Interaction of Genotype,Environment,and Management on Organ-Specific Critical Nitrogen Dilution Curve in Wheat

The organ-specific critical nitrogen(N_(c))dilution curves are widely thought to represent a new approach for crop nitrogen(N)nutrition diagnosis,N management,and crop modeling.The N_(c) dilution curve can be described by a power function(N_(c)=A_(1)·W^(−A2)),while parameters A_(1) and A_(2) control the starting point and slope.This study aimed to investigate the uncertainty and drivers of organ-specific curves under different conditions.By using hierarchical Bayesian theory,parameters A_(1) and A_(2) of the organ-specific N_(c) dilution curves for wheat were derived and evaluated under 14 different genotype×environment×management(G×E×M)N fertilizer experiments.Our results show that parameters A_(1) and A_(2) are highly correlated.Although the variation of parameter A_(1) was less than that of A_(2),the values of both parameters can change significantly in response to G×E×M.Nitrogen nutrition index(NNI)calculated using organ-specific N_(c) is in general consistent with NNI estimated with overall shoot N_(c),indicating that a simple organ-specific N_(c) dilution curve may be used for wheat N diagnosis to assist N management.However,the significant differences in organ-specific N_(c) dilution curves across G×E×M conditions imply potential errors in N_(c) and crop N demand estimated using a general N_(c) dilution curve in crop models,highlighting a clear need for improvement in N_(c) calculations in such models.Our results provide new insights into how to improve modeling of crop nitrogen–biomass relations and N management practices under G×E×M.

China’s Actions to Achieve Universal Health Coverage for Children

The United Nations General Assembly adopted the 2030 Sustainable Development Goals(SDGs)in September 2015.Through this,governments have also committed to universal health coverage(UHC)as a development sub-goal within the SDG health field(1-2).In its effort to fully safeguard universal child health,China has established a management system for child healthcare to provide all children with continuous healthcare services starting at birth.Since 1950,China has implemented a system of local physicians who take responsibility for child healthcare services within their areas.Under the three-tier management system at the county,township,and village levels,every newborn is included in the child health management system starting immediately after birth.The urban and rural community-based health facilities in the area provide unified,standardized,and systematic healthcare services.Moreover,since 2009,child health management has been included in the National Essential Public Health Service Program,which mandates that community-based primary health facilities provide free health management services for children 0–6 years old in their area.This has led to equitable and comprehensive coverage of health services for these children.When children enter school at the age of six,they receive an annual routine physical examination at school.In 2020,the national coverage for neonatal visits,the health management rate for children under 3 and the health management rate for children under 7 all surpassed 92%,while the prevalence of stunting among children under 5 continued to decline.In addition,urban-rural disparities in major child health indicators have also narrowed significantly.Moreover,the inoculation rate of children in National Immunization Programme Vaccines has remained over 99%since 2011(3).

Comparing COVID-19 Case Prediction Between ARIMA Model and Compartment Model—China,December 2019–April 2020

Introduction:To compare the performance between the compartment model and the autoregressive integrated moving average(ARIMA)model that were applied to the prediction of new infections during the coronavirus disease 2019(COVID-19)epidemic.Methods:The compartment model and the ARIMA model were established based on the daily cases of new infection reported in China from December 2,2019 to April 8,2020.The goodness of fit of the two models was compared using the coefficient of determination(R2).Results:The compartment model predicts that the number of new cases without a cordon sanitaire,i.e.,a restriction of mobility to prevent spread of disease,will increase exponentially over 10 days starting from January 23,2020,while the ARIMA model shows a linear increase.The calculated R2 of the two models without cordon sanitaire were 0.990 and 0.981.The prediction results of the ARIMA model after February 2,2020 have a large deviation.The R2 of complete transmission process fit of the epidemic for the 2 models were 0.964 and 0.933,respectively.Discussion:The two models fit well at different stages of the epidemic.The predictions of compartment model were more in line with highly contagious transmission characteristics of COVID-19.The accuracy of recent historical data had a large impact on the predictions of the ARIMA model as compared to those of the compartment model.&Joint first authors.

Dysfunctions of ATP-binding cassette,subfamily D,member 1(ABCD1)cause X-linked adrenoleukodystrophy,a rare neurodegenerative disease that affects all human tissues.Residing in the peroxisome membrane,ABCD1 plays a role in the translocation of very long-chain fatty acids for theirβ-oxidation.Here,the six cryo-electron microscopy structures of ABCD1 in four distinct conformational states were presented.In the transporter dimer,two transmembrane domains form the substrate translocation pathway,and two nucleotide-binding domains form the ATP-binding site that binds and hydrolyzes ATP.The ABCD1 structures provide a starting point for elucidating the substrate recognition and translocation mechanism of ABCD1.Each of the four inward-facing structures of ABCD1 has a vestibule that opens to the cytosol with variable sizes.Hexacosanoic acid(C26:0)-CoA substrate binds to the transmembrane domains(TMDs)and stimulates the ATPase activity of the nucleotide-binding domains(NBDs).W339 from the transmembrane helix 5(TM5)is essential for binding substrate and stimulating ATP hydrolysis by substrate.ABCD1 has a unique C-terminal coiled-coil domain that negatively modulates the ATPase activity of the NBDs.Furthermore,the structure of ABCD1 in the outward-facing state indicates that ATP molecules pull the two NBDs together and open the TMDs to the peroxisomal lumen for substrate release.The five structures provide a view of the substrate transport cycle and mechanistic implication for disease-causing mutations.

De novo serine synthesis regulates chondrocyte proliferation during bone development and repair

The majority of the mammalian skeleton is formed through endochondral ossification starting from a cartilaginous template.Cartilage cells, or chondrocytes, survive, proliferate and synthesize extracellular matrix in an avascular environment, but the metabolic requirements for these anabolic processes are not fully understood. Here, using metabolomics analysis and genetic in vivo models, we show that maintaining intracellular serine homeostasis is essential for chondrocyte function. De novo serine synthesis through phosphoglycerate dehydrogenase(PHGDH)-mediated glucose metabolism generates nucleotides that are necessary for chondrocyte proliferation and long bone growth. On the other hand, dietary serine is less crucial during endochondral bone formation, as serine-starved chondrocytes compensate by inducing PHGDH-mediated serine synthesis.Mechanistically, this metabolic flexibility requires ATF4, a transcriptional regulator of amino acid metabolism and stress responses.We demonstrate that both serine deprivation and PHGDH inactivation enhance ATF4 signaling to stimulate de novo serine synthesis and serine uptake, respectively, and thereby prevent intracellular serine depletion and chondrocyte dysfunction. A similar metabolic adaptability between serine uptake and de novo synthesis is observed in the cartilage callus during fracture repair.Together, the results of this study reveal a critical role for PHGDH-dependent serine synthesis in maintaining intracellular serine levels under physiological and serine-limited conditions, as adequate serine levels are necessary to support chondrocyte proliferation during endochondral ossification.

Trend Analysis and Intervention Effect starting Point Detection of COVID-19 Epidemics Using Recalibrated Time Series Models—Worldwide,2020

Objective:This study aimed to identify a model for short-term coronavirus disease 2019(COVID-19)trend prediction and intervention evaluation.Methods:We compared the autoregressive integrated moving average(ARIMA)model and Holt exponential smoothing(Holt)model on predicting the number of cumulative COVID-19 cases in China.Based on the mean absolute percentage error(MAPE)value,the optimal model was selected and further tested using data from the United States,Italy and Republic of Korea.The intervention effect starting time points and abnormal trend changes were detected by observing the pattern of differences between the predicted and real trends.Results:The recalibrated ARIMA model with a 5-day prediction time span has the best model performance with MAPEs ranged between 2%and 5%.The intervention effects started to show on February 7 in the mainland of China,March 5 in Republic of Korea and April 27 in Italy,but have not been detected in the US as of May 19.Temporary abnormal trends were detected in Korea and Italy,but the overall epidemic trends were stable since the effect starting points.Conclusion:The recalibrated ARIMA model can detect the intervention effects starting points and abnormal trend changes;thus to provide valuable information support for epidemic trend analysis and intervention evaluation.

Architected materials can achieve enhanced properties compared to their plain counterparts.Specific architecting serves as a powerful design lever to achieve targeted behavior without changing the base material.Thus,the connection between architected structure and resultant properties remains an open field of great interest to many fields,from aerospace to civil to automotive applications.Here,we focus on properties related to mechanical compression,and design hierarchical honeycomb structures to meet specific values of stiffness and compressive stress.To do so,we employ a combination of techniques in a singular workflow,starting with molecular dynamics simulation of the forward design problem,augmenting with data-driven artificial intelligence models to address the inverse design problem,and verifying the behavior of de novo structures with experimentation of additively manufactured samples.We thereby demonstrate an approach for architected design that is generalizable to multiple material properties and agnostic to the identity of the base material.
