Identification of Plasma Membrane Aquaporin in Guard Cells of Vicia faba and Its Role in stomatal movement

Water channels or aquaporins are the main pathways of water transport. Both the existence and function of aquaporins in die guard cells of Vicia faba L. were investigated both by using RD28 cDNA and RD28 antibody as probes, and by controlling stomatal movement as a parameter combined with antibody and inhibitor of aquaporins respectively. The results revealed that RD28 mRNA, encoding a plasma membrane aquaporin, expressed in ale mesophyll cells and vascular tissues of V. faba, especially in guard cells. And the location of RD28-like proteins was mainly on plasma membrane of guard cells. The addition of 25 mumol/L HgCl2, an aquaporin blocker, and antibody of RD28 as well, greatly suppressed the stomatal opening or guardcell protoplast swelling induced by fusicoccin and light, and closing induced by abscisic acid. However, 5 mmol/L, beta-mercaptoethanol, a reverse reagent of aquaporin blocker, reversed the inhibitory effect of HgCl2 Pretreatment oil stomatal opening ( i.e., HgCl2 was removed after HgCl2 pretreatment for 10 min). The results suggest that the aquaporins in V. faba are associated with stomatal movement.

As a possible peptide primary messenger, extracellular calmodulin (CaM) may regulate processes such as cell proliferation, pollen germination and expression of some genes. Stomata open or close quickly in response to environmental stimuli. CaM was found to be extracellular both in guard cells of broad bean leaves and in epidermal cells by immuno-electron microscopy and immuno-fluorescence microscopy techniques. Exogenous purified CaM enhanced stomatal closure and inhibited stomatal opening with an optimal concentration of 10(-8) mol/L; CaM antagonist W7-agarose and anti-CaM serum, which were membrane-impermeable macromolecules, inhibited stomatal closure and promoted stomatal opening. All these data showed that endogenous extracellular CaM. of guard cells did promote stomatal closure and inhibit stomatal opening, and could he active only outside the cells. Therefore under natural conditions, the endogenous extracellular CaM of guard cells might regulate stomatal movements as a primary messenger together with other signal molecules, and might be an important linkage between environmental stimuli and cell responses.

Hyperosmotic stress caused by drought is a detrimental threat to plant growth and agricultural productivity due to limited water *** are gateways of transpiration and gas exchange,the swift adjustment of stomatal aperture has a strong influence on plant drought *** intensive investigations of stomatal closure during drought stress in past decades,little is known about how sequential signals are integrated during complete ***,we discovered that the rapid Ca^(2+) signaling and subsequent abscisic acid(ABA)signaling contribute to the kinetics of both F-actin reorganizations and stomatal closure in Arabidopsis thaliana,while stomatal CLOSURE-RELATED ACTIN BINDING PROTEIN1(SCAB1)is the molecular switch for this entire *** the early stage of osmotic shock responses,swift elevated calcium signaling promotes SCAB1 phosphorylation through calcium sensors CALCIUM DEPENDENT PROTEIN KINASE3(CPK3)and *** phosphorylation restrained the microfilament binding affinity of SCAB1,which bring about the Factin disassembly and stomatal closure *** the osmotic stress signal continued,both the kinase activity of CPK3 and the phosphorylation level of SCAB1 attenuated *** further found that ABA signaling is indispensable for these attenuations,which presumably contributed to the actin filament reassembly process as well as completion of stomatal ***,the dynamic changes of SCAB1 phosphorylation status are crucial for the kinetics of stomatal *** together,our results support a model in which SCAB1 works as a molecular switch,and directs the microfilament rearrangement through integrating the sequentially generated Ca^(2+) and ABA signals during osmotic stress induced stomatal closure.

In recent years, adenosine tri-phosphate （ATP） has been reported to exist in apoplasts of plant cells as a signal molecule. Extracellular ATP （eATP） plays important roles in plant growth, development, and stress tolerance. Here, extra- cellular ATP was found to promote stomatal opening of Arabidopsis thaliana in light and darkness. ADP, GTP, and weakly hydrolyzable ATP analogs （ATPγS, Bz-ATP, and 2meATP） showed similar effects, whereas AMP and adenosine did not affect stomatal movement. Apyrase inhibited stomatal opening. ATP-promoted stomatal opening was blocked by an NADPH oxidase inhibitor （diphenylene iodonium） or deoxidizer （dithiothreitol）, and was impaired in null mutant of NADPH ox- idase （atrbohD/F）. Added ATP triggered ROS generation in guard cells via NADPH oxidase. ATP also induced Ca^2＋ influx and H ＋ efflux in guard cells. In atrbohD/F, ATP-induced ion flux was strongly suppressed. In null mutants of the heterotrimeric G protein α subunit, ATP-promoted stomatal opening, cytoplasmic ROS generation, Ca^2＋ influx, and ^H＋ efflux were all sup- pressed. These results indicated that eATP-promoted stomatal opening possibly involves the heterotrimeric G protein, ROS, cytosolic Ca^2＋, and plasma membrane H＋-ATPase.

: The regulation of stomatal movement is one of the most important signaling networks in plants. The H+-ATPase at the plasma membrane of guard cells plays a critical role in the stomata opening, while there are some conflicting results regarding the effectiveness of the plasma membrane H+-ATPase inhibitor, vanadate, in inhibiting stomata opening. We observed that 2 mmol/L vanadate hardly inhibited light-stimulated stomata opening in epidermal peels of Vicia faba L., but significantly inhibited dark- and ABA-induced stomatal closure. These results cannot be explained with the previous findings that H+-ATPase was inhibited by vanadate. In view of the fact that vanadate is an inhibitor of protein tyrosine phosphatases (PTPases), we investigated whether the stomatal movement regulated by vanadate is through the regulation of PTPase. As expected, phenylarsine oxide (PAO), a specific inhibitor of PTPase, has very similar effects and even more effective than vanadate. Typical PTPase activity was found in guard cells of V. faba; moreover, the phosphatase activity could be inhibited by both vanadate and PAO. These results not only provide a novel explanation for conflicting results about vanadate modulating stomatal movement, but also provide further evidence for the involvement of PTPases in modulating signal transduction of stomatal movement.

We know ambient CO is continuing rising and this rising CO causes plants stomatal pores closing and thus regulates plant gas exchange---CO influx into plants and water loss by ***,little is known about the early CO sensing mechanisms that control plant gas ***,we found that CO-binding proteins,βCA1 andβCA4, are required for[CO]regulation of stomatal movements and also stomatal development, functioning as the earliest components in guard cell signaling ***, CA-mediated stomatal CO responses are not directly linked to photosynthesis and originate in guard *** cell over-expression of either CA1 or CA4 in wild-type plants greatly enhances water use *** physiological research showed that CA-mediated catalysis is a major mechanism forβCA-mediated CCvinduced stomatal closing and elevated HCO,more than CO and protons,functions as a small-molecule transducer to activate anion channels in guard *** this pathway,Cais required for HCO activation of S-type anion channel and the activation is mediated by SLAC1. Furthermore,several proteins were identified as interactors of CA1 and CA4,indicating there is another mechanism for CA1 and CA4 in CO regulation of stomatal movement.
