Mannanase improves the growth performance of broilers by alleviating inflammation of the intestinal epithelium and improving intestinal microbiota

This experiment aimed to discuss and reveal the effect and mechanism of mannanase on intestinal inflammation in broilers triggered by a soybean meal diet. In this experiment, 384 Arbor Acres broilers at 1 d old were randomly divided into 3 treatment groups. The broilers were fed a corn-soybean meal basal diet, a low-energy diet (metabolizable energy reduced by 50 kcal/kg), and a low-energy diet supplemented with 100 mg/kg mannanase for 42.d. The low-energy diet increased feed conversion ratio from 0 to 42.d, reduced ileal villus height and villus height-to-crypt depth ratio and upregulated the expression of nuclear factor kappa B ( NF-κB ) in the ileum ( P P P CLDN1 ) and zonula occludens-1 ( ZO-1 ) in the jejunum at 2. d, downregulated the expression of ilealNF-κB , and increased the content of isobutyric acid in the cecum of broilers ( P Lactobacillus reuteri in the ileum of broilers. The addition of mannanase increased the relative abundance ofLactobacillus-KC45b andLactobacillus johnsonii in broilers. Furthermore, a low-energy diet reduced the relative abundance ofButyricicoccus in the intestine of broilers and inhibited oxidative phosphorylation and phosphoinositol metabolism. Mannanase increased the relative abundance ofOdoribacter , promoted energy metabolism and N-glycan biosynthesis, and increased the activities of GH3 and GH18. It is concluded that mannanase could improve the growth performance of broilers by reducing the expression ofNF-κB in the ileum, increasing the production of SCFA in the cecum, suppressing intestinal inflammation, balancing the intestinal microbiota, reducing damage to the intestinal barrier, and improving the efficiency of nutrient utilization to alleviate the adverse effects caused by the decrease in dietary energy *** The PDF

An Enterococcus faecalis isolate harboring both aerobic and anaerobic uric acid metabolic pathways alleviates hyperuricemia in mice

Probiotics have been demonstrated to be effective and safe for hyperuricemia (HUA) and gout treatment. Hence， identifying microbes with high uric acid- or purine-metabolizing ability is highly important. Due to the high load of uric acid in chicken feces， we hypothesized that the chicken gut would be an excellent source for new uric acid or purine-metabolizing microbes. In this study， we examined chicken gut contents to isolate an Enterococcus faecalis strain (designated CML390) that displayed high uric acid-degrading ability. In vitro experiments indicated that E. faecalis CML390 could effectively metabolize uric acid under both aerobic and anaerobic conditions， and the richer the nutrients， the higher the rate of uric acid degradation. By integrating genomics， transcriptomics， and metabolomics analyses， we revealed two pathways involved in aerobic and anaerobic uric acid degradation， respectively， in this isolate. E. faecalis CML390 relies on oxygen to metabolize uric acid into substances such as allantoin under aerobic conditions， while it also harbors a conserved uric acid-inducible gene cluster for anaerobic uric acid metabolism. An in vivo experiment showed that gavage with live E. faecalis CML390 in a mouse model of HUA (established using a combination of uric acid and potassium oxonate) significantly reduced serum uric acid levels and alleviated HUA. Our findings contribute to the exploration of the pathways and mechanisms of uric acid metabolism in intestinal microorganisms and provide a candidate probiotic for the treatment of HUA and gout.

Multiscale observation in wide-spatial radar surveillance based on coherent FDA design

Wide-spatial radar surveillance missions are challenging tasks, requiring an increased power budget and agility in transmission aimed at extracting information from multiple targets in different environments. These requirements necessitate high transmitting degrees of freedom(DOF) to achieve the objective of multiscale observation for specific tasks in specialized regions of interest. Herein, we exploit the multiscale observation ability in wide-spatial radar surveillance based on frequency diverse array(FDA)radar. The proposed method facilitates spatial anisotropic control of multiple radar resources, including the transmitting waveforms, beampattern, and bandwidth. By utilizing a coherent FDA radar, we offer principles for the selection of baseband waveforms, in addition to the quantitative design of the beampattern gain and optimal bandwidth from the perspective of detection. The feasibility of the proposed method is validated through numerical experiments, thus indicating the potential in wide-spatial radar surveillance. Moreover,this work can be regarded as a preliminary attempt to gauge the efficacy of the computational array, a novel academic concept.

Understanding the mechanism underlying the anti-inflammatory effect of dietary flavonoids on IBD: a focus on gut microbiota and its metabolites involved in SCFA， bile acid， tryptophan and lipopolysaccharides

Inflammatory bowel disease (IBD) is a chronic gastrointestinal disease with a high incidence. Treatment for IBD includes medications and diet， and common anti-inflammatory medications have limitations like drug resistance and serious adverse effects. Accumulating evidence has demonstrated that dietary flavonoids exhibit an alleviative effect on IBD by influencing gut microbiota. The microbiota-derived metabolites also regulate IBD and maintain intestinal homeostasis. In this review， we investigate the therapeutic effect of gut microbiota and metabolites on IBD by intestinal immune and intestinal barrier function. We demonstrate the underlying mechanism of dietary flavonoids as an anti-inflammatory molecule alleviating IBD by regulating gut microbiota， short chain fatty acid (SCFA)， bile acid (BA)， tryptophan (Trp) metabolism and lipopolysaccharides (LPS)-toll-like receptor 4 (TLR4) signaling pathway. Based on structural differences of flavonoids， we summarize the recent research progress on the role of different dietary flavonoids in alleviating IBD by gut microbiota and metabolites in animal and clinical trials. This review indicates that dietary flavonoids targeting gut microbiota and metabolites provide a promising strategy for the treatment of inflammation and novel insights into the management of IBD.

Eukaryotic genomes are hierarchically packaged into cell nucleus,affecting gene *** genome is organized into multiscale structural units,including chromosome territories,compartments,topologically associating domains(TADs),and DNA *** identification of these hierarchical structures has benefited from the development of experimental approaches,such as 3C-based methods(Hi-C,ChIA-PET,etc.),imaging tools(2.-FISH,3D-FISH,Cryo-FISH,etc.)and ligation-free methods(GAM,SPRITE,etc.).In recent two decades,numerous studies have shown that the 3D organization of genome plays essential roles in multiple cellular processes via various mechanisms,such as regulating enhancer activity and promoter-enhancer ***,there are relatively few studies about the 3D genome in livestock ***,studies for exploring the function of 3D genomes in livestock are urgently needed to provide a more comprehensive understanding of potential relationships between the genome and production *** this review,we summarize the recent advances of 3D genomics and its biological functions in human and mouse studies,drawing inspiration to explore the 3D genomics of livestock *** then mainly focus on the biological functions of 3D genome organization in muscle development and its implications in animal breeding.

A Video Captioning Method by Semantic Topic-Guided Generation

In the video captioning methods based on an encoder-decoder,limited visual features are extracted by an encoder,and a natural sentence of the video content is generated using a ***,this kind ofmethod is dependent on a single video input source and few visual labels,and there is a problem with semantic alignment between video contents and generated natural sentences,which are not suitable for accurately comprehending and describing the video *** address this issue,this paper proposes a video captioning method by semantic topic-guided ***,a 3D convolutional neural network is utilized to extract the spatiotemporal features of videos during the ***,the semantic topics of video data are extracted using the visual labels retrieved from similar video *** the decoding,a decoder is constructed by combining a novel Enhance-TopK sampling algorithm with a Generative Pre-trained Transformer-2.deep neural network,which decreases the influence of“deviation”in the semantic mapping process between videos and texts by jointly decoding a baseline and semantic topics of video *** this process,the designed Enhance-TopK sampling algorithm can alleviate a long-tail problem by dynamically adjusting the probability distribution of the predicted ***,the experiments are conducted on two publicly used Microsoft Research Video Description andMicrosoft Research-Video to Text *** experimental results demonstrate that the proposed method outperforms several state-of-art ***,the performance indicators Bilingual Evaluation Understudy,Metric for Evaluation of Translation with Explicit Ordering,Recall Oriented Understudy for Gisting Evaluation-longest common subsequence,and Consensus-based Image Description Evaluation of the proposed method are improved by 1.2.,0.1%,0.3%,and 2.4% on the Microsoft Research Video Description dataset,and 0.1%,1.0%,0.1%,and 2.8% on the Microsoft Research-Video to Text dataset

Bio-inspired Design and Inverse Kinematics Solution of an Omnidirectional Humanoid Robotic Arm with Geometric and Load Capacity Constraints

Inspired by the driving muscles of the human arm,a 4-Degree of Freedom(DOF)concentrated driving humanoid robotic arm is proposed based on a spatial double parallel four-bar *** four-bar mechanism design reduces the inertia of the elbow-driving unit and the torque by 76.65%and 57.81%,*** the human pose regulation strategy that the human arm picks up a heavy object by adjusting its posture naturally without complicated control,the robotic arm features an integrated position-level closed-form inverse solution method considering both geometric and load capacity *** method consists of a geometric constraint model incorporating the arm angle(φ)and the Global Configuration(GC)to avoid joint limits and singularities,and a load capacity model to constrain the feasible domain of the arm ***,trajectory tracking simulations and experiments are conducted to validate the feasibility of the proposed inverse solution *** simulated maximum output torque,maximum output power and total energy consumption of the robotic arm are reduced by up to 2.0%,13.3%,and 33.3%,*** experimental results demonstrate that the robotic arm can bear heavy loads in a human-like posture,effectively reducing the maximum output torque and energy consumption of the robotic arm by 1.83%and 5.03%,respectively,while avoiding joints beyond geometric and load capacity *** proposed design provides a high payload–weight ratio and an efficient pose control solution for robotic arms,which can potentially broaden the application spectrum of humanoid robots.

Promoting electrocatalytic nitrogen reduction to ammonia via dopant boron on two-dimensional materials

Electrocatalytic nitrogen reduction reaction(NRR) is a key process for producing energy efficient and environment friendly ammonia. Boron-doped two-dimensional materials are highly promising as NRR ***, the interaction between doped boron and matrix materials on NRR catalytic performance is still unclear,which is limiting the development of catalysts containing boron for NRR. Here, NRR on different boron-doped twodimensional(2.) materials was explored by first principle theory. It is found that adsorption energy of intermediate*NNH(E*NNH) can be used as a descriptor to characterize the catalytic activity for NRR. Boron-adsorbed black phosphorus(B-BP) is demonstrated showing excellent NRR catalytic activity and suppressing hydrogen evolution reaction. It is disclosed that the excellent catalytic performance of boron-doped structures comes from a proper quantitatively electron transfer(0.8e) between nitrogen in*NNH and the active site boron atom. This work not only established a descriptor for NRR catalytic activity on B-doped 2. materials, but also screened out a potential NRR catalyst. More importantly, the intrinsic reason and mechanism of high catalytic activity of boron-doped structures were proposed. This work provides a design principle for exploring high-performance NRR electrocatalysts containing boron.

Balanced Fracturing and Cold-welding of Magnesium during Ball Milling Assisted by Carbon Coating:Experimental and Molecular Dynamic Simulation

The lignite-derived carbon from self-protection pyrolysis was employed to balance the fracturing and cold-welding of magnesium during ball *** size analysis indicates that the introduction of lignite-derived carbon can effectively reduce the particle size of Mg while the introduction of graphite does no ***,the effect of lignite-derived carbon on crystallite size reduction of Mg is also better than graphite.A moderate cold-welding phenomenon was observed after ball-milling Mg with the lignite-derived carbon,suggesting less Mg is wasted on the milling vials and *** dynamic simulations reveal that the balanced fracturing and cold-welding of magnesium during ball milling is mainly attributed to the special structure of the lignite-derived carbon:graphitized short-range ordered stacking function as dry lubricant and irregular shape/sharp edge function as milling *** preliminary findings in current study are expected to offer implications for designing efficient Mg-based hydrogen storage materials.

A Weakly-Supervised Crowd Density Estimation Method Based on Two-Stage Linear Feature Calibration

In a crowd density estimation dataset,the annotation of crowd locations is an extremely laborious task,and they are not taken into the evaluation *** this paper,we aim to reduce the annotation cost of crowd datasets,and propose a crowd density estimation method based on weakly-supervised learning,in the absence of crowd position supervision information,which directly reduces the number of crowds by using the number of pedestrians in the image as the supervised *** this purpose,we design a new training method,which exploits the correlation between global and local image features by incremental learning to train the ***,we design a parent-child network(PC-Net)focusing on the global and local image respectively,and propose a linear feature calibration structure to train the PC-Net simultaneously,and the child network learns feature transfer factors and feature bias weights,and uses the transfer factors and bias weights to linearly feature calibrate the features extracted from the Parent network,to improve the convergence of the network by using local features hidden in the crowd *** addition,we use the pyramid vision transformer as the backbone of the PC-Net to extract crowd features at different levels,and design a global-local feature loss function(L2..We combine it with a crowd counting loss(LC)to enhance the sensitivity of the network to crowd features during the training process,which effectively improves the accuracy of crowd density *** experimental results show that the PC-Net significantly reduces the gap between fullysupervised and weakly-supervised crowd density estimation,and outperforms the comparison methods on five datasets of Shanghai Tech Part A,ShanghaiTech Part B,UCF_CC_50,UCF_QNRF and JHU-CROWD++.
