Dissecting the genetic regulation of gene expression is critical for understanding phenotypic variation and species evolution.However,our understanding of the transcriptional variability in sweet potato remains limited.Here,we analyzed two publicly available datasets to explore the landscape of transcriptomic variations and its genetic basis in the storage roots of sweet potato.The comprehensive analysis identified a total of 724,438 high-confidence single nucleotide polymorphisms(SNPs)and 26,026 expressed genes.Expression quantitative trait locus(eQTL)analysis revealed 4408 eQTLs regulating the expression of 3646 genes,including 2261 local eQTLs and 2147 distant eQTLs.Two distant eQTL hotspots were found with target genes significantly enriched in specific functional classifications.By combining the information from regulatory network analyses,eQTLs and association mapping,we found that IbMYB1-2 acts as a master regulator and is the major gene responsible for the activation of anthocyanin biosynthesis in the storage roots of sweet potato.Our study provides the first insight into the genetic architecture of genome-wide expression variation in sweet potato and can be used to investigate the potential effects of genetic variants on key agronomic traits in sweet potato.

sweet cherry(Prunus avium)is an economically significant fruit species in the genus Prunus.However,in contrast to other important fruit trees in this genus,only one draft genome assembly is available for sweet cherry,which was assembled using only Illumina short-read sequences.The incompleteness and low quality of the current sweet cherry draft genome limit its use in genetic and genomic studies.A high-quality chromosome-scale sweet cherry reference genome assembly is therefore needed.A total of 65.05 Gb of Oxford Nanopore long reads and 46.24 Gb of Illumina short reads were generated,representing~190x and 136x coverage,respectively,of the sweet cherry genome.The final de novo assembly resulted in a phased haplotype assembly of 344.29 Mb with a contig N50 of 3.25 Mb.Hi-C scaffolding of the genome resulted in eight pseudochromosomes containing 99.59%of the bases in the assembled genome.Genome annotation revealed that more than half of the genome(59.40%)was composed of repetitive sequences,and 40,338 protein-coding genes were predicted,75.40%of which were functionally annotated.With the chromosomescale assembly,we revealed that gene duplication events contributed to the expansion of gene families for salicylic acid/jasmonic acid carboxyl methyltransferase and ankyrin repeat-containing proteins in the genome of sweet cherry.Four auxin-responsive genes(two GH3s and two SAURs)were induced in the late stage of fruit development,indicating that auxin is crucial for the sweet cherry ripening process.In addition,772 resistance genes were identified and functionally predicted in the sweet cherry genome.The high-quality genome assembly of sweet cherry obtained in this study will provide valuable genomic resources for sweet cherry improvement and molecular breeding.

A novel aeration strategy in repeated-batch fermentation for efficient ethanol production from sweet sorghum juice

To improve the efficiency of ethanol production in a batch fermentation from sweet sorghum juice under a very high gravity(VHG)condition(~290 g/L of total sugar)by Saccharomyces cerevisiae NP01,repeatedbatch fermentation under an aerated condition(2.5 vvm for the first 4 h during every cycle)was done in a5-L fermenter.The average ethanol concentration(P),productivity(Qp)and yield(Yp/s)for five successive cycles were 112.31 g/L,1.55 g/L·h^-1 and 0.44,respectively with 80.97%sugar consumption.To complete sugar consumption,the total sugar of the juice was reduced to a high gravity(HG)level(~240 g/L).The results showed that yeast extract was not necessary for ethanol production,and aeration during every other cycle i.e.,alternating cycles,was sufficient to promote both yeast growth and ethanol production.The average P,Qpand Yp/svalues for eight successive cycles with aeration during alternating cycles were97.58 g/L,1.98 g/Láh and 0.41,respectively with 91.21%sugar consumption.The total fatty acids in the yeast cells under the aerated condition were^50%higher than without aeration,irrespective the initial sugar concentration,whereas the ergosterol contents under aeration condition were^29%to 49%higher than those without aeration.

The accumulation and productivity of shale gas are mainly controlled by the characteristics of shale reservoirs;study of these characteristics forms the basis for the shale gas exploitation of the Lower Cambrian Niutitang Formation(Fm),Southern China.In this study,core observation and lithology study were conducted along with X-ray diffraction(XRD)and electronic scanning microscopy(SEM)examinations and liquid nitrogen(N2)adsorption/desorption and CH4 isothermal adsorption experiments for several exploration wells in northwestern Hunan Province,China.The results show that one or two intervals with high-quality source rocks(TOC>2 wt%)were deposited in the deep-shelf facies.The source rocks,which were mainly composed of carbonaceous shales and siliceous shales,had high quartz contents(>40 wt%)and low clay mineral(50 nm in diameter).The shale reservoir has strong adsorption capacity for CH4.The Langmuir volume(VL)varied from 1.63 to 7.39 cm^3/g,with an average of 3.95 cm^3/g.The characteristics of shale reservoir are controlled by several factors:(1)A deep muddy continental shelf is the most favorable environment for the development of shale reservoirs,which is controlled by the development of basic materials.(2)The storage capacity of the shale reservoir is positively related to the TOC contents and plastic minerals and negatively related to cement minerals.(3)High maturity or overmaturity leads to the growth of organic pores and microfractures,thereby improving the reservoir storage capacity.It can be deduced that the high percentage of residual gas in Niutitang Fm results from the

sweets （sugars will eventually be exported transporters） are a novel class of recently identified sugar transporters that play important roles in diverse physiological processes. However, only a few species of the plant sweetgene family have been functionally identified. Up till now, there has been no systematic analysis of the sweetgene family in Cucurbitaceae crops. Here, a genome-wide characterization of this family was conducted in cucumber（Cucumis sativus L.）. A total of 17 Cssweetgenes were identified, which are not evenly distributed over the seven cucumber chromosomes. Cucumber sweet protein sequences possess seven conserved domains and two putative serine phosphorylation sites. The phylo- genetic tree of the sweet genes in cucumber, Arabidopsis thaliana, and Oryza sativa was constructed, and all the sweet genes were divided into four clades. In addition, a number of putative cis-elements were identified in the promoter regions of these Cssweet genes： nine types involved in phytohormone responses and eight types involved in stress responses. Moreover, the transcript levels of Cssweetgenes were analyzed in various tissues using quantitative real-time polymerase chain reaction. A majority （70.58%） of the Cssweet genes were confined to reproductive tissue development. Finally, 18 putative watermelon Clasweetgenes and 18 melon Cmsweetgenes were identified that showed a high degree of similarity with Cssweetgenes. The results from this study provided a basic understanding of the Cssweetgenes and may also facilitate future research to elucidate the function of sweet genes in cucumber and other Cucurbitaceae crops.

Genome-wide association study of vitamin E in sweet corn kernels

Vitamin E,consisting of tocopherols and tocotrienols,serves as a lipid-soluble antioxidant in sweet corn kernels,providing nutrients to both plants and humans.Though the key genes involved in the vitamin E biosynthesis pathway have been identified in plants,the genetic architecture of vitamin E content in sweet corn kernels remains largely unclear.In the present study,an association panel of 204 inbred lines of sweet corn was constructed.Seven compounds of vitamin E were quantified in sweet corn kernels at 28 days after pollination.A total of 119 loci for vitamin E were identified using a genome-wide association study based on genotyping by sequencing,and a genetic network of vitamin E was constructed.Candidate genes identified were involved mainly in RNA regulation and protein metabolism.The known gene ZmVTE4,encodingγ-tocopherol methyltransferase,was significantly associated with four traits(α-tocopherol,α-tocotrienol,theα/γ-tocopherol ratio,and theα/γ-tocotrienol ratio).The effects of two causative markers on ZmVTE4 were validated by haplotype analysis.Finally,two elite cultivars(Yuetian 9 and Yuetian 22)with a 4.5-fold increase in the sum ofα-andγ-tocopherols were developed by marker-assisted selection,demonstrating the successful biofortification of sweet corn.Three genes(DAHPS,ADT2,and cmu2)involved in chorismate and tyrosine synthesis were significantly associated with theα/γ-tocotrienol ratio.These results shed light on the genetic architecture of vitamin E and may accelerate the nutritional improvement of sweet corn.

Based on the sweet potato starch,cationic starch,acetic starch and cationic-acetic compoundedly modified starch were made through chemical denaturalization.The above three kinds of static rheological parameter and dynamic rheological parameter were measured,respectively.The experimental result reveals that the thermal stability of starchy viscosity increases after chemical denaturalization.Under the condition of identical shearing rate,the shear stress of cationic-acetic ester compoundedly modified sweet potato starch paste is the largest among these kinds of sweet potato starch.This attributes to a phenomenon of shearing thinning.Furthermore,raw sweet potato starch has a larger gel intensity than that of modified starch.

sweet sorghum mutants induced by^(12)C(6+)-ion irradiation were planted under different soil salinity conditions to investigate the mechanisms maintaining the transport and spatial distribution of Na^+. The functions of the synergistic responses of NHX, AKT1, and SOS1 related to Na^+ accumulation were investigated in control(KFJT-CK) sorghum and KF1210-3 and KF1210-4 mutants. The results indicated that the NHX, AKT1, and SOS1 proteins in sweet sorghum are mainly involved in the transport, exclusion, and spatial distribution of Na^+,respectively. In addition to physiological parameters, we also measured the expression levels of NHX, AKT1, and SOS1 genes. The experimental results indicated that 150 m M Na Cl induced marked increases in the transcripts of NHX and SOS1 after 8 and 12 h in the KF1210-3,KF1210-4, and KFJT-CK cultivars. In contrast, however, a decrease in AKT1 was observed. On the basis of our results, we propose a model in which cooperation amongNHX, AKT1, and SOS1 facilitates Na^+ homeostasis in sweet sorghum in response to an increase in salt concentration. Accordingly, study of the regulatory mechanisms in sweet sorghum generated by carbon ion irradiation is essential for the selection of salt-tolerant cultivars.

sweet(sugars will eventually be exported transporter)是一类著名的糖类运输体,在植物的生长发育、果实成熟、糖类运输和胁迫反应中发挥重要作用。本研究在枣(Ziziphus jujuba)中鉴定了27个sweet基因,并对它们的保守基序、基因结构、染色体定位、进化关系、启动子顺式作用元件等进行分析,对亚细胞定位进行预测。结果表明,Zjsweet基因家族成员不均匀地分布在11条染色体上,均具有1~2个MtN3保守结构域,含多个内含子。根据系统进化关系,27个Zjsweet成员可分为3个分支,同一分支具有相似的基因结构和基序。Zjsweet基因启动子区域包含激素、胁迫等多个逆境相关的顺式元件。转录组数据分析表明,基因Zjsweet9、Zjsweet22和Zjsweet23可能参与枣果实中蔗糖积累的调控,Zjsweet22、Zjsweet23和Zjsweet25可能参与对高温胁迫的调控。总之,Zjsweet在果实发育过程及高温胁迫中扮演着重要的角色,这些结果有助于更好地理解sweet基因,从而为进一步探索枣sweet基因功能提供参考。

Like olfaction,the sense of taste allows the detection and discrimination of chemicals in the environment.However,while olfaction is specialized in the detection of volatile chemicals,taste is restricted to the detection of contact-chemicals.Two families of mammalian taste receptors,T1R and T2R,involved in recognition of sweet,umami(the taste of monosodium glutamate)and bitter stimuli have been identified and characterized.Although much progress has been made in studies on the basic mechanisms of taste recognition and signal transduction in mammals,we are still far from a full understanding of different taste qualities.This review presents a current perspective on sweet,bitter and umami taste receptors and their signal transduction mechanism.We also discuss the evolution of taste and taste-related molecules.
