The Effects of Low Intensity Endurance Activity on Various Physiological Parameters and Exercise Induced Oxidative Stress in Dogs

The study of canine athlete physiology has focused on endurance sled dog racing and high intensity short duration Greyhound racing, yet the number of dogs competing in low intensity endurance activities is rising due to the increased numbers of hunting and companion dog activities. There is little information on the physiological effect of longer duration low intensity endurance activities. We set out to evaluate the serum biochemistry, oxidative stress, and cortisol response before and after two consecutive days of exercise in ten healthy unconditioned male dogs. Exercise sessions consisted of 120 minutes on an exercise wheel at 11 km/hour on 2 consecutive days. Blood was collected at four time points: 24 hours pre-exercise (sample 1, Day 0, resting);2 min post-exercise on days 1 and 2, (samples 2 and 3, respectively);and 20 hours post-exercise, collected on day 3(sample 4). Hematocrit, blood gases, serum chemistry, uric acid, cortisol, and F2-isoprostanes were determined. Serum biochemistry and hematocrit suggested hemoconcentration, mild muscle damage and respiratory alkalosis during exercise, which was expected in the unconditioned canine athlete. In addition, plasma indices of oxidative damage (F2-isoprostanes) increased, as did plasma uric acid (an endogenous antioxidant). Importantly, similar to human studies, plasma F2-isoprostanes decreased 24 hours after exercise suggesting a protective effect of exercise. Serum cortisol concentrations were also markedly elevated at the end of exercise on both days suggesting that timing of sampling may play a role in interpreting cortisol results when looking at previous field studies.

Polyphenolic compounds, widely distributed in plant kingdom, have been exhaustively studied for their bioactive properties specially antioxidant activity. However, they are extensively metabolized by human organism and the resulting metabolites are largely responsible for their effects. Furthermore, they may interact with the endogenous antioxidant network being this possibility scarcely studied. Plasma antioxidant network encompasses antioxidant enzymes and other substances such as uric acid. In addition, ascorbic acid is the major compound representing water soluble compartment both in foods and human body. The interaction of this vitamin with phenolic compound is largely unexplored. This work aims to study if there is a synergic effect between phenolic metabolites and main antioxidants (uric and ascorbic acid). For this purpose, the antioxidant activity was evaluated in terms of ORAC (oxygen radical absorbance capacity) and FRAP (ferricreducing antioxidant power) as these tests involved either HAT (Hydrogen Atom Transfer) or SET (Single Electron Transfer) mechanisms. Additionally, a kinetic studied was developed to test if the rate constant presented a synergic effect. Protocatechuic acid, 3,4-dihydroxyphenylacetic acid, 3,4-dihydroxyphenylpropionic acid and 3-hydroxyphenylacetic acid were selected as they were metabolites of polyphenol compounds such as anthocyanins, quercetin, neohesperidin, chlorogenic acid and hesperetin present in wines, orange and strawberries. A synergic effect was proved for the combination of ascorbic acid with 3,4-dihydroxyphenylacetic acid, both in terms of antioxidant activity and potent increase of velocity of the antioxidant reaction that took place.

Gout is a form of inflammatory arthritis characterized by sharp pain and severe swelling that often causes severe physical disability. Gout is caused by the chronic elevation of uric acid levels in the blood and is known as the disease of kings due to its strong association with a diet rich in fructose and beer. Recent studies suggest that a high uric acid concentration is the result of a dynamical process that highlights the interactions between leptin production, insulin resistance, low muscle mass and a diet rich in fructose. Once individuals develop hyperuricemia, reach a high uric acid concentration in excess of 7 mg/dL for men and 6 mg/dL for women, they become susceptible to developing gout. We propose a novel dynamic system to analyze and determine the connections between a diet involving different levels of fructose (in both adult men and women in the U.S.) and the concentration of uric acid in the blood. Our model simulations suggest that adult males under a diet containing levels of fructose stimulating a 0.5 uric acid growth rate, could develop hyperuricemia after around 10,000 days, while it only takes women about 5000 days with a diet stimulating a 0.4 growth rate.

Introduction: To investigate the relationship of xanthine oxidase activity with nephrotic syndrome disorder in children, and the optimization of the enzyme activity conditions in the disorder. Material and methods: Sera of children with nephrotic syndrome (NS) (60 samples) were obtained from Central Child Teaching Al Karama Hospital (CCTAH), from 2nd Mar. 2013 to 28th Feb. 2014. Sera of the patients were assayed for xanthine oxidase (XO) activity using colorimetric absorbance technique. The obtained results were compared with the enzyme activity of normal children (70 samples) as control. Results and conclusions: The results revealed a significant (P uric acid level in sera of NS patients (497.52 ± 3.21 μmol/L) compared with that in normal group (298.12 ± 1.70 μmol/L). Elevation was found in urea level in sera of NS patient (10.69 ± 7.55 mmol/L) compared with that of normal group (4.57 ± 1.27 mmol/L). It was appeared, there is a role of XO activity in the pathogenesis of endothelial injury during glomerular lesion in NS and that was confirmed by comparing XO activity and other related conditions with the activity of normal volunteers.

Background: Metabolic syndrome is the major cause for life threatening disorders such as cardiovascular diseases and type 2 diabetes. These disorders are associated with hyperuricemia and the number is growing among the urban population. Methods: A cross sectional study was done among Kodava population by conducting health camps in Mysore district. Metabolic syndrome was defined according to Joint Interim Statement criteria. Anthropometry was done and blood pressure readings were noted. Clinical markers like fasting glucose, triglyceride, high density lipoprotein, CVD markers and uric acid levels were analyzed. Results: The prevalence of metabolic syndrome was 60.77% and the utmost occurrence was in 41 - 60 age groups. Women were more affected than men (31.58%) and MetS became pronounced with advance of age. Biochemical levels of C-reactive protein, ApolipoproteinB/ApolipoproteinA1 ratio and uric acid wereraised (P uricemic conditions. Life style modifications and therapeutic approach are critical prerequisite. However, there is an urgent need for further health camps for the awareness, and prevention of MetS and its associated risk factors among Kodavas.

Objective: To study the correlation of serum Hcy and UA contents with coronary plaque property changes in diabetic patients with coronary heart disease. Methods: Patients who were diagnosed with type 2 diabetes mellitus in Dongguan Branch of Yan'an University Affiliated Hospital between May 2013 and June 2017 were selected and divided into the CHD+DM group who were complicated by coronary heart disease and the DM group who were not complicated by coronary heart disease according to the results of coronary CTA, and the healthy subjects who received physical examination in Dongguan Branch of Yan'an University Affiliated Hospital during the same period were selected as control group. The contents of homocysteine (Hcy), uric acid (UA), platelet activation indexes and protease indexes in serum were measured. Results: Serum Hcy, UA, sSema4D, sP-selectin, sE-selectin, MMP1, MMP3, CatK and ADAMTS4 contents of DM group and CHD+DM group were greatly higher than those of control group whereas Klotho and TIMP2 contents were greatly lower than those of control group;serum Hcy, UA, sSema4D, sP-selectin, sE-selectin, MMP1, MMP3, CatK and ADAMTS4 contents of CHD+DM group were greatly higher than those of DM group whereas Klotho and TIMP2 contents were greatly lower than those of DM group;serum sSema4D, sP-selectin, sE-selectin, MMP1, MMP3, CatK and ADAMTS4 contents of CHD+DM group were positively correlated with Hcy and UA contents while Klotho and TIMP2 contents were negatively correlated with Hcy and UA contents. Conclusion: The abnormal increase of serum Hcy and UA contents in diabetic patients with coronary heart disease can promote the activation and aggregation of platelet, the degradation of plaque fibrous cap and the decrease of plaque stability.

In this study,the changes of clinical symptoms and signs,contents of uric acid inthe blood and urine and RNAase activities were observed to investigate the therapeutic role of prickingblood plus cupping in the treatment of gouty *** was observed that the effective rate was100％in the pricking blood group and 68.43％in the medicine ***,there was a statisticallysignificant difference in chemical indexes between the two *** results indicate that prickingblood may cure the disease by promoting the excretion of uric acid,inhibiting the synthesis of uric acidand regulating the metabolism of pruines in organism.
