Flumorph is a carboxylic acid amide（CAA）fungicide expressing high activity against *** sensitivity of 120 Phytophthora sojae isolates to flumorph was *** baseline sensitivities were distributed as a unimodal curve with a mean EC50 of 0.54μg/ml and no spontaneous resistance was *** types of stable resistant mutants,exhibiting different resistance levels to CAAs,were obtained by flumorph *** I with a low resistance level was screened out from a wild-type isolate by exposure to a low concentration of *** II,Type III,Type IV and Type V,with a higher resistance level to CAAs,were obtained with a higher frequency by exposing Type I to a higher concentration of flumorph.A point mutation I1027V was found in the cellulose synthase 3（CesA3）in Type *** from I1027V,V1025L,G1020A,G1020S and Q992H were discovered respectively in Type II,Type III,Type IV and Type V isolates as ***-direct mutagenesis of the above mutations via CRISPR/Cas9 technology confirmed that I1027V indeed confer the low resistance to CAAs and with the assistance of V1025L,G1020A/S and Q992H,the resistance ability of *** with I1027V to CAAs were proved significantly *** consideration of the resistance development process from sensitive to low resistance,then to high resistance,it is deduced that the genotype of CesA3 gene in *** will probably evolve stepwise from the wild type to Type I,then to Type II,Type III,Type IV or Type V under the selection of flumorph.
