＜正＞spinal motor neuron shares the same neurotransmitter（ACh） with sympathetic preganglionic neuron.Can spinal motoneurons establish synapses with sympathetic postganglionic neurons? In this study,superior cervical ganglion neuron（SCGN） were labeled by Dil,and SMN-SCGN synaptic model were established by co-culture.Co-culture was double-immunofluorescently labeled by synatophysin（p38） antibody and ChAT antibody to illustrate the synaptogenesis in the presynaptic neurons,followed by quantitative study.Western blot and RT-PCR were also performed to confirm the changes of p38 content in the co-culture.Miniature spontaneous postsynaptic current（mSPC） was recorded in SCGN so as to observe the influence of co-culture and verify the formation of functional synapses.We found:（1） Primary culture of SMN and SCGN achieved satisfactory growth states.For SMN culture,neurons were got aging on the 14th day,In contrast,SCGN still kept excellent growth state on the 16th day.Double-fluorescent staining of NF200,ChAT or GFAP and Hoechst for nucli revealed that neuronal purity was up to 87.8%,with the purity of SMN being 70.9%and purity of SCGN being 95%.（2） In SCGN culture group,neurons could be labelled by Dil on day 1,and the red fluororescence could last many days without obvious negative influence on the survival of SCGN.（3） SMN and SCGN could be co-cultured on the cover-slip that have been microlandized;quantitative studies showed that co-culture groups significantly outnumbered either single culture group in the number of synapse.（4） Results of western blot indicated that the content of p38 was greatly increased in co-culture group compared with SMN culture group.（5） RT-PCR experiment showed that the level of p38 mRNA was significantly higher in co-culture group than either single culture group or the summation of the two groups.（6） On day 4 in vitro,mSPC was detected in SCGN in the co-culture group,while no similar postsynaptic current was detected in mere SCGN culture group.In conclusion,all the results revealed that SMN-SCGN

spinal cord injury （SCI） results in permanent loss of function leading to often devastating personal, economic and social problems. A contributing factor to the permanence of SCI is that damaged axons do not regenerate, which prevents the re-establishment of axonal circuits involved in function. Many groups are working to develop treatments that address the lack of axon regeneration after SCI. The emergence of biomaterials for regeneration and increased collaboration between engineers, basic and translational scientists, and clinicians hold promise for the development of effective therapies for SCI. A plethora of biomaterials is available and has been tested in various models of SCI. Considering the clinical relevance of contusion injuries, we primarily focus on polymers that meet the specific criteria for addressing this type of injury. Biomaterials may provide structural support and/or serve as a delivery vehicle for factors to arrest growth inhibition and promote axonal growth. Designing materials to address the specific needs of the damaged central nervous system is crucial and possible with current technology. Here, we review the most prominent materials, their optimal characteristics, and their potential roles in repairing and regenerating damaged axons following SCI.
