DNA hybridization chain reaction and DNA supersandwich self-assembly for ultrasensitive detection

The fabrication of sensitive sensors with high selectivity is highly desirable for the detection of some important biomarkers,such as nucleic acids,proteins,small molecules and *** hybridization chain reaction(HCR) and DNA supersandwich self-assembly(SSA) are two prevalent enzyme-free signal amplification strategies to improve sensitivity of the *** this review,we firstly describe the characteristics about DNA HCR and DNA SSA,and then summarize the advances in the one-dimensional DNA nanostructures assisted by HCR and *** review has been divided into three parts according to the two signal amplification methods and highlights recent progress in these two strategies to improve the detection sensitivity of proteins,nucleic acids,small molecules and ions.

Conducting polymer engineered covalent organic framework as a novel electrochemical amplifier for ultrasensitive detection of acetaminophen

Two-dimensional covalent organic framework(COF)has distinctive properties that offer potential opportunities for developing advanced electrode *** this work,a core-shell material composed of TAPB-DMTP-COF(TAPB,1,3,5-tris(4-aminophenyl)benzene;DMTP,2,5-dimethoxyterephaldehyde)core and conducting polymer shell,TAPB-DMTP-COF@PANI,was synthesized solvothermally using a polymerization *** structural cha racteristics of the prepared composite were revealed by X-ray diffraction patterns(XRD),fourier transform infrared spectra(FTIR),X-ray photoelectron spectroscopy(XPS),transmission electron microscopy(TEM).The electrochemical analyses were verified by subsequent monitoring of trace levels of *** resultant composite not only facilitated acetaminophen to interact with absorption sites byπ-πstacking effect and hydrogen bonding but also overcame the poor conductivity of *** the optimal conditions,a low limit of detection of 0.032μmol/L and wide linear range of 0.10-500μmol/L were *** electrochemical platform was almost unaffected by other interfering substances,and successfully applied for the practical detection of acetaminophen in commercial tablet,human blood serum and *** enhanced performance makes this COF based core-shell composite a promising material in electrochemical senso r.

In this manuscript,we first report an ultrasensitive detection assay of microRNA by combing asymmetric polymerase chain reaction(A-PCR)and loop-mediated isothermal amplification(LAMP)*** A-PCR obtained an extended single strand to form LAMP stem-loop structure under isothermal amplification *** used miRNAs as a loop primer probe in LAMP reaction and completed its ultrasensitive and rapid *** established method furnished a fast,specific and efficient detection of target miRNA with a detection limit as low as 10 amol/L in 90 min.

In this article, we report a novel approach for the development of ultrasensitive multi-analytes detection and ***, we used a novel phthalimide-functionalized pillar[5]arene(PF) and a bis-bromohexane-functionalized pillar[5]arene(BF) to construct a linear supramolecular polymer(PFBF). Then, the obtained linear supramolecular polymer could form a stable two-dimensional supramolecular polymer gel(PFBF-G) in cyclohexanol with strong aggregation induced emission(AIE). Interestingly, the PFBF-G shows ultrasensitive fluorescent response for Fe, Hg and Ag in gel states, by rationally introduced Fe and Hg into the PFBF-G, the obtained metallogels(PFBF-FeG and PFBF-HgG) could ultrasensitive fluorescently sense Fand Br, *** importantly, the xerogel of PFBF-G shows excellent separation properties for these metal ions and the absorption rates were up to 99.95%

Protein biomarker detection are crucial in the clinical diagnosis 1. In recent years, Mass spectrometry(MS) has been wildly applied biomarker analysis either in proteomic aspect or target biomarker detection because of its low detection limits, large dynamic range and better spectral resolution. However, ultrasensitive direct detecting of large biomolecules, such as proteins and DNA etc., remains challenging. Signal amplification strategies using small molecules as mass reporters enable the higher sensitive detection of large molecules 2, 3. Different from conventional ionization technique, ambient ionization technique offers MS more possibilities in fast screening and detection. In our work, novel mass tag was designed and synthesized. Combining ambient ionization MS with the signal amplification strategy, the ultrasensitive detection method was established for DNA and protein biomarkers. Attomolar detection limit of target analytes were achieved through this approach, which showed great promise for large biomolecules detection in early disease diagnosis and single-cell analysis.
