Arg-X Protease-Sensitive in supramolecular structures of Interphase Cell Nucleus during Growth Morphogenesis Mature Germs of Wheat

Abstract： In the present study was investigated Arg-X protease-sensitive in supramolecular-genome compartments （nucleoplasm, chromatin, nuclear matrix）, during the period of the transcriptional activation of chromatin when the growth processes was initiated in the mature germs of winter and transformed from it spring wheat. The germs have been separated from endosperm from 0 h （air-dry seed） up to 21 h in each 3 h after the start of seeds soaking. Cell nucleus have been allocated from germs and cleared, and then from them supramolecular-genome compartments were extracted by increasing ionic strength of solution. The Arg-X （tryptase） activity was assessed by cleavage of Arg-X bonds in the arginine-enriched protein protamine in all nuclear fractions. In the present study have shown what Arg-X protease-sensitives zones can be located on the supramolecular structures of chromatin matrix in processes of realization of ontogenetic programs of development in mature germs of the winter and transformed from it spring wheat. Arg-X protease-sensitive can translocate and coordinated in heteropolymer structures on the same genetic matrix. Questions of epigenetic mechanisms are discussed.

Main observation and conclusion Polymer-supramolecular double-network hydrogels(PS-DN hydrogels)often show much improved recovery rates than conventional double-network hydrogels because of the fast self-assembling properties,making them attractive candidates for tissue engineering and flexible ***,as the supramolecular network is dynamic and susceptible to break under low strains,the overall mechanical properties of PS-DN hydrogels are still ***,we report the mechanical properties for PS-DN hydrogels can be significantly improved by tuning the supramolecular network structures.A single amino acid change of the self-assembling peptide can tune the assembled structures from nanofiber to *** a microscopic structural change can greatly increase the Young's modulus(107.4 kPa),fracture stress(0.48 MPa),and toughness(0.38 MJ·m^(–3))of the PS-DN ***,the structural change also leads to slightly faster recovery rates(supramolecular network structures.

The dual ligand system of V-shaped 5-（4-carboxy-2-nitrophenoxy）-isophthalic acid（Hcn-H2ipa） and dipyridy-type molecules produces two new coordination polymers： {[Ni（cn-Hipa）（bpa）（H2O）3]·1.5H2O}n 1 and {[Cu（Hcn-ipa）（ppe）2]}n 2（bpa = 1,2-bis（4-pyridyl）ethane, and ppe = trans-1-（2-pyridyl）-2-（4-pyridyl）ethylene）. Two complexes have been characterized by single-crystal X-ray diffraction, elemental analysis, IR, and thermogravimetric analysis（TGA）. Complex 1 is of monoclinic system, space group C2 with a = 25.936（3）, b = 7.6100（10）, c = 16.558（2）, β = 105.9470（10）°, V = 3142.4（7） A3, Dc = 1.415 g/cm3, Mr = 669.23, Z = 4, F（000） = 1388, μ = 0.687 mm-1, the final R = 0.0397 and w R = 0.0967 for 12070 observed reflections with I 〉 2σ（I）. Complex 2 belongs to the triclinic system, space group P1 with a = 10.165（5）, b = 14.946（7）, c = 15.506（11） A, α = 106.461（7）, β = 102.454（8）, γ = 107.861（5）°, V = 2029（2） A3, Dc = 1.266 g/cm3, Mr = 773.20, Z = 2, F（000） = 794, μ = 0.595 mm-1, the final R = 0.0538 and w R = 0.1378 for 15488 observed reflections with I 〉 2σ（I）. Two complexes show that the 1D polymeric chains are cohered together by extensive H-bonds to complete stable supramolecular microporous frameworks.

Luminescent spin crossover(SCO) materials have attracted significant interest owing to their potential applications in magneto-optical switches. However, the majority of previously reported FeII-based SCO complexes are adversely affected by fluorescence quenching in the solid-state. Here, we have constructed the first mononuclear FeIIcomplex decorated with an aggregation-induced emission(AIE) luminophore(i.e., tetraphenylethylene) that exhibits synergistic SCO and fluorescence behavior. Intriguingly, we obtained two types of crystals in different solvent systems, both displaying distinct magnetic bistability and fluorescence properties. The fluorescence intensity was observed to track the magnetic susceptibility, which confirmed that SCO and solid-state fluorescence operate synergistically. We introduce a novel approach for the construction of luminescent SCO compounds using an AIEgen as a luminophore, which leads to fluorescence emission in the solid-state, thus allowing us to study the synergy between SCO and fluorescence.

The purpose of this study was to analyze spatio-temporal dynamics of localization of protease-sensitive sites Arg-X in non-histone and histone blocks of heteropolymer suprastructures （nucleoplasm, chromatin, nuclear matrix） as possible zones affecting the conformational rearrangements of the total interphase chromatin at the induction of increasing morphogenesis of mature embryos-germs of spring and transformed from its winter wheat. Germinated embryos-germs were detached from endosperm after 24 hours from the start of soaking. Cell nuclei have been allocated from embryos-germs and cleared, and then from their heteropolymer suprastructures （nucleoplasm, chromatin loosely bound with nuclear matrix and chromatin tightly bound with nuclear matrix, and nuclear matrix） were extracted by increasing ionic strength of solution. From isolated nuclear suprastructures, non-histone proteins were separated from histones using ion exchange chromatography. Trypsin-like complexes from non-histone proteins and histone blocks were isolated using the affinity chromatography. The Arg-X （tryptase） activity was assessed by cleavage of Arg-X bonds in the arginine-enriched protein protamine. Hypersensitivity to the Arg-X proteolysis in trypsin-like complexes detected at the level suprastructures of chromatin tightly bound with the nuclear matrix was shown. The most active changes of the nuclear proteome have occurred at the level of the non-histone proteins and the core histones （H2A ＋ H2B）（H3 ＋ H4） of induced to growth embryos-seedlings of winter wheat （compared to the initial spring form of wheat）. Perhaps hypersensitivity to the Arg-X activity of the trypsin-like complexes in the non-histone proteins and the core blocks of chromatin tightly bound with nuclear matrix have been entrenched during the transforming of the winter wheat from the initial spring wheat.

The purpose of the given work was the experimental analysis of features of Arg-X proteolysis in proteom of supramolecular structures of bacterial cells during their life cycle. The basic attention was devoted to relaxation of Arg-X sites of proteom in association with the evolutionary significance ofArg-rich histones in the eukaryotic kingdom. These properties were not studied in the prokaryotes. Cells ofE. coli were grown to the stationary phase, collected by centrifugation and washed. All cells were taken over from 50 min to 430 min at intervals of 20 min and were preserved in glycerol. The supramolecular structures were fractionated from bacterial cells by increasing ionic strength of solution. The Arg-Xactivity was assessed by cleavage of Arg-Xbonds in the arginine-enriched protein protamine in all cell fractions. We have shown that during the stationary phase in the life cycle of E. coli, there are a high continuous activity of the Arg-X processing at the level of＂cytoskeleton＂ of the cell and bright cyclic activity in the cytoplasm.

supramolecular Nanostructures from Self-Assembly of Various-Shaped Rod Building Block Oligomers

H-shaped, n-shaped, bent-shaped and linear rod-coil molecules, consisting of biphenyls and a p-terphenyl or phenyl unit linked together with ether bonds as a rod segment and poly(ethylene oxide)(PEO) with various degree of polymerization as coil segments were synthesized. These molecules contain lateral alkyl groups in the middle of benzene ring and lateral methyl groups between the rod and coil segments. The self-assembling behavior of these molecules was investigated by means of DSC, POM, AFM, TEM and SAXS in the bulk and aqueous solutions. Molecules with non lateral units self-assemble into a lamellar and a hexagonal perforated layer structure in the crystalline phase and the liquid crystalline phase, respectively. While, molecules incorporating lateral methyl groups between the rod and coil segments spontaneously self organize into hexagonal perforated layer, oblique columnar or 3-D body centered tetragonal structures. In aqueous solutions, molecules with non lateral units self-assemble into nanoribbon or fibrous aggregates, whereas, molecules with a lateral groups exhibit self-organizing capacity to form helical fibers. Notably, CD experiments together with AFM investigations of these molecules reveal that the helical sense switches to opposite-handedness, depending on the temperature of aqueous solution. These results reveal that self-assembling behavior of these molecules is dramatically influenced by lateral alkyl groups in the middle of rod segment and lateral methyl groups between the rod and coil segments.
