in Fohsc704 and six introns in Fohsc705 protein were found. However, the HSP70 protein sequences in our study were ended with EKKN and GIFL, which were different from the reported Fo HSP70s. Various expression patterns of Fohsc704 and Fohsc705 were found in both genders of F. occidentalis under thermal stress. The expression of Fohsc704 and Fohsc705 reached to the highest level at –12 and –8°C in male adults, respectively, and Fohsc705 expressed the highest level at 33°C in female adults. In conclusion, HSP70s of F. occidentalis in our study are novel heat shock proteins. There were difference in expression patterns of the two hsc70s in genders of F. occidentalis, and the two HSP70s play important roles in the thermotolerance of F. occidentalis.

Selective and stable elimination of endosymbionts from multiple-infected whitefly Bemisia tabaci by feeding on a cotton plant cultured in antibiotic solutions
