Mastitis often occurs during women’s lactation period， causing great psychological and physical pain to women. Fucoidan， a sulfated polysaccharide obtained from brown algae， has much broader biological properties. However， the roles of fucoidan in lipopolysaccharides (LPS)-induced mastitis are still undiscovered. The present study was aimed to evaluate the influences of fucoidan on LPS-induced mouse mastitis and investigate its possible mechanisms. The expression profiles of fucoidan acting on mastitis were analyzed by Network pharmacology. Additionally， mechanism experiments verified the mechanism of fucoidan on mastitis. The results of in vivo study displayed that the treatment of fucoidan to LPS-stimulated mouse mastitis decreased the inflammatory damage， proinflammatory cytokines level and repaired the completeness of blood-milk barrier. In the study of mouse mammary epithelial cells， fucoidan suppressed the secretion of reactive oxygen species (ROS)and enhanced the activity of antioxidant enzymes. Molecular experiments suggested that fucoidan promoted Nrf2.nuclear import and autophagy via activating the AMPK signaling pathway. The above results indicated that in mastitis， fucoidan promoted Nrf2.nuclear import and autophagy via activating the AMPK signaling pathway， thereby suppressing oxidative stress and inflammatory injury to the mammary glands and protecting the blood-milk barrier.

Heart failure(HF)is a clinical hallmark of cardiovascular syndrome leading to substantial morbidity,repeated hospitalization,and enormous healthcare *** elderly populations continue to increase,biomarker-guided diagnosis and treatment for age-related cardiac decline have become *** evidence demonstrates that N-terminal proBNP(NT-proBNP)can provide a unique window into the diagnosis and risk stratification with ***,a simple yet robust aptasensor is developed for on-site recognizing the NT-proBNP by its targeting aptamer,thus achieving the accurate diagnosis of *** aptasensing system is prepared by absorbing the fluorophore-labeled aptamer strand onto the graphene oxide(GO),leading to efficient quenching without possible off-site signal *** aptamer strand can specifically identify target NT-proBNP molecules via a versatile conformational transformation,resulting in the desorption of the aptamer-NTproBNP complexes from GO and re-generation fluorescence signal,thus allowing sensitive detection of NT-proBNP in 37 clinical blood *** advantage of the high specificity of aptamer-guaranteed recognitions of NT-proBNP,this aptasensor system readily achieves better diagnostic performance for HF than commercially adopted chemiluminescence immunoassay(Siemens,CLIA)in hospitals in terms of accuracy(89.2.vs 83.8%),specificity(89.5%vs 84.2.),and positive predictive value(88.9%vs 83.3%).This work provides a stable option for the diagnosis and treatment of elderly HF-related diseases inclinics.

Characterization of early maturing elite genotypes based on MTSI and MGIDI indexes:an illustration in upland cotton(Gossypium hirsutum L.)

Through linear tracking method to deal with nonlinear systems, this paper proposed a dynamic state-feedback controller design method, to deal with the problem of stabilizing nonlinear systems. Under suitable assumptions on the linearized system, we prove that the proposed method can stabilize the system to the expected equilibrium state. The method can be implemented based on either model or data with high computational efficiency for only linear matrix inequalities to be solved. We make a certain extension for a more general situation, proposing the dynamic output-feedback controller design method. The effectiveness of the proposed method in nonlinear systems is illustrated through the simulation.

Background Globally,the cultivation of cotton is constrained by its tendency for extended periods of *** maturity plays a potential role in rainfed-based multiple cropping system especially in the current era of climate *** the current study,a set of 2. diverse Gossypium hirsutum genotypes were evaluated in two crop seasons with three planting densities and assessed for 11 morphological traits related to early *** study aimed to identify genotype(s)that mature rapidly and accomplish well under diverse environmental conditions based on the two robust multivariate techniques called multi-trait stability index(MTSI)and multi-trait genotype-ideotype distance index(MGIDI).Results MTSI analysis revealed that out of the 2. genotypes,three genotypes,viz.,NNDC-30,A-2.and S-32.accomplished well in terms of early maturity traits in two ***,three genotypes were selected using MGIDI method for each planting densities with a selection intensity of 15%.The strengths and weaknesses of the genotypes selected based on MGIDI method highlighted that the breeders could focus on developing early-maturing genotypes with specific traits such as days to first flower and boll *** selected genotypes exhibited positive genetic gains for traits related to earliness and a successful harvest during the first and second ***,there were negative gains for traits related to flowering and boll *** The study identified three genotypes exhibiting early maturity and accomplished well under different planting *** multivariate methods(MTSI and MGIDI)serve as novel approaches for selecting desired genotypes in plant breeding programs,especially across various growing *** methods offer exclusive benefits and can easily construe and minimize multicollinearity issues.

Background Aloe ankoberensis M.G. Gilbert & Sebsebe and A. debrana Christian are Ethiopian endemic species currently classified as endangered and least concern, respectively under International Union for Conservation of Nature(IUCN) categories. Recent studies indicate that climate change is anticipated to significantly influence the distribution of plant species. Therefore, this study aimed to model the distribution of A. ankoberensis and A. debrana under different climate change scenarios in the North Shewa Zone, Amhara National Regional State of Ethiopia. Thirty-six and 397 georeferenced presence points for A. ankoberensis and A. debrana, respectively, and 12.environmental variables were used to simulate their current and future distributions. The ensemble model approach was used to examine the current and future(2.50 and 2.70) climatic suitability for both species under three shared socio-economic pathway(SSP) climate scenarios(SSP 2.6, 4.5 and 8.5).Results The performance of ensemble model was excellent for A. ankoberensis with score of area under curve(AUC) 0.96 and true skill statistics(TSS) 0.88, and good for A. debrana with score of AUC 0.87 and TSS 0.63. The main variables that affected the species' distributions were mean diurnal range of temperature, annual precipitation, and elevation. According to the model, under the current climate conditions, 98.32., 1.01%, 0.52., and 0.15% were not suitable, lowly, moderately, and highly suitable areas, respectively for A. ankoberensis, and 63.89%, 2..35%, 12.54%, and 0.2.% were not suitable, lowly, moderately and highly suitable areas, respectively for A. debrana. Under future climate scenarios, suitable habitats of these species could shrink. In addition, under all climate change scenarios, it is anticipated that highly suitable areas for both species and moderately suitable areas for A. ankoberensis will be lost completely in the future unless crucial interventions are done on *** The results indicat

Chemokine CXCL12.plays a crucial role in both direct bactericidal activity and phagocytosis in humans. However, the mechanisms and evolutionary functions of these processes in vertebrates remain largely unknown. In this study, we found that the direct bactericidal activity of CXCL12.is highly conserved across various vertebrate lineages, including Arctic lamprey (Lampetra japonica), Basking shark (Cetorhinus maximus), grass carp (Ctenopharyngodon idella), Western clawed frog (Xenopus tropicalis), Green anole (Anolis carolinensis), chicken(Gallus gallus), and human (Homo sapiens). CXCL12.also has been shown to promote phagocytosis in lower and higher vertebrates. We then employed C. idella CXCL12. (CiCXCL12.) as a model to further investigate its immune functions and underlying mechanisms. CiCXCL12. exerts direct broad-spectrum antibacterial activity by targeting bacterial acidic phospholipids, resulting in bacterial cell membrane perforation, and eventual lysis. Monocytes/macrophages are attracted to the infection sites for phagocytosis through the rapid production of CiCXCL12. during bacterial infection. CiCXCL12. induces CDC42.and CDC42.GTPase activation, which in turn mediates F-actin polymerization and cytoskeletal rearrangement. The interaction between F-actin and Aeromonas hydrophila facilitates bacterial internalization into monocytes/macrophages. Additionally, A. hydrophila is colocalized within early endosomes, late endosomes and lysosomes, ultimately degrading within phagolysosomes. CiCXCL12. also activates PI3K-AKT, JAK-STAT5 and MAPK-ERK signaling pathways. Notably, only the PI3K-AKT signaling pathway inhibits LPS-induced monocyte/macrophage apoptosis. Thus, CiCXCL12. plays key roles in reducing tissue bacterial loads, attenuating organ injury, and decreasing mortality rates. Altogether, our findings elucidate the conserved mechanisms underlying CXCL12.mediated bactericidal activity and phagocytosis, providing novel perspectives into the immune functions

Arbuscular mycorrhizae status of host plant affects offspring’s phenotype and transcriptome expression in a low-phosphorus environment

Although the effects of arbuscular mycorrhizal fungi(AMF)on host plants have been well documented,whether the effects of AMF on parental generations affect offspring performance is not fully *** conducted a common garden experiment to determine whether AMF status of host plants(Medicago truncatula)affects phenotype and transcriptome expression of their *** from four type parental treatments(low-phosphorus(P)soil without AMF,low-P soil with AMF,high-P soil without AMF and high-P soil with AMF)were grown under low-P(LPS)and normal-P soil(OHS)*** fowering pattern of LP offspring was similar to their parents,such that plants with AMF fowered earlier than those without AMF under OHS condition but were opposite under LPS *** transcriptome differential analysis showed that some differential transcripts(45 for parental plants growing under low-P condition and 3 for parental plants growing under high-P condition)expression patterns between offspring were comparable,and that only affected by parental AMF status regardless of the P environment that offspring was ***(146 for parental plants growing under low-P condition and 2.for parental plants growing under high-P condition),however,were affected both by the parental AMF status and the offspring P *** addition,the number of differential transcripts between offspring whose parental plants grew under high-P condition was far less than under low-P *** results indicate that AMF may not only affect the current generation of host plants but also affect the offspring especially when their parents have experienced a stressful environment.
