A high-precision multi-dimensional microspectroscopic technique for morphological and properties analysis of cancer cell

Raman and Brillouin scattering are sensitive approaches to detect chemical composition and mechanical elasticity pathology of cells in cancer development and their medical treatment *** application is,however,suffering from the lack of ability to synchronously acquire the scattering signals following three-dimensional(3D)cell morphology with reasonable spatial resolution and signal-to-noise ***,we propose a divided-aperture laser differential confocal 3D Geometry-Raman-Brillouin microscopic detection technology,by which reflection,Raman,and Brillouin scattering signals are simultaneously in situ collected in real time with an axial focusing accuracy up to 1 nm,in the height range of 200μ*** divided aperture improves the anti-noise capability of the system,and the noise influence depth of Raman detection reduces by 35.4%,and the Brillouin extinction ratio increases by 22 dB.A high-precision multichannel microspectroscopic system containing these functions is developed,which is utilized to study gastric cancer *** a result,a 25%reduction of collagen concentration,42%increase of DNA substances,17%and 9%decrease in viscosity and elasticity are finely resolved from the 3D *** findings indicate that our system can be a powerful tool to study cancer development new therapies at the sub-cell level.

Correlation of serum trace element changes with inflammatory cytokines and oxidative stress indexes in patients with urinary tract infection

Objective: To study the correlation of serum trace element changes with inflammatory cytokines and oxidative stress indexes in patients with urinary tract infection (UTI). Methods: The patients who were diagnosed with lower urinary tract infection in our hospital between March 2013 and December 2017 were selected as the UTI group and the healthy volunteers who received physical examination were selected as the control group. Serum was collected to measure the contents of trace elements and inflammatory cytokines, and urine was collected to measure the contents of oxidative stress indexes. Results: The content of Zinc (Zn) in serum of the UTI group was significantly lower than that of the control group (P0.05);the contents of C-reactive protein (CRP), amyloid protein A (SAA), procalcitonin (PCT), intercellular adhesion molecule-1 (ICAM-1), interleukin-2 (IL-2), interleukin-4 (IL-4), interleukin-5(IL-5), interleukin-17(IL-17) and transforming growth factor-β1 (TGF-β1) in serum as well as the contents of malondialdehyde (MDA) and advanced oxidation protein product (AOPP) in urine of the UTI group were significantly higher than those of the control group and negatively correlated with the content of Zn in serum (r=-0.514, -0.573, -0.475, -0.636, -0.612, -0.585,-0.492, -0.537, -0.611, -0.476 and -0.542, p=0.015, 0.011, 0.024, 0.007, 0.009, 0.010, 0.020, 0.014, 0.009, 0.024 and 0.013) while the contents of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT) in urine were significantly lower than those of the control group and positively correlated with the content of Zn in serum (r=0.592, 0.657 and 0.610, p=0.010, 0.002 and 0.009). Conclusion: Serum trace element Zn significantly decreases in patients with urinary tract infection and it is closely related to the occurrence of disease as well

Efficiency and safety evaluation of Baimai ointment for alleviating hypermyotonia following cerebral infarction

Baimai ointment has Shujin and Huoluo efficiency. It has been confirmed by basic pharmacodynamic experiments that Baimai ointment has some therapeutic effects on stroke. However, the detailed evaluations of Baimai ointment on clinically treating stroke patients for function recovery are still insufficient with little reports. OBJECTIVE： To observe the efficiency and safety of Baimai ointment on alleviating hypermyotonia following cerebral infarction DESIGN： A randomized, parallel and controlled experiment. SETTING： Department of Neurology, Beijing aerospace General hospital. PARTICIPANTS： A total of 120 patients with acute front-cycle cerebral infarction were selected from Department of Neurology, Beijing aerospace General hospital from July to December 2006 and met the diagnosis criteria modified by Forth National Seminar on Cerebrovascular Disease. The experiment had got confirmed consent from local ethic committee. All 120 patients with cerebral infarction were randomly divided into experimental group （n =80; 32 in the acute phase, 32 in the recovery phase and 16 in the sequela phase） and control group （n =40; 16 in the acute phase, 16 in the recovery phase and 8 in the sequela phase）. METHODS： Within 4 weeks after the patients were enrolled in this study, no other traditional Chinese medicines containing similar components were adopted besides routine supporting therapy. The Baimai ointment being consisted of jianghuang, Roudoukou, Gansong, Yangqishi, Gancao, She, shannai, Zanghuixiang, Zangchangpu, Huajiao and dianhua was used by trial group （20 g/ampoule, Tibet Lingzhiqizheng Tibetan Drug Factory; batch number： 050403）, 2-3 times per day, the ointment was moderately coated traveling parts of Baimai. MAIN OUTCOME MEASURES： The evaluation of National Institutes of Health Stroke Scale （NIHSS）, Modified Rankin Scale and Modified Ashworth Scale were performed before administration, 2 and 4 weeks after administration. Recent advances in the dark adaptation investigations

Dark adaptation is a highly sensitive neural function and may be the first symptom of many status including the physiologic and pathologic entity, suggesting that it could be instrumental for diagnose. However, shortcomings such as the lack of standardized parameters, the long duration of examination, and subjective randomness would substantially impede the use of dark adaptation in clinical work. In this review we summarize the recent research about the dark adaptation, including two visual cycles-canonical and cone-specific visual cycle, affecting factors and the methods for measuring dark adaptation. In the opinions of authors, intensive investigations are needed to be done for the widely use of this significant visual function in clinic.

Clinical and pathological portraits of axillary presentation breast cancer and effects of preoperative systemic therapy

There is a lack of investigation into the biological characteristics and preoperative systemic therapy （PST） for occult breast cancer （OBC）. For this study, departmental records in Breast Disease Center of Peking University First hospital from January 2008 to December 2015 were retrospectively reviewed to identify cases of OBC. Eleven cases were included, and all patients were female, with a median age of 56 （range： 29-75） years. The sensitivity of magnetic resonance imaging （MRD was I00%, and the false positive rate was 33.3%. Based on histologic analysis of the axillary node, 9 （81.8%） cases were grade 3, and 2 （18.2%） cases were grade 2; 4 （36.4%） cases were 〉10% estrogen receptor （ER） positive and 6 （54.5%） human epidermal growth receptor 2 （HER2） positive. Nine cases （81.8%） exhibited over 30% Ki67 expression. PST was performed in 5 of the 11 cases. The lymph node response rate was 100% （5/5）, but no complete remission was achieved. In conclusion, aggressive subtypes were predominant among the included cases, and PST should be considered for OBC treatment options.

Gene editing in T cell therapy

The adoptive transfer of engineered T cells for the treatment of cancer, autoimmunity, and infectious disease is a rapidly growing field that has shown great promise. Gene editing holds tremendous potential for further improvements of T cell therapy. Here we review the applications of gene editing in various T cell therapies, focusing on antiviral strategies and cancer immunotherapies, and discuss the challenges and future prospects.

Validation and modification of the AJCC 8th TNM staging system for pancreatic ductal adenocarcinoma in a Chinese cohort:A nationwide pancreas data center analysis

Objective:To validate the 8 th edition of the American Joint Committee on Cancer(AJCC)staging system for pancreatic ductal adenocarcinoma(PDAC)in a Chinese cohort of radically resected patients and to develop a refined staging system for ***:Data were collected from the China Pancreas Data Center(CPDC)for patients with resected PDAC in 2016 and 2017,and cancer-specific survival(CSS)was evaluated using the Kaplan-Meier method and log-rank *** and multivariate analyses based on Cox regression were performed to identify prognostic *** recursive partitioning analysis(RPA),Kaplan-Meier method,and log-rank test were performed on the training dataset to generate a proposed modification for the 8 th TNM staging system utilizing the preoperative carbohydrate antigen(CA)19-9 *** was performed for both staging systems in the validation ***:A total of 1,676 PDAC patients were retrieved,and the median CSS was significantly different between the 8 th TNM groupings,with no significant difference in survival between stage IB and *** analysis of T and N stages demonstrated a better prognostic value in the N *** analysis showed that the preoperative serum CA19-9 level was the strongest prognostic indicator among all the independent risk *** patients with CA19-9>500 U/mL had similar survival,and we proposed a new staging system by combining IB and IIA and stratifying all patients with high CA19-9 into stage *** modified staging system had a better performance for predicting CSS than the 8 th AJCC staging ***:The 8 th AJCC staging system for PDAC is suitable for a Chinese cohort of resected patients,and the N category has a better prognostic value than the T *** modified staging system has superior accuracy in predicting survival than the 8 th AJCC TNM staging system.

Four-year dynamic observation and study on standardized management of elderly patients with type 2 diabetes in Beijing Yongding Road Community

Purpose:This study aimed to understand the joint standard of blood glucose,blood li-pids,and blood pressure in elderly patients>60 years of age with type 2 diabetes in the Beijing Yongding Road ***:One hundred ninety-two elderly patients>60 years of age who were diagnosed with type 2 diabetes in Yongding Road Community served as the investigation subjects,under-went standard management,and the dynamic changes in blood glucose,blood lipids,and blood pressure were monitored for 48 ***:At the end of the observation period,the standard rates of fasting blood glucose and postprandial blood glucose were 72.55%and 80.00%,respectively,which were increased compared with 55.73%and 56.08%at baseline(P0.05).The standard rates of TG and LDL-C were 76.71%and 60.38%,respectively,which were increased compared with 54.69%and 34.74%at base-line,and the standard rate of HDL-C was 13.64%,which was decreased compared with 40.10%at baseline(P<0.01).The standard rate of BP was 58.33%,which was increased compared with 38.54%at baseline(P<0.01).The optimal control rate of blood glucose for 48 months(the standard times of the total measurement times in 48 months≥75%)was higher;the fasting blood glucose was 52.17%,the 2-h postprandial blood glucose was 60.22%,and the HbA1c was 46.45%.The optimal control rate of blood lipids was lower;LDL-C was 17.49%and HDL-C was 13.59%.The optimal control rate of BP was 9.13%.At the end of the observation period,the levels of fasting blood glucose and postprandial blood glucose were decreased by 0.7 mmol/L and 1.48 mmol/L,respectively,compared with the base-line(P<0.01).The level of HbA1c was decreased by 0.18%compared with the baseline(P<0.05).The levels of LDL-C and HDL-C were decreased by 0.4 mmol/L and 0.23 mmol/L,respectively,compared with the baseline(P<0.01).The levels of SBP and DBP were decreased by 4 mmHg compared with the bas

The interactions between bloodstream and vascular structure on aortic dissecting aneurysmal model: A numerical study

Stent-graft implantation is an important means of clinical treatment for aortic dissecting aneurysm （ADA）. However, researches on fluid dynamics effects of stent were rare. Computer simulation was used to investigate the interactions between bloodstream and vascular structure in a stented ADA, which endures the periodic pulse velocity and pressure. We obtained and analyzed the flow velocity distribution, the wall displacement and wall stress in the ADA. By comparing the different results between a non-stented and a stented ADA, we found that the insertion of a vascular graft can make the location of maximum stress and displacement move from the aneurysm lumen wall to the artery wall, accompanied with a greatly decrease in value. These results imply that the placement of a stent-graft of any kind to oc-clude ADA will result in a decreased chance of rupture.

AIM：To investigate the effects of hydrogen-rich saline（HRS）on microglia activation and Sirtuin type 1（Sirt1）in rats with N-methyl-N-nitrosourea（MNU）-induced retinitis pigmentosa（RP）.METHODS：Rats were divided into norm（N）group,model（M）group and HRS（H）*** in M and H groups were given saline and HRS respectively prior to and after administration of *** one day（d1）and d3 afterwards,electroretinogram and histological examination were performed to confirm the effects of HRS on retinal function and structure of MNU-induced *** staining of anti-ionized calcium-binding adapter molecule 1（Iba1）,a maker of microglia cells,was performed,with quantitative real-time polymerase chain reaction（qRT-PCR）for its m RNA ***,Sirt1 m RNA and protein expression in the retinas were detected by Western blot and ***：HRS preserved the retinal function and mitigated the reduction of photoreceptor degeneration in MNU-treated *** presence of microglia cells was somewhat more obvious in H group than that in M group at *** suppressed the further activation of microglia cells,with the number of microglia cells less than that of M group at *** of qRT-PCR of Iba1 were consistent with those of immunofluorescence staining,with the m RNA expression of Iba1 in H group more intensive than that of M group at d1（P〈0.05）,while less than that of M group at d3（P〈0.05）.Furthermore,the Sirt1 m RNA and protein expression decreased after MNU administration,while HRS mitigated the MNU-induced downregulation of ***：HRS can effectively keep microglia activation induced by MNU to an appropriate extent,while upregulate Sirt1 in MNU-induced RP.
