Succession of bacteria communities during production and application of dairy bedding by membrane-covered aerobic fermentation

The cost of dairy manure treatment and bedding material purchase increases the operating cost of the dairy ***-covered aerobic fermentation system has been widely used for dealing with dairy manure and recycling the final product as bedding ***,the microbial safety in each processing step is still *** better understand the bacterial community dynamics during the whole bedding conversion process,a full-chain and large-scale experiment including 1.-day membrane-covered aerobic fermentation and 1.-day bedding material application was *** results showed that the pile temperatures in the fermentation stage rapidly increased to 80°C and maintained>50°C for more than 1. days and the use of fermentation product as bedding material provided cows with a stable and comfortable bedding *** Chao1.and Shannon index decreased at the end of the fermentation stage and remained stable in the application stage,indicating that membrane-covered aerobic fermentation effectively killed some pathogenic bacteria and guaranteed both the maturity and stability of the final *** dominant bacteria in the fermentation stage were Acinetobacter,Thermus,and Rhodothermus at genus *** common potential pathogens of mastitis(Staphylococcus,Enterococcus,Serratia,Pseudomonas,Corynebacterium,Mycobacterium,and Bacillus)were found at the end of fermentation stage but the relative abundance was low(0.0025%-0.2727%).The dominant bacteria in the application stage mainly included Acinetobacter,Pseudomonas,and Flavobacterium at the genus *** relative abundance of Pseudomonas increased in the application stage,which was a reminder to the dairy farm to pay attention to the disinfection and timely replacement of bedding material to prevent the occurrence of dairy *** results of this study contributed deep understanding of the microorganism-driven bedding conversion process and provide practical guidance and cautions for the bedding

Reductive soil disinfestation(RSD)is an important tool for sustainable agricultural ***,the differences in soil bacterial communities and their community assembly processes among RSD and other treatment strategies(e.g.,biochar and chemical fumigation)are still subject to open *** this study,soils subjected to various treatments–un-treated control(CK),chemical soil fumigation with CaCN2(CF),1.biochar(1.B),3%biochar(3%B),and reductive soil disinfestation(RSD)are *** samples were collected,incubated,and then used for growth of tomato *** Sloan neutral community model indicates that stochastic processes dominate in bacterial community assembly for both biochar and CF *** contrast,this work shows that RSD treatment can have a strong impact on soil bacterial community *** relative abundance of Firmicutes increased during unplanted soil incubation,whereas Proteobacteria and Bacteroidetes dominated in the rhizosphere after planting of *** stochasticity ratio reveals that deterministic selection played an important role in the bacterial assembly under RSD *** found that RSD amendment yielded lower biomass than that for other treatments after 28 days of tomato *** results suggest that although RSD treatment has great potential to rebuild soil bacterial ecology by shaping bacterial communities and their assembly processes,it is important to monitor and manage soil conditions(e.g.,soil nutrients or physical properties)before planting to ensure plant productivity.

Study on the Regulation of ROS by Acupuncture and Moxibustion to Relieve Inflammation in the Treatment of Sciatica

Objective: To observe the analgesic effect of acupuncture and moxibustion on chronic constriction injury (CCI) pain model rats, and to investigate the mechanism of constriction constriction injury. Methods: 32 SPF SD rats were divided into Control Group, Sham Surgery Group, Model Group and Acupuncture and moxibustion Group, with 8 rats in each group. The Model Group, Acupuncture and moxibustion Group and sciatic nerve ligation were used to establish the CCI pain rat model. The sham operation group only separated the nerve, and the control group did not do any treatment. After modeling, the Acupuncture and moxibustion Group was given acupuncture treatment, while the control group and the Model Group did not do any intervention. 7 days for 1.course of treatment, continuous treatment for 3 courses of sampling and detection. Paw withdrawal mechanical threshold (PWMT) and Paw withdrawal thermal latency (PWTL) were observed. Biochemical detection of reactive oxygen species (ROS), malondialdehyde (MDA), Super Oxide Dismutase (SOD), glutathione (GSH);Serum, tumor necrosis factor-α (TNF-α), interleukin-1. (IL-1.) and interleukin-6 (IL-6) levels were detected by ELISA. Results: Compared with the control group, PWMT and PWTL in the Model Group were decreased (P α, IL-1. and IL-6 levels or contents were increased (P α, IL-1. and IL-6 levels or contents were decreased (P Conclusion: Acupuncture and moxibustion can reduce the ROS level, enhance the antioxidant capacity of the body, reduce the damage of sciatic nerve caused by inflammation, and relieve sciatic pain.

T-cell acute lymphoblastic leukemia(T-ALL),a heterogeneous hematological malignancy,is caused by the developmental arrest of normal T-cell *** development of targeted therapeutic regimens is impeded by poor knowledge of the stage-specific aberrances in this *** this study,we performed multi-omics integration analysis,which included mRNA expression,chromatin accessibility,and gene-dependency database analyses,to identify potential stage-specific druggable targets and repositioned drugs for this *** multi-omics integration helped identify 29 potential pathological genes for *** genes exhibited tissue-specific expression profiles and were enriched in the cell cycle,hematopoietic stem cell differentiation,and the AMPK signaling *** these,four known druggable targets(CDK6,TUBA1.,TUBB,and TYMS)showed dysregulated and stage-specific expression in malignant T cells and may serve as stage-specific targets in *** TUBA1. expression level was higher in the early T cell precursor(ETP)-ALL cells,while TUBB and TYMS were mainly highly expressed in malignant T cells arrested at the CD4 and CD8 double-positive or single-positive ***6 exhibited a U-shaped expression pattern in malignant T cells along the naıve to maturation ***,mebendazole and gemcitabine,which target TUBA1. and TYMS,respectively,exerted stage-specific inhibitory effects on T-ALL cell lines,indicating their potential stage-specific antileukemic role in ***,our findings might aid in identifying potential stage-specific druggable targets and are promising for achieving more precise therapeutic strategies for T-ALL.

In-situ electrochemical study on the eff ects of Fe(Ⅲ)on kinetics of pyrite acidic pressure oxidation

Fe(Ⅲ)has been proved to be a more eff ective oxidant than dissolved oxygen at ambient temperature,however,the role of Fe(Ⅲ)in pyrite acidic pressure oxidation was rarely discussed so *** this paper,in-situ electrochemical investigation was performed using a flow-through autoclave system in acidic pressure oxidation *** results illustrated that increasing Fe(Ⅲ)concentrations led to raising in redox potential of the solution,and decreased passivation of pyrite caused by deposition of elemental *** of Fe(Ⅲ)at pyrite surface was a fast reaction with low activation energy,it was only slightly promoted by rising ***,the oxidation rate of pyrite at all investigated Fe(Ⅲ)concentrations increased obviously with rising temperatures,the anodic reaction was the rate-limiting step in the overall *** energy of pyrite oxidation decreased from 47.74 to 28.79 kJ/mol when Fe(Ⅲ)concentration was increased from 0.05 to 0.50 g/L,showing that the reaction kinetics were limited by the rate of electrochemical reaction at low Fe(Ⅲ)concentrations,while,it gradually turned to be diffusion control with increasing Fe(Ⅲ)concentrations.

In fractured geothermal reservoirs,the fracture networks and internal fluid flow behaviors can significantly impact the thermal *** this study,we proposed a non-Darcy rough discrete fracture network(NR-DFN)model that can simultaneously consider the fracture evolution and non-Darcy flow dynamics in studying the thermo-hydro-mechanical(THM)coupling processes for heat extraction in geothermal *** further employed the model on the Habanero enhanced geothermal systems(EGS)project located in ***,our findings illustrate a clear spatial-temporal variation in the thermal stress and pressure perturbations,as well as uneven spatial distribution of shear failure in 3D fracture *** shear failure is mainly concentrated in the first fracture ***,channeling flow have also been observed in DFNs during heat extraction and are further intensified by the expansion of fractures driven by thermal ***,the combined effect of non-Darcy flow and fracture evolution triggers a rapid decline in the resulting heat rate and *** NR-DFN model framework and the Habanero EGS's results illustrate the importance of both fracture evolution and non-Darcy flow on the efficiency of EGS production and have the potential to promote the development of more sustainable and efficient EGS operations for stakeholders.

PbrARF4 contributes to calyx shedding of fruitlets in ‘Dangshan Suli’ pear by partly regulating the expression of abscission genes

Fruitlet calyx shedding in pear plants is apparently regulated via numerous pathways that involve both environmental triggers and phytohormones cues such as auxin. In this study, we found at 1. days after full bloom (DAFB) higher levels of indoleacetic acid (IAA) and tryptophan (Trp) in calyx persistence fruitlet (CPF) than calyx shedding fruitlet (CSF) ofDanshan Suli’ pear (Pyrus bretschneideri Rhed.). Consisting with this, the activity of indolealdehyde oxidase (IAAIdO), which promotes IAA synthesis, was remarkably increased, and that of peroxidase(POD), which degrades IAA, dropped markedly in CPF but not in CSF. Further, qRT-PCR results revealed that most of 31.PbrARFs (encoding auxin response factors) in Pyrus bretschneideri were highly expressed in CPF, whereas PbrARF4, PbrARF24 and PbrARF26 were significantly downregulated in CPF vis-a-vis CSF. Phylogenetic analysis revealed that 6 PbrARFs clustered in the group III, where PbrARF4 showed the closest affinity with AtARF1.that promotes organ abscission, indicating a putative role of PbrARF4 in mediating the process of calyx shedding in pear. In fact, the ectopic overexpression of PbrARF4 in Solanum lycopersicum resulted in an earlier-formed and deeper abscission layer (AL) in the transgenic plants, whose calyxes were more prone to wilt at the mature red stage (MR) compared with the control plants (wild-type). More importantly, expression levels of the abscission genes SILS and Sl Cel2 in transgenic plants overexpressing PbrARF4 were significantly upregulated in comparation with the WT, whereas those of Sl BI and Sl TAPG2 were considerably inhibited. Further, PbrJOINTLESS and PbrIDA,the two genes related to calyx shedding in pear, were up-regulated more in CSF than CPF. The findings contribute to a better understanding of PbrARFs involved in fruitlet calyx shedding of pear, which could prove beneficial to improving the quality of pear fruit.
