Translocation of telomerase reverse transcriptase coincided with ATP release in postnatal cochlear supporting cells

The spontaneous bursts of electrical activity in the developing auditory system are derived from the periodic release of adenosine triphosphate(ATP)by supporting cells in the Kölliker’s ***,the mechanisms responsible for initiating spontaneous ATP release have not been *** previous study revealed that telomerase reverse transcriptase(TERT)is expressed in the basilar membrane during the first postnatal *** role in cochlear development remains *** this study,we investigated the expression and role of TERT in postnatal cochlea supporting *** results revealed that in postnatal cochlear Kölliker’s organ supporting cells,TERT shifts from the nucleus into the cytoplasm over *** found that the TERT translocation tendency in postnatal cochlear supporting cells in vitro coincided with that observed in *** analysis showed that TERT in the cytoplasm was mainly located in mitochondria in the absence of oxidative stress or apoptosis,suggesting that TERT in mitochondria plays roles other than antioxidant or anti-apoptotic *** observed increased ATP synthesis,release and activation of purine signaling systems in supporting cells during the first 10 postnatal *** phenomenon that TERT translocation coincided with changes in ATP synthesis,release and activation of the purine signaling system in postnatal cochlear supporting cells suggested that TERT may be involved in regulating ATP release and activation of the purine signaling *** study provides a new research direction for exploring the spontaneous electrical activity of the cochlea during the early postnatal period.

GJB2 gene mutations are the most common causes of autosomal recessive non-syndromic hereditary *** individuals suffering from severe to profound GJB2-related deafness,cochlear implants have emerged as the sole remedy for auditory *** previous studies have highlighted the crucial role of preserving cochlear neural components in achieving favorable outcomes after cochlear ***,we generated a conditional knockout mouse model(Cx26-CKO)in which Cx26 was completely deleted in the cochlear supporting cells driven by the Sox2 *** Cx26-CKO mice showed severe hearing loss and massive loss of hair cells and Deiter’s cells,which represented the extreme form of human deafness caused by GJB2 gene *** addition,multiple pathological changes in the peripheral auditory nervous system were found,including abnormal innervation,demyelination,and degeneration of spiral ganglion neurons as well as disruption of heminodes in Cx26-CKO *** findings provide invaluable insights into the deafness mechanism and the treatment for severe deafness in Cx26-null mice.

Translocation of a fish spike from the pharynx to the thyroid gland:A case report

BACKGROUND A fish spike stuck in the throat is a common ear,nose,and throat(ENT)***,it is very rare for a fish spike to reach the thyroid tissue through the throat,which is very dangerous and can lead to pharyngeal fistula,cervical abscess,mediastinal abscess,and thyroid *** and timely management can help reduce complications,especially in elderly *** SUMMARY In the case presented here,the causative factor was dentures,but improper management aggravated the *** the case presented here,an elderly woman with a history of accidentally swallowing fish bones for 20 d had a sensation of foreign bodies in her ***,computed tomography(CT)of the neck showed that the left side of the thyroid gland had a dense shadow in the form of a *** If a fishbone foreign body is not visible during endoscopic examination but the patient has significant symptoms,the surgeon should be aware that the fishbone may be lodged in the *** avoid a misdiagnosis,ultrasound,CT,and other tests can be used to clarify the diagnosis.T The first step in treating a fish bone in the thyroid gland is to determine the position of the foreign body and the extent of the infection,and to develop a personalized surgical plan for its *** the same time,scientific information should be made available to the general public so that people know that if a fish bone is accidentally lodged,they should not force it to be swallowed or be spit out by inducing vomiting,which are incorrect methods and may aggravate the condition or even cause it to migrate outside the cavity,leading to serious complications,as in this reported case.

The application of the strip-shaped cymba conchae orthosis in the nonsurgical correction of complex auricular deformity

Objective: This study aims to evaluate the efficacy and safety of using a strip-shaped cymba conchae orthosis for the nonsurgical correction of complex auricular deformities. Methods: Clinical data were collected from 2020 to 2021 for 6 patients who underwent correction using a stripshaped cymba conchae orthosis. The indications, corrective effects, and complications associated with use of the orthosis were analyzed. Results: There were four indications for treatment: cryptotia with helix adhesion;cryptotia with grade I microtia;cryptotia with excessive helix thickness;and auricular deformity beyond the treatment time window(≥6 months). Excellent corrective effects were observed in all 6 patients. Complications occurred in one patient, who recovered after symptomatic treatment. Conclusion: The use of a strip-shaped cymba conchae orthosis alone or combined with a U-shaped helix orthosis presents a feasible approach for correcting complex auricular deformities or deformities beyond the treatment time window in pediatric patients.

Locoregionally advanced laryngeal and hypopharyngeal cancers(LA-LHCs)are traditionally treated with surgery followed by postoperative radiotherapy,which impairs speech and swallowing functions and reduces the quality of life.^(1,2)The use of induction chemotherapy(IC)as a larynxpreserving approach for LA-LHCs has been verified and refined.^(3)However,the short-term tumor response to IC varies,non-responders usually show poor survival and little benefit.

Objective Traditionally tumors of the parapharyngeal space(PPS)are resected through transcervical *** recent approaches include endoscopic approaches or transoral robotic surgery(TORS)without directions on when to use which *** objective was to find objective parameters to choose the most suitable *** It is a retrospective study containing 6 patients from May 2019 to May 2021 with tumors of the PPS treated in the department of otolaryngology and Head-Neck Surgery at the Hospital of Lucerne,*** The data was analysed in average 53 months after *** resection was completed with TORS in 3 patients and endoscopically in 3 *** operation time was 114 *** major complications *** evidence of tumor was found in magnetic resonance imaging studies postoperatively in all *** We conclude that a resection via TORS or endoscopic technique is safe and ***,we postulate that the further a tumor is located in the upper lateral area of the PPS,an approach via TORS is less possible.

According to the World Health Organization’s world report on hearing,nearly 2.5 billion people worldwide will suffer from hearing loss by 2050,which may contribute to a severe impact on individual life quality and national *** hearing loss(SNHL)occurs commonly as a result of noise exposure,aging,and ototoxic drugs,and is pathologically characterized by the impairment of mechanosensory hair cells of the inner ear,which is mainly triggered by reactive oxygen species accumulation,inflammation,and mitochondrial *** recent advances have been made in understanding the ability of cochlear repair and regeneration,there are still no effective therapeutic drugs for *** herbal medicine which is widely distributed and easily accessible in China has demonstrated a unique curative effect against SNHL with higher safety and lower cost compared with Western *** we present trends in research for Chinese herbal medicine for the treatment of SNHL,and elucidate their molecular mechanisms of action,to pave the way for further research and development of novel effective drugs in this field.

Airway management of a patient with linear immunoglobulin A bullous dermatosis:A case report

BACKGROUND There is limited literature on managing the airway of patients with linear immunoglobulin A(IgA)bullous dermatosis,a rare mucocutaneous disorder that leads to the development of friable *** clinical decision making is necessary when there is a risk of bleeding into the airway,and a multidisciplinary team approach may lead to decreased patient morbidity during these high-risk scenarios,especially when confronted with an unusual cause for *** SUMMARY A 45-year-old African American female presented to our ambulatory surgical center for right corneal transplantation due to corneal perforation after blunt trauma in the setting of cicatricial conjunctivitis and diffuse corneal neovascularization from linear IgA bullous *** diagnosis of IgA dermatosis was recent,and the patient had been lost to *** severity of the disease and extent of airway involvement was unknown at the time of the *** airway bleeding was noticed upon intubation and the otorhinolaryngology team had to be called to the operating *** patient required transfer to the intensive care unit where a multidisciplinary team was involved in her *** patient was extubated on postoperative day *** A multidisciplinary approach to treating this disease is the best course of action before a surgical *** our case,key communication between the surgery,anesthesia,and dermatology teams led to the quick and safe treatment of our patient’s *** surgery should not be considered for these cases unless they are in full remission and there is no mucous membrane involvement.

The construction and functionalization of lanthanide-organic cages have been a research hotspot in coordination *** from the unique luminescent and magnetic properties of lanthanide ions,the potential applications of lanthanide-organic cages in biological imaging applications are promising,but only a few corresponding explorations have been *** rein,we introduced a series of lanthanide tetrahedral cages,(Eu_(n)Gd_(4-n)L_(4))(sol)_8(n=0,1,2,3 and 4,where L=(4,4',4"-tris(4,4,4-trifluoro-1,3-dioxobutyl)-triphenylamine,and sol=dimethyl sulfoxide and methanol),with potential applications in biological *** the good luminescent and magnetic properties of Eu^(3+) and Gd^(3+) ions,Eu_(4)L_(4) and Gd_(4)L_(4) complexes show a high luminescence quantum yield of 29% in DMSO and large longitudinal relaxivity(r_(1)) of 11.4(mmol/L)^(-1)/s at 0.5 T in water containing 1 vol% DMSO,*** heterobimetallic Eu-Gd complexes achieve a dual imaging mode by combining luminescent and magnetic centers in one *** the Gd:Eu ratio increases from 1:3 to 3:1,the r_(1) values increases from 5.33 to 8.64(mmoI/L)^(-1)/s,higher than that of commercial contrast agent Gd-DOTA(3.23(mmol/L)^(-1)/s).Owing to their low toxicity and good cell imaging ability toward ECA 109 cells,the selfassembled Eu-Gd heterobimetallic tetrahedral cages could be potential candidates for the multimodal imaging contrast agent.
