Development of a multiplex reverse transcription-PCR assay for simultaneous detection of garlic viruses

A preliminary screening for garlic viruses in garlic plants in Hunan,China,using existing monoplex（simplex） reverse transcription-polymerase chain reaction（RT-PCR） procedures detected four viruses/virus *** viruses/virus groups were Onion yellow dwarf virus（OYDV）,Leek yellow stripe virus（LYSV）,Shallot latent virus（SLV） and allexiviruses（e.g.,garlic viruses A,B,C,D,E,X）.Sequence analysis of the projected allexivirus amplicons revealed the allexivirus in the infected garlic plants was Garlic virus D（GarV-D）,which shared 92-97%sequence identities with various isolates from the world.A multiplex RT-PCR（mRT-PCR） was therefore developed to simultaneously detect and differentiate the four viruses/virus *** achieve this,four primer pairs targeting allexiviruses,OYDV,LYSV and SLV were *** anticipated amplicon sizes are 183 bp（allexiviruses）,265 bp（OYDV）,404 bp（LYSV） and 592 bp（SLV）,*** primer pairs produced virus-specific fragments in both simplex and multiplex formats,thus confirming the efficacy of the newly developed mRT-PCR for detection of these *** mRT-PCR further was evaluated by applying it to garlic plant samples collected in two geographic locations in ***,OYDV,LYSV and SLV were detected in 50.9,40.3,28.3 and 58.5%of leaf samples,respectively;and mixed infections with two or more viruses accounted for 54%of the garlic *** results obtained by mRT-PCR were confirmed by simplex RT-PCR *** conclusion,this newly developed mRT-PCR provides a rapid,sensitive and reliable method for the detection and identification of major garlic viruses.

Use of transcriptome sequencing to explore the effect of CSRP3 on chicken myoblasts

The mechanisms that regulate the specificity and maintenance of chicken muscle fiber types remain largely unknown. In mammals, CSRP3 has been shown to play a vital role in the maintenance of typical muscle structure and function. This study investigated the role that CSRP3 plays in chicken skeletal muscle. First, the antibody against chicken CSRP3 protein was prepared, and the expression levels of the mRNA and protein of the CSRP3 gene in four chicken skeletal muscles with different myofiber compositions were compared. Then the effects of CSRP3 silencing on the expression profile of chicken myoblast transcriptomes were analyzed. The results showed that the expression levels of the mRNA and protein of the CSRP3 gene were both associated with the composition of fiber types in chicken skeletal muscles. A total of 650 genes with at least 1.5-fold differences(Qchain genes(MyH1B and MyH1E)were upregulated(Q<0.05) upon CSRP3 silencing. These results suggested that CSRP3 plays a crucial role in chicken myofiber composition, and affects the distribution of chicken myofiber types, probably by regulating the expression of MyH1B and MyH1E.

Since the rapid spread of the Asian chestnut gall wasp(ACGW) throughout south-east Europe in the last few years,the possibilities of its control have been increasingly *** to constraints in available suppression measures in forest stands,biological control is recognized as the most suitable action to lower the abundance of the new invasive *** sinensis,as a specialized parasitoid chalcid wasp of ACGW,was introduced to suppress the pest in Italy in 2005,and later in Croatia,Hungary(2014) and Slovenia(2015).We investigated the native parasitoid complex associated with ACGW in Slovenia,Croatia and Hungary over 8 years of *** found 41 species of native parasitoids,eight of which are the first records on ACGW in Europe,adding up to a total of 51 species known to parasitize *** findings represent a large complex of cynipid-parasitoid ***,the abundance of native parasitoid populations on ACGW have demonstrated a temporal surge between the invasion of a new alien host(ACGW) and the onset of its specialized parasitoid,*** results indicate that the introduced parasitoid acts as a very successful biocontrol of ACGW,outcompeting native *** outcome should be considered as beneficial through the maintenance of ecological balance in affected forest ecosystems.

Factors Affecting Teff (Eragrostis tef) Market Supply in Woliso and Becho Districts of South West Shoa Zone Oromia Regional State, Ethiopia

This study was aimed to analyze teff (Eragrostis tef) market chain in south west Shoa zone with objective of factors affecting teff market supply using two stage ordinary least square approaches. The majority of Ethiopia’s population earns its livelihood primarily from agriculture. Cereals teff is the first in Ethiopia area coverage and production. Teff (Eragrostis tef) is a major staple food crop in Ethiopia. Both primary and secondary data were used in this study. Primary data was collected from 138 sampled farmers and 38 traders from both districts by using semi-structured interview. The OLS (ordinary least square) model results showed that seven explanatory variables significantly affected the quantity of teff supplied to the market supplied by smallholder producers. Age, education level and current market price were negatively and significantly affecting teff market supply. Distance to the nearest market, farm size, perception and quantity produced were positively and significantly influencing marketed supply of teff. Policy implications that were to take place highly recommendation those are relevant to improve teff marketing system in the study area which indicated production and market orientation were set based on the significant variables and raised problems by the stakeholders. To improve market supply of teff in the study area resolving the prevailing production problems deems a necessary condition.

Nondestructive determination of the freshness change in bighead carp heads under variable temperatures by using excitation-emission matrix fl uorescence and back-propagation neural networks

This study established back-propagation neural networks(BPNNs)for evaluating the freshness of bighead carp(Hypophthalmichthys nobilis)heads during chilled storage via fluorescence spectroscopy using an excitation-emission matrix(EEM).The total volatile basic nitrogen(TVB-N)and total aerobic count(TAC)of fish increased obviously during storage at 0,4,8,12,and 16°C,while sensory scores decreased with increasing storage *** EEM fluorescence intensity was measured,and its change was correlated with the freshness indicators of the *** characteristic components of EEM data were extracted by parallel factor analysis,and two freshness indicators were used to construct the EEM-BPNNs *** results demonstrated that the relative errors of the EEM-BPNNs model for TVB-N and TAC were less than 14%.This result indicated that the EEM-BPNNs model could determine the freshness of fish in cold chains in a rapid and nondestructive way.

Using benzene carboxylic acids to prepare zirconium-based catalysts for the conversion of biomass-derived furfural

Benzene carboxylic acid (BCAs) are common and useful chemical blocks, which can be derived from the abundant low rank coals (LRCs) via oxidative degradation. In this work, we proposed a novel strategy to utilize BCAs as raw materials to prepare catalysts with transition metal zirconium, and the prepared catalysts were applied into the conversion of the renewable biomass resources. Typical model BCAs in the oxidative products of LRCs, including pyromellitic acid, trimesic acid (TMSA), trimellitic acid, and benzoic acid, were used as the block to construct the Zr- BCAs catalysts. The chemoselective conversion of furfural into furfuryl alcohol (FAL). an important reaction in the biomass conversion chain, is chosen to evaluate the activity of the catalysts. The preparation conditions of the catalysts and experiment factors during the reaction were systematically investigated. The prepared catalysts were characterized by SEM, TEM, XRD and TG-DTG. The results showed that the prepared catalysts were efficient for the conversion of furfural into FAL, among which Zr-TMSA gave the highest activity. Zr-TMSA could be recycled for ten times without obvious deactivation, indicating an excellent stability. The strategy proposed in this work may be beneficial for the value-added utilization of both LRCs and biomass resources.

A new grey predictionmodel for forecasting the automobiles ownership in China

Pomegranate peel polyphenols improves diet-induced obesity and promotes adipose browning development through gut microbiota in mice

Gut microbiota regulate the activation of adipose browning， which promote energy dissipation and combat diet-induced obesity. Pomegranate peel polyphenols (PPPs) have been shown to reduce obesity， regulate lipid metabolism in adipose tissue， and modulate the composition of gut microbiota in animal fed high-fat diet (HFD). However， the role of gut microbiota in the improvement of obesity by PPPs has not been elucidated. In current study， we applied antibiotics to inhibit gut microbiota in mice fed HFD and treated with PPPs. The results showed that the inhibition of gut microbiota impair the effect of PPPs on reducing obesity and promoting adipose browning， and change the fecal metabolomic profiles in respond to PPPs. Moreover， the inhibition of gut microbiota supressed the promotive effects of PPPs on the levels of Akkermansia and microbiota-related metabolites， such as urolithin A， short-chain fatty acids (SCFAs)， and bile acids (BAs)， which were associated with activating adipose browning. Therefore， our results suggested that the presence of gut microbiota is essential for PPPs to ameliorate HFD-induced obesity. The related bacteria or metabolites generated by the interaction between PPPs and microbiota promote adipose browning and facilitate the beneficial effects of PPPs.

Antimicrobial usage and resistance in beef production

Antimicrobials are critical to contemporary high-intensity beef production. Many different antimicrobials are approved for beef cattle, and are used judiciously for animal welfare, and controversially, to promote growth and feed efficiency. Antimicrobial administration provides a powerful selective pressure that acts on the microbial community, selecting for resistance gene determinants and antimicrobial-resistant bacteria resident in the bovine flora. The bovine microbiota includes many harmless bacteria, but also opportunistic pathogens that may acquire and propagate resistance genes within the microbial community via horizontal gene transfer. Antimicrobial-resistant bovine pathogens can also complicate the prevention and treatment of infectious diseases in beef feedlots,threatening the efficiency of the beef production system. Likewise, the transmission of antimicrobial resistance genes to bovine-associated human pathogens is a potential public health concern. This review outlines current antimicrobial use practices pertaining to beef production, and explores the frequency of antimicrobial resistance in major bovine pathogens. The effect of antimicrobials on the composition of the bovine microbiota is examined, as are the effects on the beef production resistome. Antimicrobial resistance is further explored within the context of the wider beef production continuum, with emphasis on antimicrobial resistance genes in the food chain, and risk to the human population.
