Over the past two decades, several fluorescence-and non-fluorescence-based optical microscopes have been developed to break the diffraction limited barrier. In this review, the basic principles implemented in microscopy for super-resolution are described. Furthermore, achievements and instrumentation for super-resolution are presented. In addition to imaging, other applications that use super-resolution optical microscopes are discussed.

Although small EVs(sEVs)have been used widely as biomarkers in disease diagnosis,their heterogeneity at single EV level has rarely been *** is because high-resolution characterization of sEV presents a major challenge,as their sizes are below the optical diffraction ***,we report that upconversion nanoparticles(UCNPs)can be used for super-resolution profiling the molecular heterogeneity of *** show that Er3+-doped UCNPs has better brightness and Tm3+-doped UCNPs resulting in better resolution beyond diffraction *** an orthogonal experimental design,the specific targeting of UCNPs to the tumour epitope on single EV has been cross validated,resulting in the Pearson’s R-value of 0.83 for large EVs and~65%co-localization double-positive spots for ***,super-resolution nanoscopy can distinguish adjacent UCNPs on single sEV with a resolution of as high as 41.9 *** decreasing the size of UCNPs from 40 to 27 nm and 18 nm,we observed that the maximum UCNPs number on single sEV increased from 3 to 9 and 21,*** work suggests the great potentials of UCNPs approach“digitally”quantify the surface antigens on single EVs,therefore providing a solution to monitor the EV heterogeneity changes along with the tumour progression progress.

As a new method of cell-cell communication,tunneling nanotubes(TNTs)play important roles in cell-cell signaling and mass ***,a lack of powerful tools to visualize dynamic TNTs with high temporal/spatial resolution restricts the exploration of their formation and cleavage,hindering the complete understanding of its ***,we present the first example of using stochastic optical reconstruction microscopy(STORM)to observe the tube-like structures of TNTs linking live cells with an easily prepared fluorescent *** of this new imaging microscopy,the cleavage process of TNTs was observed with a high spatial resolution.

Recently, neighbor embedding based face super-resolution(SR) methods have shown the ability for achieving high-quality face images, those methods are based on the assumption that the same neighborhoods are preserved in both low-resolution(LR) training set and high-resolution(HR) training set. However, due to the "one-to-many" mapping between the LR image and HR ones in practice, the neighborhood relationship of the LR patch in LR space is quite different with that of the HR counterpart, that is to say the neighborhood relationship obtained is not true. In this paper, we explore a novel and effective re-identified K-nearest neighbor(RIKNN) method to search neighbors of LR patch. Compared with other methods, our method uses the geometrical information of LR manifold and HR manifold simultaneously. In particular, it searches K-NN of LR patch in the LR space and refines the searching results by re-identifying in the HR space, thus giving rise to accurate K-NN and improved performance. A statistical analysis of the influence of the training set size and nearest neighbor number is given, experimental results on some public face databases show the superiority of our proposed scheme over state-of-the-art face hallucination approaches in terms of subjective and objective results as well as computational complexity.

Fluorescence polarization microscopy(FPM)aims to detect the dipole orientation of fluorophores and to resolve structural information for labeled organelles via wide-field or confocal *** FPM often suffers from the presence of a large number of molecules within the diffraction-limited volume,with averaged fluorescence polarization collected from a group of dipoles with different ***,we apply sparse deconvolution and least-squares estimation to fluorescence polarization modulation data and demonstrate a super-resolution dipole orientation mapping(SDOM)method that resolves the effective dipole orientation from a much smaller number of fluorescent molecules within a sub-diffraction focal *** further apply this method to resolve structural details in both fixed and live *** the first time,we show that different borders of a dendritic spine neck exhibit a heterogeneous distribution of dipole ***,we illustrate that the dipole is always perpendicular to the direction of actin filaments in mammalian kidney cells and radially distributed in the hourglass structure of the septin protein under specific *** accuracy of the dipole orientation can be further mapped using the orientation uniform factor,which shows the superiority of SDOM compared with its wide-field counterpart as the number of molecules is decreased within the smaller focal *** the inherent feature of the orientation dipole,the SDOM technique,with its fast imaging speed(at sub-second scale),can be applied to a broad range of fluorescently labeled biological systems to simultaneously resolve the valuable dipole orientation information with super-resolution imaging.

In this Letter,we report on the investigations of nonlinear scattering of plasmonic nanoparticles by manipulating ambient *** create different local thermal hosts for gold nanospheres that are immersed in oil,encapsulated in silica glass and also coated with silica *** terms of regulable effective thermal conductivity,silica coatings are found to contribute significantly to scattering *** from the enhanced thermal stability and the reduced plasmonic coupling provided by the shell-isolated nanoparticles,we achieve super-resolution imaging with a feature size of 52 nm(λ/10),and we can readily resolve pairs of nanoparticles with a gap-to-gap distance of 5 nm.

Image super-resolution is essential for a variety of applications such as medical imaging,surveillance imaging,and satellite imaging,among ***,the most popular color image super-resolution is performed in each color channel *** this paper,we show that the super-resolution quality can be further enhanced by exploiting the cross-channel *** by the High-Quality Linear Interpolation(HQLI)demosaicking algorithm by Malvar et al.,we design an image super-resolution scheme that integrates intra-channel interpolation with cross-channel details by isotropic linear *** its simplicity,our super-resolution method achieves the accuracy comparable with the existing fastest state-of-the-art super-resolution algorithm at 20 times faster *** is well applicable to applications that adopt traditional interpolations,for improved visual quality at trivial computation *** comparative study verifies the effectiveness and efficiency of the proposed super-resolution algorithm.
