Adenosine triphosphate mediates the pain tolerance effect of manual acupuncture at Zusanli(ST36) in mice

OBJECTIVE:To investigate the mechanisms behind the effects of acupuncture in Traditional Chinese medicine,we delved into the adenosine triphosphate/peripheral purinergic P2X receptor 3(ATP/P2X3)receptor signaling system as an indicator of the body's energy state,commonly referred to as"Qi".METHODS:The tail-flick test was utilized to explore the impact of acupuncture on pain tolerance threshold(PTT)in mice,while also assessing adenosine(ADO)levels and adenylate energy charge(EC)at Zusanli(ST36).The study further investigated the dose-dependent effects of acupuncture on PTT and ADO levels at Zusanli(ST36).To shed light on the underlying mechanisms of acupuncture's effects,the study examined the impact of ATP,a P2X3 receptor antagonist,and adenosine disodium on PTT following acupuncture ***:acupuncture at Zusanli(ST36)led to significant improvements in PTT in mice,with the most effective interventions being twirling for 2 min and needle retention for 28 *** interventions also resulted in significant increases in ATP *** effects of acupuncture were further augmented by administration of different doses of ATP at Zusanli(ST36),and pretreatment with a P2X3 receptor antagonist decreased *** EC peaked at 30 min after intraperitoneal injection of ATP,and pretreatment with various doses of *** 30 min prior to acupuncture increased PTT in a dose-dependent ***,pretreatment with an *** intramuscular injection of adenosine disodium enhanced the effects of ***:This research provides compelling evidence that ATP is involved in the regulation of PTT through acupuncture,revealing new avenues for achieving enhanced clinical outcomes.

Objective To explore the demographic and disease-related factors associated with acupuncture response in patients with chronic tension-type headache(CTTH).Methods Using data from a randomized clinical trial(218 cases)consisting of 4 weeks of baseline assessment,8 weeks of treatment,and 24 weeks of follow-up,participants were regrouped into responders(at least a 50%reduction in monthly headache days at week 16 compared with baseline)and ***-three demographic and disease-related factors associated with acupuncture response in 183 participants were analyzed by multivariable logistic *** One hundred and nineteen(65.0%)participants were classified as *** factors were significantly independently associated with acupuncture response,including treatment assignment,headache intensity at baseline,and 2 domains[general health(GH)and social functioning(SF)]from the 36-Item Short Form Health Survey quality of life *** assignment was associated with non-response:participants receiving true acupuncture were 3-time more likely to achieve a CTTH response than those receiving superficial acupuncture[odds ratio(OR)0.322,95%confidence interval(CI)0.162 to 0.625,P=0.001].Compared with patients with mild-intensity headache,patients with moderate-intensity headache were twice as likely to respond to acupuncture(OR 2.001,95%CI 1.020 to 4.011,P=0.046).The likelihood of non-response increased by 4.5%with each unit increase in the GH grade(OR 0.955,95%CI 0.917 to 0.993,P=0.024)while decreased by 3.8%with each unit increase in the SF grade(OR 1.038,95%CI 1.009 to 1.069,P=0.011).Conclusions Greater headache intensity,lower GH score,and higher SF score were associated with better acupuncture responses in CTTH *** 3 factors require independent validation as predictors of acupuncture effectiveness in CTTH.

Regulatory role of electroacupuncture on satellite glial cell activity in the colon and dorsal root ganglion of rats with irritable bowel syndrome

OBJECTIVE: To investigate the role of satellite glial cells in irritable bowel syndrome(IBS) and the effect of electroacupuncture(EA) at the Tianshu(ST25) and Shangjuxu(ST37) combination. METHODS: A model for visceral hypersensitivity in IBS was induced through colorectal distension(CRD) stimulation. Clean-grade male Sprague-Dawley(SD) rats were randomly divided into four groups: a normal group(NG), a model group(MG), an electroacupuncture group(EA), and a glial cell inhibitor group(FCA). Bilateral EA(2/100 Hz, 1 mA, 30 min) was administered at the Tianshu(ST25) and Shangjuxu(ST37) in week 6. Abdominal withdrawal reflex(AWR) scores were used to assess the behavioral response associated with visceral hyperalgesia, while hematoxylin-eosin staining was employed to evaluate pathological changes in the colon. The protein and mRNA levels of glial fibrillary acidic protein(GFAP) in the colon and colon-related dorsal root ganglion(DRG) were analyzed using immun-ofluorescence, immunohistochemistry, Western blotting, real-time polymerase chain reaction. The impact of EA on electrophysiological properties of colon-related DRG neurons was observed through whole-cell patch clamp analysis. RESULTS: EA significantly reduced the visceral pain behavior scores in rats with IBS in response to graded(20, 40, 60, 80 mm Hg) CRD stimulation. Additionally, EA downregulated the protein and mRNA expression levels of GFAP in the colon and colon-related DRG of rats with IBS. EA also regulated the resting membrane potential, rheobase and action potential of colon-related DRG neurons in rats with IBS. CONCLUSIONS: EA can regulate the excitatory properties of colon-related DRG neurons by downregulating the protein and mRNA expression of GFAP in the colon and colon-related DRG, indicating a potential neurobiological mechanism by which EA relieves visceral hypersensitivity in rats with IBS.

Combined Treatment with Bojungikgi-tang(Buzhong Yiqi Decoction)and Riluzole Attenuates Cell Death in TDP-43-Expressing Cells

Objective To examine the effect of combined treatment with Bojungikgi-tang(BJIGT,Buzhong Yiqi Decoction)and riluzole(RZ)in transactive response DNA-binding protein 43(TDP-43)stress granule(SG)cells,a amyotrophic lateral sclerosis(ALS)cell line using transcriptomic and molecular *** TDP-43 SG cells were pretreated with BJIGT(100µg/mL),RZ(50µmol/L),and combined BJIGT(100µg/mL)/RZ(50µmol/L)for 6 h before treatment with lipopolysaccharide(LPS,200µmol/L).Cell viability assay was performed to elucidate cell toxicity in TDP-43 SC cells using a cell-counting kit-8(CCK8)assay *** expression levels of cell death-related proteins,including Bax,caspase 1,cleaved caspase 3 and DJ1 in TDP-43 SG cells were examined by Western blot *** autophagy-related proteins,including pmTOR/mTOR,LC3b,P62,ATG7 and Bcl-2-associated athanogene 3(Bag3)were investigated using immunofluorescence and immunoblotting *** Cell viability assay and Western blot analysis showed that combined treatment with BJIGT and RZ suppressed LPS-induced cell death and expression of cell death-related proteins,including Bax,caspase 1,and DJ1(P<0.05 or P<0.01).Immunofluorescence and immunoblotting assays showed that combined treatment with BJIGT and RZ reduced LPS-induced formation of TDP-43 aggregates and regulated autophagy-related protein levels,including p62,light chain 3b,Bag3,and ATG7,in TDP-43-expressing cells(P<0.05 or P<0.01).Conclusion The combined treatment of BJIGT and RZ might reduce inflammation and regulate autophagy dysfunction in TDP-43-induced ALS.
