Objective In vivo multi-channel extracellular recording provides the neuroscientists a new insight into the functional mode of a large group of neurons at the neuronal circuitry ***,different types of neurons in a local circuit can be recorded simultaneously and a rough way to distinguish these cell types is based on the waveform differences of the action *** we introduced optogenetic tools to target the specific type of the neurons（such as the pyramidal cells in neocortex） and recorded the population responses following the optical stimulation in order to identify the targeted neurons’ *** We expressed channelrhodopsin-2（ChR2） under the control of special promoter to target a specific type of neurons（such as using CaMKIIα promoter to label the excitatory pyramidal cells）,and then we implanted an optrode system into the virus transfection *** stimulation evoked many multiunit activities and we well isolated some single *** set up 3 necessary conditions to pick out a ChR2-positive cell:（1） firing probability following the light pulses ＞ 90%;（2） response latency ＜ 3 ms;（3） the waveform correlation between spontaneous and light-evoked spike ＞ *** We recorded and identified 15neurons as the ChR2- positive cell in 3 mice and all of them showed classic pyramidal cell firing *** We identified the specific type of neurons through multi-channel extracellular recording results,confirming it will be an useful tool to distinguish the functions of different type of neurons in a specific behavior in the future study.
