Ageing exacerbates ribosome pausing to disrupt cotranslational proteostasis

Ageing is accompanied by a decline in cellular proteostasis,which underlies many age-related protein misfolding diseases1,2.Yet,how ageing impairs proteostasis remains unclear.As nascent polypeptides represent a substantial burden on the proteostasis network3,we hypothesized that altered translational efficiency during ageing could help to drive the collapse of proteostasis.Here we show that ageing alters the kinetics of translation elongation in both Caenorhabditis elegans and Saccharomyces cerevisiae.Ribosome pausing was exacerbated at specific positions in aged yeast and worms.

Targeting apoptosis is one of the major strategies for cancer therapy. Essentially, most of the conventional cancer therapeutic drugs that are in the clinical use induce apoptosis and in part necrosis of malignant cells and therefore prevent cancer progression and metastasis. Although these cytotoxic anticancer drugs are important weapons for killing cancers, their toxic side effects limited their application. The molecularly targeted therapeutics that are based on the deeper understanding of the defects in the apoptotic signaling in cancers are emerging and have shown promising anticancer activity in selectively killing cancers but not normal cells. The examples of molecular targets that are under exploration for cancer therapy include the cell surface receptors such as TNFR family death receptors, the intrinsic Bcl-2 family members and some other intracellular molecules like p53, MDM2, IAP, and Smac. The advance in the high-throughput bio-technologies has greatly accelerated the progress of cancer drug discovery.

This study characterizes the 19 kDa protein expressed by Mycobacterium avium subspecies paratuberculosis (MAP) as a glycolipoprotein, providing the foundation for future experiments regarding its antigenicity and role in disease pathogenicity. We have previously shown that a 4.8 kb insert from MAP will produce a 16 kDa recombinant protein when expressed in Escherichia coli and 19 kDa recombinant protein when expressed in M. smegmatis (smeg19K). The difference of 3 kDa in size of these expressed proteins may be related to post translational modifications that occur in Mycobacterium species. We hypothesized that smeg19K is a glycolipoprotein since BLAST analysis revealed approximately 76% amino acid identity between the MAP 19 kDa protein and a known lipoglycoprotein, the 19 kDa protein of M. tuberculosis. This prediction was confirmed by the following positive staining of smeg19K with Sudan Black 4B, a postelectrophoresis dye used to stain for lipids. Smeg19K has also stained positively for glycosylation with the lectin concavalin A, a highly specific stain for mannose residues. As expected, treatment with tunicamycin (an antibiotic known to inhibit N-glycosylation) and treatment with deglycosylation assay (non-specific for mannose), showed no reduction in size of 19 kDa glycolipoprotein.

Government work report reflects the most authoritative information about government work conditions at present and its future plans.With the guidance of Toury s translational norm theory,this thesis focuses on the analysis of the translation of 2015 government work report from three aspects,namely initial norms,preliminary norms and operational norms.Based on those relevant analysis,it concludes that Toury' s translational norm theory is valuable to summarize some common principles about the translation of government documents from macroscopic and microscopic views.

Adaptation is the pursuit of active, deliberate measures to enhance humankind’s capacity to manage water supply and attenuate demand in the face of climate uncertainty. This article contends that worsening constraints upon freshwater due to climate variability demand concerted, imaginative, science-based solutions. These solutions must join creative management to co-production of climate knowledge. Through a series of case studies, we analyze the need for adaptation approaches to prevail over climate variability, and the role of these factors to facilitate their implementation. We also examine how translation of climate knowledge is helping spur adaptation at various spatial levels. These experiences point to the challenges in adaptation, and the adversity various regions will be faced if we do not.

The overview shows that the scientific interest in social behaviour in mice has exponentially grown in the last two decades in parallel with advances in biotechnology and the emergence of genetically engineered mice. Most of the studies are psychopharmacological or look for the neurochemical bases of social behaviour and its alterations. However, the rol of social behaviour per se is increasing mainly in those research works aimed to model neuropsychiatric and neurode-generative diseases. In fact, at the translational level, the study of social behaviour in murine models is relevant because changes in social behaviour are present in most neuropsychiatric and neurodegenerative disorders as well as in other diseases that, directly or indirectly, affect the sphere of social relationships. The consideration of social behaviour in the experimental design of basic and translational research works using murine models may improve the predictive validity of new preventive and/or therapeutic strategies. The present work provides conceptual description of social behaviour in mice, the tests used to measure it and analyzes its increasing interest, mostly in the area of neuroscience. It reviews the 821 scientific studies (in English) included in the MEDLINE database from 1930 to December 2012. Keywords used for the search where those related to the different kinds of social behaviour (spontaneous or induced) in mice and took into account the diversity of experimental paradigms (dyads, groups, parental relationships, isolation) and the wide spectrum of behavioural tests available.

Anti-Inflammatory Pre-Treatment to Reduce Mobilization-Induced Liver Inflammation in Mice: Novel Model to Study Liver Injury

Background: Liver surgery requires mobilization of the liver resulting in injury. Mobilization is the predominant cause of hepatocyte damage during liver surgery and jeopardizes post-operative liver function. Previously, mobilization-induced liver injury was found to be associated with inflammation. So far, anti-inflammatory drugs to potentially prevent liver inflammation following liver mobilization were not tested. In this study, we aimed to establish an in vivo mouse model of mobilization-induced liver injury and to evaluate the effect of anti-inflammatory pre-treatment before liver mobilization on liver inflammation. Methods: To develop a mouse model for mobilization-induced liver injury, C57BL/6 mice (n = 8) underwent surgery during which the liver was mobilized by gentle manipulation of the lobes with cotton-wool applicators for 15 minutes. In two control groups, the liver was left alone or was subjected to laparotomy only. An additional group was added that received anti-TNF treatment (Infliximab) 2 days prior to surgery. Liver samples were obtained 2 hours after mobilization and liver inflammation was analyzed by histology and inflammatory gene expression. Results: Gentle liver mobilization resulted in acute liver inflammation as indicated by increased recruitment of inflammatory cells and elevated inflammatory gene expression compared to controls. Infliximab pre-treatment had no effect on the inflammatory response in the liver. Conclusion: Our current model provides an excellent opportunity to study the effects of pre-treatment with anti-inflammatory drugs on mobilization-induced liver inflammation. Further studies are needed to investigate whether anti-inflammatory mediators can be used to prevent liver inflammation.
