The superfamily of G protein-coupled receptors (GPCRs) contains immense structural and functional diversity and mediates a myriad of biological processes upon activation by various extracellular *** roles of GPCRs have been established in bone development,remodeling,and *** human GPCR mutations impair bone development or metabolism,resulting in *** we summarize the disease phenotypes and dysfunctions caused by GPCR gene mutations in humans as well as by deletion in *** date,92 receptors (5 glutamate family,67 rhodopsin family,5 adhesion,4 frizzled/taste2 family,5 secretin family,and 6 other 7TM receptors) have been associated with bone diseases and dysfunctions (36 in humans and 72 in animals).By analyzing data from these 92 GPCRs,we found that mutation or deletion of different individual GPCRs could induce similar bone diseases or dysfunctions,and the same individual GPCR mutation or deletion could induce different bone diseases or dysfunctions in different populations or animal *** from human diseases or dysfunctions identified 19 genes whose mutation was associated with human BMD:9 genes each for human height and osteoporosis;4 genes each for human osteoarthritis (OA) and fracture risk;and 2 genes each for adolescent idiopathic scoliosis (AIS),periodontitis,osteosarcoma growth,and tooth *** from gene knockout animals found 40 GPCRs whose deficiency reduced bone mass,while deficiency of 22 GPCRs increased bone mass and BMD;deficiency of 8 GPCRs reduced body length,while 5 mice had reduced femur size upon GPCR ***,deficiency in 6 GPCRs induced osteoporosis;4 induced osteoarthritis;3 delayed fracture healing;3 reduced arthritis severity;and reduced bone strength,increased bone strength,and increased cortical thickness were each observed in 2 GPCR-deficiency *** ever-expanding number of GPCR mutation-associated diseases warrants accelerated molecular analysis,population

A modified technique for tibial bone sparing in unicompartmental knee arthroplasty

Anorexia and fasting are host adaptations to acute infection, inducing a metabolic switch towards ketogenesis and the production of ketone bodies, including β-hydroxybutyrate (BHB) 1-6. However, whether ketogenesis metabolically influences the immune response in pulmonary infections remains unclear. Here we report impaired production of BHB in humans with SARS-CoV-2-induced but not influenza-induced acute respiratory distress syndrome (ARDS). CD4^(+) T cell function is impaired in COVID-19 and BHB promotes both survival and production of Interferon-γ from CD4^(+) T cells. Using metabolic tracing analysis, we uncovered that BHB provides an alternative carbon source to fuel oxidative phosphorylation (OXPHOS) and the production of bioenergetic amino acids and glutathione, which is important for maintaining the redox balance. T cells from patients with SARS-CoV-2-induced ARDS were exhausted and skewed towards glycolysis, but can be metabolically reprogrammed by BHB to perform OXPHOS, thereby increasing their functionality.

Although some studies had suggested a bidirectional relationship between diabetes and periodontal disease, there were no appropriate tools for nurses to evaluate oral status and oral health behaviors in patients with diabetes. Therefore, the Diabetes Oral Health Assessment Tool (DiO-HAT ?) was developed with items contributed by health care professionals (diabetologists, periodontal specialists, a preventive dentist, a Certified Nurse in Diabetes Nursing, a national registered dietitian, registered nurses, a dental hygienist, and nursing researchers) who were involved in the medical care of patients with diabetes. Subsequently, a survey of 700 Diabetes Nurse Specialists (DNS) was conducted to determine their score of recognition and implementation of the DiOHAT ?, however, 304 participants (43.4%) responded. Constructive concept validation and the Cronbach’s alpha coefficient for all assessment items were 0.931, indicating high reliability: Factor 1, Patient’s oral health status (α = 0.831);Factor 2, Implementation of oral health behaviors (α = 0.890);Factor 3, Information transmission regarding dental visits (α = 0.862);and Factor 4, Perceptions and knowledge of oral health behaviors (α = 0.793). Although the mean score of recognition of DiOHAT ? was 3.5 ± 0.4 points, the mean value of the implementation score was 1.5 ± 0.5 points (obtained using a 4-grade scale). The implementation scores were significantly lower than the recognition scores for all items (p ndings suggested that the DNS were not inclined to implement all items of DiOHAT ?, despite recognizing their importance.

Low molecular weight heparin reduces arterial blood lactic acid content and increases estimated glomerular filtration rate in patients with moderate Covid-19 pneumonia

Background:Coronavirus disease 2019(Covid-19)remains a serious health threat *** aimed to investigate whether low molecular weight heparin(LMWH)can promote organ function recovery in moderate Covid-19 pneumonia ***:We initiated an LMWH protocol in Covid-19 patients with increased D-dimer,body mass index>30 kg/m^(2) or a history of diabetes from January 18,2020 at Shanghai Public Health Clinical *** this retrospective study,we assigned moderate Covid-19 pneumonia patients admitted between January 18th and April 18,2020 receiving the LMWH protocol to the LMWH *** patients who met the inclusion criteria but did not receive LMWH protocol were included in the control group by 1:2 propensity score *** clinical information,indicators for renal function,arterial blood gas analyses,arterial blood lactic acid content(mmol/L),and coagulation indexes at 0 day,3 days,7 days,and 11 days after admission were recorded and compared between the two ***:There were 41 patients in the LMWH group and 82 patients in the control *** information in both groups were *** to the control group,the arterial blood lactic acid content(mmol/L)at day 11(1.3[1.1,1.7]vs.1.2[0.9,1.3],P=0.016)was reduced in the LMWH *** estimated glomerular filtration rate(eGFR)in the LMWH group was higher than that in the control group at day 7(108.54[89.11,128.17]vs.116.85[103.39,133.47],P=0.039)and day 11(113.74[94.49,126.34]vs.128.31[112.75,144.12],P=0.003).The serum creatinine levels(Scr)in the LMWH group were lower than that in the control group at day 7(62.13[51.47,77.64]vs.55.49[49.50,65.75],P=0.038)and day 11(63.35[50.17,75.73]vs.51.62[44.62,61.24],P=0.005).Conclusions:LMWH treatment can reduce arterial blood lactic acid levels and improve eGFR in moderate Covid-19 pneumonia *** controlled trials are warranted to further investigate this issue.

A safety evaluation of profound hypothermia-induced suspended animation for delayed resuscitation at 90 or 120min

Background: The successful treatment of military combat casualties with penetrating injuries is significantly dependent on the time needed to get the patient to an adequate treatment facility. Profound hypothermia induced suspended animation for delayed resuscitation(SADR) is a novel approach for inducing cardiac arrest and buying additional time for such injuries. However, the time used to safely administer circulatory arrest(CA) is controversial. The goal of this study was to evaluate the safety of hypothermia-induced SADR over 90 and 120 min time ***: Sixteen male BAMA minipigs were randomized into two groups: CA90 group(90 min, n =8) and CA120 group(120 min, n =8). Cannulation of the right common carotid arteries and internal jugular veins was performed to establish cardiopulmonary bypass for each animal. Through the perfusion of cold organ preservation solution(OPS), cardioplegia and profound hypothermia(15℃) were induced. After CA, cardiopumonary bypass(CPB) was restarted, and the animals were gradually re-warmed and resuscitated. The animals were assisted with ventilators until spontaneous breathing was achieved. The index of hemodynamic perioperative serum chemistry values [alanine transaminase(ALT), aspartate aminotransferase(AST), creatinine(CR), lactic dehydrogenase(LDH) and troponin T(TnT)] and survival were observed from pre-operation to 7 days ***: Fifteen animals were enrolled in the experiment, while 1 animal in CA120 group died from surgical error. All 8 animals in CA90 group recovered, with only 1 animal displaying mild disability. However, in CA120 group, only 2 animals survived with severe disability, and the other 5 animals died after 2 days post-operation. In CA90 group, the perioperative serum chemistry values increased at 1 day post-operation(ALT 84.43±18.65 U/L; AST 88.99±23.19 U/L; Cr 87.90±24.49μmol/L; LDH 1894.13±322.26 U/L; TnT 0.849±0.135 ng/ml) but decreased to normal or almost normal levels at 7 days p

Multidrug resistance(MDR) and targeted therapies remain as vital challenges in tumor *** provide promising application for cancer theranostics as for their excellent advantages of tumor-targeted cytotoxicity and imaging *** present study,we developed N-((2-hydroxy-3-trimethylammonium) propyl) chitosan chloride(HTCC)/alginate encapsulated FeO magnetic nanoparticles(HTCC-MNPs) and applied it into MDR gastric cancer both in vivo and in ***-MNPs were fabricated using ALG,FeO and HTCC in ionic gelation *** size and physical characteristics of nanoparticles were detected by dynamic light scattering,transmission electron microscopy(TEM) and zeta potential ***-MNPs exhibited excellent water solubility and biocompatibility and significantly reduced cell viability in drug-resistant cancer cell line SGC7901/ADR cells but not in normal gastric cells(P ndrial membrane potential loss,decreased ATP production and excessive reactive oxygen species(ROS) generation as compared with control group(P nd tumor volume measurement revealed that HTCC-MNPs markedly inhibited in vivo tumor growth(P nd decreased tumor volume via the induction of cellular autophagy and apoptosis,which was attributed to mitochondrial dysfunction and excessive ROS accumulation.

High-resolution medical images have important medical value,but are difficult to obtain *** by hardware equipment and patient’s physical condition,the resolution of directly acquired medical images is often not ***,many researchers have thought of using super-resolution algorithms for secondary processing to obtain high-resolution medical ***,current super-resolution algorithms only work on a single scale,and multiple networks need to be trained when super-resolution images of different scales are *** definitely raises the cost of acquiring high-resolution medical ***,we propose a multi-scale superresolution algorithm using *** algorithm combines a metalearning approach with an enhanced depth of residual super-resolution network to design a meta-upscale *** meta-upscale module utilizes the weight prediction property of meta-learning and is able to perform the super-resolution task of medical images at any ***,we design a non-integer mapping relation for super-resolution,which allows the network to be trained under non-integer magnification *** to the state-of-the-art single-image super-resolution algorithm on computed tomography images of the pelvic *** meta-learning multiscale superresolution algorithm obtained a surpassing of about 2%at a smaller model *** on different parts proves the high generalizability of our ***-scale super-resolution algorithms using meta-learning can compensate for hardware device defects and reduce secondary harm to patients while obtaining high-resolution medical *** can be of great use in imaging related fields.
