Tracking Tonoplast Protein Behaviors in Intact vacuoles Isolated from Arabidopsis Leaves

Dear Editor Membrane proteins perform a vast array of functions in vital cellular activities, including transport of molecules, responses to stimuli, and regulation of immune reactions （Proft et al., 2006; Bohm et al., 2014）. Real-time behaviors of proteins, such as their lateral diffusion and dynamic distribution, affect their ac- tivity and functions （Bai et al., 2008）. Consequently, the tracking and analysis of the behaviors of membrane proteins in living cells can further our understanding of the mechanisms regulating the biological functions of proteins.

Simplification of vacuole structure during plant cell death triggered by culture filtrates of Erwinia carotovora

vacuoles are suggested to play crucial roles in plant defense-related cell death. During programmed cell death, previous live cell imaging studies have observed vacuoles to become simpler in structure and have implicated this simplification as a prelude to the vacuole＇s rupture and consequent lysis of the plasma membrane. Here, we examined dynamics of the vacuole in cell cycle-synchronized tobacco BY-2 （Nicotiana tabacum L. cv. Bright Yellow 2） cells during ceil death induced by application of culture filtrates of Erwinia carotovora. The filtrate induced death in about 90% of the cells by 24 h. Prior to cell death, vacuole shape simplified and endoplasmic actin filaments disassembled; however, the vacuoles did not rupture until after plasma membrane integrity was lost. Instead of facilitating rupture, the simplification of vacuole structure might play a role in the retrieval of membrane components needed for defense-related cell death.

Cryopreservation enhances vacuolization in human spermatozoa

Objective:To evaluate the impact of freezing–thawing on the human sperm head vacuoles and the potential value of motile sperm organelle morphology examination for selection of frozen-thaw spermatozoa.Methods: In 30 sperm samples from infertile men, analysis for conventional sperm parameters (motility, vitality, and normal morphology) and a morphological analysis at high magnification for vacuoles examination were done before cooling and after warming. For description of sperm head vacuoles, two hundred spermatozoa were examined and were classified into three groups according to presence and vacuole areas including no vacuole group (free of any vacuole), small vacuole group (occupy not more than 4% of the nuclear area), and large vacuole group (occupy more than 4% of the normal nuclear area).Results:Significant reduction of progressive motility and vitality was observed following cryopreservation (Pvacuoles increased slightly, but not significantly after cryopreservation (P=0.296).Conclusions:Motile sperm organelle morphology examination is a powerful research tool for investigating spermatozoa abnormalities such as vacuoles that are increased post cryopreservation.
