Lipid phosphorylation by diacylglycerol kinase(DGK)that produces phosphatidic acid(PA)plays important roles in various biological processes,including stress responses,but the underlying mechanisms remain ***,we show that DGK5 and its lipid product PA suppress ABA biosynthesis by interacting withABA-DEFICIENT2(ABA2),a key ABA biosynthesis enzyme,to negatively modulate plant responseto abiotic stress tested in Arabidopsis *** of DGK5 function rendered plants less damaged,whereas overexpression(OE)of DGK5 enhanced plant damage to water and salt *** dgk5 mutant plants exhibited decreased total cellular and nuclear levels of PA with increased levels of diacylglycerol,whereas DGK5-OE plants displayed the opposite ***,we found that both DGK5 and PA bind to the ABA-synthesizing enzyme ABA2 and suppress its enzymatic ***,the dgk5 mutant plants exhibited increased levels of ABA,while DGK5-OE plants showed reduced ABA *** addition,we showed that both DGK5 and ABA2 are detected in and outside the nuclei,and loss of DGK5 function decreased the nuclear association of *** found that both DGK5 activity and PA promote nuclear association of *** together,these results indicate that both DGK5 and PA interact with ABA2 to inhibit its enzymatic activity and promote its nuclear sequestration,thereby sup-pressing ABA production in response to abiotic *** study reveals a sophisticated mechanism by which DGK5 and PA regulate plant stress responses.

To the Editor:Frailty status represents one of the core determinants for the individualization of glycated hemoglobin(HbA1c)targets in the elderly ***,the most validated tools to quantify frailty,which are complex and involve multiple dimensions of assessment,are rarely used in the clinical practice of diabetes ***,a simple and practical metric for the evaluation of frailty is *** strength is an easily measurable,objective,and reproducible metric for assessing frailty status.[1]It is correlated with overall muscle strength and has been reported to be signicantly related to important health outcomes both in the general population[2,3]and in patients with diabetes.[4]Therefore,grip strength could be a useful marker for patient strati-cation in diabetes *** this context,this study aimed to examine the associations between HbA1c and all-cause mortality across categories of grip strength,as a measure of frailty status,in older adults with type 2 diabetes in the UK *** researches had applied to access the UK Biobank database with the application approval number of 82812.

Lung cancer is a prevalent malignancy,and fatalities of the disease exceed 400,000 cases *** squamous cell carcinoma(LUSC)has been recognized as the most common pathological form of lung *** comprehensive understanding of molecular features related to LUSC progression has great significance in LUSC prognosis assessment and clinical *** this study,we aim to identify a panel of signature genes closely associated with LUSC,which can provide novel insights into the progression of *** expression profiles were retrieved from public resources including gene expression omnibus(GEO)and the cancer genome atlas(TCGA)*** expressed genes(DEGs)between LUSC specimens and normal lung tissues were identified by bioinformatics analyses.A total of 66 DEGs were identified based on two cohorts of *** plugin of Cytoscape software was utilized for the further analyses of the top 10 candidate hub genes including OGN,ABI3BP,MAMDC2,FGF7,FAM107A,SPARCL1,DCN,COL14A1,and MFAP4 and CHRDL1,which showed significant downregulation in *** LUSC cell lines were used to validate the functions of CHRDL1 and FAM107A through overexpression ***,our data revealed novel candidate tumor-suppressor genes in LUSC,suggesting previously unappreciated mechanisms in the progression of LUSC.

BACKGROUND In 2016,the Food and Drug Administration approved the first hybrid closed-loop(HCL)insulin delivery system for adults with type 1 diabetes(T1D).There is limited information on the impact of using HCL systems on patient-reported outcomes(PROs)in patients with T1D in real-world clinical *** this independent study,we evaluated glycemic parameters and PROs over one year of continuous use of Medtronic’s 670G HCL in real-world clinical *** To assess the effects of hybrid closed loop system on glycemic control and quality of life in adults with *** We evaluated 71 patients with T1D(mean age:45.5±12.1 years;59%females;body weight:83.8±18.7 kg,body mass index:28.7±5.6 kg/m2,A1C:7.6%±0.8%)who were treated with HCL at Joslin Clinic from 2017 to *** measured A1C and percent of glucose time-in-range(%TIR)at baseline and 12 *** measured percent time in auto mode(%TiAM)for the last two weeks preceding the final visit and assessed PROs through several validated quality-of-life surveys related to general health and diabetes *** At 12 mo,A1C decreased by 0.3%±0.1%(P=0.001)and%TIR increased by 8.1%±2.5%(P=0.002).The average%TiAM was only 64.3%±32.8%and was not associated with A1C,%TIR or ***,provided at baseline and at the end of the study,showed that the physical functioning submodule of 36Item Short-Form Health Survey increased significantly by 22.9%(P1).Hypoglycemia fear survey/worry scale decreased significantly by 24.9%(Pdiabetes reduced significantly by-17.2%(P=0.002).The emotional burden submodules of dietary diversity score reduced significantly by-44.7%(P=0.001).Furthermore,analysis of Clarke questionnaire showed no increase in awareness of hypoglycemic ***-5 showed no improvements in subject’s wellbeing among participants after starting the 670G HCL ***,analysis of Pittsburgh Sleep Quality Index showed no difference in sleep quality,sleep latency,or duration o

Physical inactivity remains in high levels after cardiac surgery,reaching up to 50%.Patients present a significant loss of functional capacity,with prominent muscle weakness after cardiac surgery due to anesthesia,surgical incision,duration of cardiopulmonary bypass,and mechanical ventilation that affects their quality of *** complications,along with pulmonary complications after surgery,lead to extended intensive care unit(ICU)and hospital length of stay and significant mortality *** the well-known beneficial effects of cardiac rehabilitation,this treatment strategy still remains broadly underutilized in patients after cardiac *** and ICU early mobilization have been both showed to be valid methods to improve exercise tolerance and muscle *** mobilization should be adjusted to each patient’s functional capacity with progressive exercise training,from passive mobilization to more active range of motion and resistance *** exercise testing remains the gold standard for exercise capacity assessment and optimal prescription of aerobic exercise *** the last decade,recent advances in healthcare technology have changed cardiac rehabilitation perspectives,leading to the future of cardiac *** incorporating artificial intelligence,simulation,telemedicine and virtual cardiac rehabilitation,cardiac surgery patients may improve adherence and compliance,targeting to reduced hospital readmissions and decreased healthcare costs.

In this editorial,we comment on the article by Zeng et al published in the recent issue of the World Journal of diabetes in *** focus on the epidemiological,pathophysiological,and clinical interplay between obesity and type 1 diabetes mellitus(T1DM).Overweight and obesity represent a growing threat for modern societies and people with T1DM could not be an exception to this *** exogenous insulin administration,genetic and epigenetic factors,and psy-chosocial and behavioral parameters,along with the modern way of life that incorporates unhealthy eating patterns and physical inactivity,set the stage for the increasing obesity rates in *** our knowledge of the underlying mechanisms that lead to the development of obesity and hyperglycemia expands,it becomes clear that there are overlap zones in the pathophysiology of the two main types of *** regarding strict dividing lines between“autoimmune”and“metabolic”phenotypes increase the risk of trapping physicians into ineffective therapeutic approaches,instead of individualized diabetes *** this context,the use of adjuncts to insulin therapy that have the potential to alleviate cardiorenal risk and decrease body weight can reduce the burden of obesity in patients with T1DM.
