Neoadjuvant treatment of pancreatic ductal adenocarcinoma:Whom,when and how

Pancreatic ductal adenocarcinoma(PDAC),which is notorious for its aggressiveness and poor prognosis,remains an area of great unmet medical need,with a 5-year survival rate of 10%-the lowest of all solid *** diagnosis,only 20%of patients have resectable pancreatic cancer(RPC)or borderline RPC(BRPC)disease,while 80%of patients have unresectable tumours that are locally advanced pancreatic cancer(LAPC)or have distant *** 60%of patients who undergo upfront surgery for RPC are unable to receive adequate adjuvant chemotherapy(CHT)because of postoperative complications and early cancer *** important paradigm shift to achieve better outcomes has been the sequence of therapy,with neoadjuvant CHT preceding *** surgical stages have emerged for the preoperative assessment of nonmetastatic pancreatic cancers:RPC,BRPC,and *** main goal of neoadjuvant treatment(NAT)is to improve postoperative outcomes through enhanced selection of candidates for curative-intent surgery by identifying patients with aggressive or metastatic disease during initial CHT,reducing tumour volume before surgery to improve the rate of margin-negative resection(R0 resection,a microscopic margin-negative resection),reducing the rate of positive lymph node occurrence at surgery,providing early treatment of occult micrometastatic disease,and assessing tumour chemosensitivity and tolerance to treatment as potential surgical *** this editorial,we summarize evidence concerning NAT of PDAC,providing insights into future practice and study *** research is needed to establish predictive biomarkers,measures of therapeutic response,and multidisciplinary stra tegies to improve patient-centered outcomes.

Classification of knowledge graph completeness measurement techniques

At present,although knowledge graphs have been widely used in various fields such as recommendation systems,question and answer systems,and intelligent search,there are always quality problems such as knowledge omissions and *** assessment and control,as an important means to ensure the quality of knowledge,can make the applications based on knowledge graphs more complete and more accurate by reasonably assessing the knowledge graphs and fixing and improving the quality problems at the same ***,as an indispensable part of the knowledge graph construction process,the results of quality assessment and control determine the usefulness of the knowledge *** them,the assessment and enhancement of completeness,as an important part of the assessment and control phase,determine whether the knowledge graph can fully reflect objective phenomena and reveal potential connections among *** this paper,we review specific techniques of completeness assessment and classify completeness assessment techniques in terms of closed world assumptions,open world assumptions,and partial completeness *** purpose of this paper is to further promote the development of knowledge graph quality control and to lay the foundation for subsequent research on the completeness assessment of knowledge graphs by reviewing and classifying completeness assessment techniques.

How to implement a knowledge graph completeness assessment with the guidance of user requirements

In the context of big data, many large-scale knowledge graphs have emerged to effectively organize the explosive growth of web data on the Internet. To select suitable knowledge graphs for use from many knowledge graphs, quality assessment is particularly important. As an important thing of quality assessment, completeness assessment generally refers to the ratio of the current data volume to the total data *** evaluating the completeness of a knowledge graph, it is often necessary to refine the completeness dimension by setting different completeness metrics to produce more complete and understandable evaluation results for the knowledge ***, lack of awareness of requirements is the most problematic quality issue. In the actual evaluation process, the existing completeness metrics need to consider the actual application. Therefore, to accurately recommend suitable knowledge graphs to many users, it is particularly important to develop relevant measurement metrics and formulate measurement schemes for completeness. In this paper, we will first clarify the concept of completeness, establish each metric of completeness, and finally design a measurement proposal for the completeness of knowledge graphs.

Analysis of phase preservation and interferometric offset test in sparse SAR imaging

Sparse synthetic aperture radar(SAR) imaging has emerged as a reliable microwave imaging scheme in the recent decade and excels in down-sampling reconstruction and full-sampling performance improvements such as noise, sidelobe, speckle, and ambiguity suppression. To utilize complex image products of sparse reconstruction for improvement in polarimetric, interferometric, and tomographic SAR imaging, it is necessary to evaluate the phase preservation of sparse SAR imaging. In this study, we first introduce the general alternating direction method of multipliers(ADMM) as the universal framework for sparse reconstruction algorithms and adopt chirp scaling algorithm(CSA)-based azimuth-range decouple operators to avoid expensive data storage and processing. Further, we theoretically analyze the phase preservation of the sparse reconstruction algorithm through a comparison with the reconstruction results of CSA. Finally,we conduct the interferometric offset test on the sparse reconstruction results of simulated and real Gaofen-3(GF-3) SAR data, demonstrating the phase-preserving ability of sparse methods.

Joint UAV trajectory and communication design with heterogeneous multi-agent reinforcement learning

Unmanned aerial vehicles(UAVs) are recognized as effective means for delivering emergency communication services when terrestrial infrastructures are unavailable. This paper investigates a multiUAV-assisted communication system, where we jointly optimize UAVs’ trajectories, user association, and ground users(GUs)’ transmit power to maximize a defined fairness-weighted throughput metric. Owing to the dynamic nature of UAVs, this problem has to be solved in real time. However, the problem’s non-convex and combinatorial attributes pose challenges for conventional optimization-based algorithms, particularly in scenarios without central controllers. To address this issue, we propose a multi-agent deep reinforcement learning(MADRL) approach to provide distributed and online solutions. In contrast to previous MADRLbased methods considering only UAV agents, we model UAVs and GUs as heterogeneous agents sharing a common objective. Specifically, UAVs are tasked with optimizing their trajectories, while GUs are responsible for selecting a UAV for association and determining a transmit power level. To learn policies for these heterogeneous agents, we design a heterogeneous coordinated QMIX(HC-QMIX) algorithm to train local Q-networks in a centralized manner. With these well-trained local Q-networks, UAVs and GUs can make individual decisions based on their local observations. Extensive simulation results demonstrate that the proposed algorithm outperforms state-of-the-art benchmarks in terms of total throughput and system fairness.

In recent years,there has been significant research on the application of deep learning(DL)in topology optimization(TO)to accelerate structural ***,these methods have primarily focused on solving binary TO problems,and effective solutions for multi-material topology optimization(MMTO)which requires a lot of computing resources are still ***,this paper proposes the framework of multiphase topology optimization using deep learning to accelerate MMTO *** framework employs convolutional neural network(CNN)to construct a surrogate model for solving MMTO,and the obtained surrogate model can rapidly generate multi-material structure topologies in negligible time without any *** performance evaluation results show that the proposed method not only outputs multi-material topologies with clear material boundary but also reduces the calculation cost with high prediction ***,in order to find a more reasonable modeling method for MMTO,this paper studies the characteristics of surrogate modeling as regression task and classification *** the training of 297 models,our findings show that the regression task yields slightly better results than the classification task in most ***,The results indicate that the prediction accuracy is primarily influenced by factors such as the TO problem,material category,and data ***,factors such as the domain size and the material property have minimal impact on the accuracy.

Brazilin Actuates Ferroptosis in Breast Cancer Cells via p53/SLC7A11/GPX4 Signaling Pathway

Objective:To investigate the mechanism of induction of ferroptosis by brazilin in breast cancer ***:Breast cancer 4T1 cells were divided into 6 groups:control,brazilin 1/2 half maximal inhibitory concentration(IC50),IC50,2×IC50,erastin（10 μg/mL） and capecitabine（10 μg/mL） *** effect of brazilin on the proliferation of 4T1 cells was detected by cell counting kit-8 assay,and the treatment dose of brazilin was *** effect of brazilin on the mitochondrial morphology of 4T1 cells,and the mitochondrial damage was evaluated under electron *** levels of Fe2+,reactive oxygen species（ROS）,malondialdehyde（MDA）,glutathione（GSH） and glutathione peroxidase 4（GPX4） were estimated using various detection *** invasion and migration abilities of 4T1 cells were detected by scratch assay and transwell *** expressions levels of tumor protein p53, solute carrier family 7 member 11（SLC7A11）,GPX4 and acyl-CoA synthetase long-chain family member 4（ACSL4） proteins were quantified by Western blot ***:Compared to the control group,the 10(1/2 IC50),20(IC50) and 40(2×IC50) μg/mL brazilin,erastin,and capecitabine groups showed a significant decrease in the cell survival rate,invasion and migration abilities,GSH,SLC7A11 and GPX4 protein expression levels,and mitochondrial volume and ridge（P<0.05）,and a significant increase in the mitochondria membrane density,Fe2+,ROS and MDA levels,and p53 and ACSL4protein expression levels（P<0.05）.Conclusions:Brazilin actuated ferroptosis in breast cancer cells,and the underlying mechanism is mainly associated with the p53/SLC7A11/GPX4 signaling pathway.

Bridging element-free Galerkin and pluri-Gaussian simulation for geological uncertainty estimation in an ensemble smoother data assimilation framework

The facies distribution of a reservoir is one of the biggest concerns for geologists,geophysicists,reservoir modelers,and reservoir engineers due to its high importance in the setting of any reliable decisionmaking/optimization of field development *** approach for parameterizing the facies distribution as a random variable comes naturally through using the probability *** the prior probability fields of facies come either from a seismic inversion or from other sources of geologic information,they are not conditioned to the data observed from the cores extracted from the *** paper presents a regularized element-free Galerkin(R-EFG)method for conditioning facies probability fields to facies *** conditioned probability fields respect all the conditions of the probability theory(*** the values are between 0 and 1,and the sum of all fields is a uniform field of 1).This property achieves by an optimization procedure under equality and inequality constraints with the gradient projection *** conditioned probability fields are further used as the input in the adaptive pluri-Gaussian simulation(APS)methodology and coupled with the ensemble smoother with multiple data assimilation(ES-MDA)for estimation and uncertainty quantification of the facies *** history-matching of the facies models shows a good estimation and uncertainty quantification of facies distribution,a good data match and prediction capabilities.

Medicinal plants have aroused considerable interest as an alternative to chemical drugs due to the beneﬁcial effects of their active secondary ***,the extensive use of chemical fertilizers and pesticides in pursuit of yield has caused serious pollution to the environment,which is not conducive to sustainable devel-opment in theﬁeld of medicinal *** fertilizers are a type of“green fertilizer”containing speciﬁc microorganisms that can improve the soil microbial structure,enhance plant resistance to biological and abiotic stresses,and increase the yield of medicinal *** root exudates of medicinal plants attract different micro-organisms to the rhizosphere,which then migrate further to the plant *** microbes can increase the levels of soil nutrients,and improve the physical and chemical properties of soil through nitrogenﬁxation,and phosphorus and potassium *** addition,soil microbes can promote the synthesis of hormones that increase plant growth and the accumulation of active compounds,eventually improving the quality of med-icinal *** 2022,the total value of the global microbial fertilizer market was$4.6 billion and is estimated to reach$10.36 billion by *** this review,we have summarized the types of microbial fertilizers,the coloniza-tion and migration of microorganisms to plant tissues,and the beneﬁcial effects of microbial *** addi-tion,the prospects of developing microbial fertilizers and their application for medicinal plants have also been *** aims to provide a reference for the rational application of microbial fertilizers in theﬁeld of med-icinal plants and the green and sustainable development of medicinal plant resources.
