Increasing evidence underscores the vital significance of diverse mast cell mediators in inflammatory illness,yet diagnostic criteria for mast cell activation syndrome still focus predominantly on a solitary mediator,*** interventions,conversely,tend to prioritize histamine,prostaglandins,and *** article,via a review of both experimental and clinical findings,spotlights the mechanisms of action of an important mast cell mediator–tumor necrosis factor(TNF),and elucidates the intricate linkages between mast cell dysfunction,chronic illness,and *** providing an overview of the role of mast cell-derived TNF in inflammatory illnesses and viral infections,we hypothesize the interplay between mast cell-derived TNF and mitochondrial dysfunction,microglial activation,and the hypothalamic-pituitary-adrenal axis and the pathways of action on mast cells of treatments such as high-dose melatonin and low-dose naltrexone.

Objective: To evaluate the dynamic change of Th1/Th2 cytokines in serum, peritoneal lavage fluid (PLF) and splenic macrophages (SM) in rats with severe peritonitis, and to observe the therapeutic effects of recombinant interleukin-12 (rIL-2) and Shenmai injection (SMI, 参麦注射液), a Chinese medicinal ***: Severe peritonitis (SP) model was induced by intraperitoneal injection of E. coli and B. frag, and mild peritonitis (MP) model was induced by cecal ligation and punching. Then the following experiments were done: (1) Survival rates of animals after every 6 hrs in the 72 hrs after modeling were recorded, serum and PLF levels of cytokines, including tumor necrosis factor-α (TNF-α), interferon-γ (IFN-γ), and interleukin-10 (IL-10), 6 hrs, 12 hrs, 24 hrs and 48 hrs after modeling were measured. (2) Model rats were treated with rIL-12 or SMI, the survival rate was recorded and serum levels of TNF-α, INF-γ, and IL-10 before and after treatment were measured, and (3) amount of these cytokines produced by SM were determined 6 hrs, 12 hrs and 24 hrs after treatment. The survival rates and levels of cytokines were then compared between the groups (model group treated with rIL-12 or SMI, untreated model group, and blank group).Results: Serum and PLF levels of IFN-γ, TNF-α at all the time points in SP rats were significantly lower than those in MP rats while those of IL-10 6 hrs and 12 hrs after modeling were significantly higher in the former than that in the latter (Pfactor

Objective:The aim of our study was to investigate the immune status of patients with rectal cancer and its relationship with clinicopathological ***:The serum levels of interleukin-8(IL-8),tumor necrosis factor(TNF-α) and T-cell subgroup contents were measured using a double-antibody sandwich assay of ELISA in 43 patients with rectal cancer,and compared with the normal health ***:In patients with rectal cancer,the serum levels of CD4,CD4/CD8 of T-cell subgroup in peripheral blood were significantly lower than the control group(P < 0.01),which gradually decreased with increase of Dukes stage;but the levels of CD8,IL-8 and TNF-α were higher than the control group,which gradually increased with increase of Dukes ***:The immunocompromice exists in patients with rectal cancer,there is a correlation between the contents of T-cell subgroup,IL-8 and TNF-α in serum and the Dukes stage of rectal *** immunotherapy can be used in patients with rectal cancer.

Objective： To observe the clinical efficacy of kidney-nourishing and governor vessel-regulating therapy for apoplexy sequela. Methods： Sixty subjects were equally randomized into observation group and control group, and respectively treated for 35 days. The scores of survival quality, interleukin-6（IL-6）, interleukin-8 （IL-8） and tumor necrosis factor（TNF） were measured, and the clinical efficacy were compared between two groups. Results and Conclusion： After treatment, all the observational items were improved, with better results in observation group than in control group. This therapy has better effects than regular method in the treatment of apoplexy sequela.

In this study,Protocatechuic acid(Pac), a water-soluble component of Salvia Miltiorriza Bge, on theability of the vascular endotheliun to respond to tumor necrosis factor is inveshgated. In the presenceof a protein srpthesis inhibitor, actinomycin D, tumor necrosis factor(TNF-a)induces humanumbilical vein endothelial cells(HUVECs)to undergo apoptosis. Pretreating HUVECs with Pac for 2h,enhances survival cells for 1.5 fold, suggesting that Pac inducible cytoprotective pathway occurs forcell survive. Objective:To examine whether lipoxin A 4(LXA 4) has an inhibitory effect on tumor necrosis factor-α(TNF-α)-induced proliferation of glomerular mesangial cells of rat, and explore the molecular mechanisms of signal pathway in LXA 4 actions. Methods: Glomerular mesangial cells of rat were cultured and treated with TNF-α(10 ng/ml), with or without preincubation with LXA 4 at different concentrations. Cell proliferation was evaluated by monotetrazolium (MTT) colorimetric assay. The expression of cyclin E mRNA was measured by RT-PCR. Phosphorylated Akt1(Thr308) and p27 kip1 were analyzed by Western blotting. Results: TNF-α-stimulated proliferation of mesangial cells was inhibited by LXA 4 in a dose-dependent manner. The marked increments in cyclin E mRNA expression induced by TNF-α during proliferation of mesangial cells were down-regulated by LXA 4. Threonine phosphorylated Akt1 proteins at 308 site stimulated by TNF-α was reduced by LXA 4. TNF-α-induced decrements in expression of p27 kip1 proteins was ameliorated by LXA 4 in a dose-dependent manner. Conclusion: TNF-α-induced proliferation of rat mesangial cells can be inhibited by TXA 4 through the mechanism of Akt 1/p27 kip1 pathway-dependent signal transduction.

Objective: To investigate the role of Biyan Qingdu Granule combined with micro-radiation in suppressing nasopharyngeal carcinoma and to explore its mechanism of action. Methods: FAT cells of nasopharyngeal carcinoma were injected into the hind limb axilla of F344 rats to establish a rat nasopharyngeal carcinoma tumor model. X-ray radiation was applied to the oral cavity of the rats, delivering a micro-dose of radiation to the distant hind limb axilla tumor. The general condition of the rats, including oral mucosal inflammation, the size of the nasopharyngeal tumor mass, pathological changes in the tumor tissue, and the levels of TNF-α, IL-1β, IL-6, IL-18, IL-12, ICAM-1, and VCAM-1 in the tumor tissue, were assessed. Results: Oral administration of Biyan Qingdu Granule improved the rats’ overall condition and reduced oral mucosal inflammation. Pathological examination revealed tumor cell degeneration and necrosis. ELISA analysis of tumor tissue showed significant reductions in TNF-α, IL-1β, IL-6, ICAM-1, and VCAM-1 levels, along with a notable increase in IL-18 levels. Conclusion: Biyan Qingdu Granule, when combined with micro-radiation, can inhibit tumor growth and reduce tumor volume. Its mechanism of action may involve enhancing immune function and suppressing inflammatory factors.
