MGMT is down-regulated independently of promoter DNA methylation in rats with all-trans retinoic acidinduced spina bifida aperta

O6-methylguanine DNA methyltransferase(MGMT), a DNA repair enzyme, has been reported in some congenital malformations, but it is less frequently reported in neural tube defects. This study investigated MGMT mRNA expression and methylation levels in the early embryo and in different embryonic stages, as well as the relationship between MGMT and neural tube defects. spina bifida aperta was induced in rats by a single intragastric administration of all-trans retinoic acid on embryonic day(E) 10, whereas normal control rats received the same amount of olive oil on the same embryonic day. DNA damage was assessed by detecting γ-H2 A.X in spina bifida aperta rats. Real time-polymerase chain reaction was used to examine mRNA expression of MGMT in normal control and spina bifida aperta rats. In normal controls, the MGMT mRNA expression decreased with increasing embryonic days, and was remarkably reduced from E11 to E14, reaching a minimum at E18. In the spina bifida aperta model, γ-H2 A.X protein expression was increased, and mRNA expression of MGMT was markedly decreased on E14, E16, and E18. Bisulfite sequencing polymerase chain reaction for MGMT promoter methylation demonstrated that almost all CpG sites in the MGMT promoter remained unmethylated in both spina bifida aperta rats and normal controls, and there was no significant difference in methylation level between the two groups on either E14 or E18. Our results show that DNA damage occurs in spina bifida aperta rats. The mRNA expression of MGMT is downregulated, and this downregulation is independent of promoter DNA methylation.

The history: Myelomeningocele - also known as spina bifida- is a devastating congenital anomaly of the central nervoussystem that is caused by the malformation of the spinal cordand vertebral column during embryogenesis. Depending onthe location of the spina bifida lesion on the spine, patientssuffer from neurological dysfunction ranging from paresisand incontinence to complete paralysis. The current standardof care for spina bifida is in utero surgical repair of the defect,which has been shown to minimize the secondary deficits associatedwith this disorder (Adzick et al., 2011). Despite thesesuccesses, this approach does not reliably improve neurologicfunction of affected children. Several groups, including ourown, have performed studies aimed at augmenting the inutero surgical repair of spina bifida by applying principlesof stem cell and tissue engineering to provide an enhancedprotection of the exposed neural elements (Saadai et al., 2011,2013; Wang et al., 2015; Brown et al., 2016). The ultimategoal of these studies is to improve the neurologic function inpatients while maintaining the benefits of the existing fetalsurgical treatment.

Rapid Biosynthesis Method and Characterization of Silver Nanoparticles Using <i>Zizyphus spina christi</i>Leaf Extract and Their Antibacterial Efficacy in Therapeutic Application

Antibacterial activity of biosynthesized silver nanoparticles (AgNPs) was significant in therapeutic application of nanotechnology. These researchers studied an ecofriendly and rapid method for the first time to synthesize silver nanoparticles using Zizyphus spina christi L aqueous leaves extract (ZSE), and their antibacterial properties. The extract was found to have the potential to form silver nanoparticles at room temperature within few minutes. The green synthesized silver nanoparticles were characterized using different techniques. The UV-visible spectrum of the solution containing AgNPs showed a peak at 414 nm corresponding to the plasmon absorbance of silver nanoparticles. The transmission electron microscopy (TEM) showed that the formed particles were hexagonal in shape with appreciable Nano size ranging from 21.5 to 59.67 nm. Fourier Transform Infrared Spectroscopy analysis (FTIR) of biosynthesized AgNPs affirmed the role of ZSE as reducing and capping agent of Ag+ ions to AgNPs, and X-Ray Diffraction patterns (XRD) showed that they could be indexed as face-centered-cubic structure of silver. Antibacterial activity of AgNPs was determined by well diffusion and micro plate assay methods, showing maximum inhibition zones of 24 mm, 23 mm, 15 mm and 17 mm against Staphylococcus aureus, Acinetobacter sp., Pseudomonas aeruginosa and Escherichia coli respectively. The minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC) results showed that AgNPs had MIC, MBC of 45, 57 μg/mL, 49, 61 μg/mL, 63, 90 μg/mL and 59, 82 μg/mL against S. aureus, Acinetobacter sp., P. aeruginosa and E. coli respectively. Furthermore, the green synthesized AgNPs were loaded on band-aids and screened for antibacterial activity. The AgNPs loaded on band-aids exhibited strong antibacterial effect against multi drug resistant bacteria. These nanoparticles could be used for treating wounds and preparing wound dressing. Reflections on Health of Young Adults with spina Bifida: The Contradictory Path towards Well-Being in Daily Life

Background: An individual with SB needs to deal with long standing illnesses and is often viewed as having a disability that needs to be compensated for. This medical condition is an example of malfunction of the body, though congenital, and could be seen as an outside-perspective to the individual human being. It is important that the required medical treatment is offered to an individual with SB, but such medical treatment alone would not encompass the full health spectrum for an individual in this specific situation. A question to be raised is how this specific group of individuals experience health. Aim: To describe the experience of daily life for young adults with SB through a theoretical lens of health. Method: This qualitative study was conducted using a reflective lifeworld approach with an interpretive part. In the lifeworld theory it is understood that all our doings, feelings and thoughts are experienced through the lifeworld. The interview questions were open and non-standardised. Findings: The main theme was formulated as The contradictory path towards well-being in daily life, and was constructed on the sub-themes: Not understanding and taking responsibility for the lower body, Having people standing behind me—not being allowed to grow up myself, Compared to people like me, I usually do well, I thought it would work out by itself, and A lack of structure in daily life. Conclusion: The study indicates that young adults with SB have a diminished health and well-being and that they have a contradictory path to travel towards independency which is of great concern for this group of individuals. Differences in views of independence constitute a problem and it is important for health care providers to be aware of the individuals’ perspective on independence. This notion is something that needs to be taken into account when designing support programs for these individuals.

Spatiotemporal expression of leukemia inhibitory factor receptor protein during neural tube development in embryos with neural tube defects

Leukemia inhibitory factor receptor(LIFR),as a neuroregulatory cytokine receptor,generally shows a neuroprotective effect in central nervous system injuries.In this study,to understand the effect of LIFR on pathogenesis of neural tube defects,we explored spatiotemporal expression of LIFR at different stages of fetal development in normal and neural tube defect embryos.spina bifida aperta was induced with all-trans retinoic acid on embryonic day 10 in rats,and the spatiotemporal expression of LIFR was investigated in spina bifida aperta rats and healthy rats from embryonic day 11 to 17.Real time-polymerase chain reaction and western blot assay were used to examine mRNA and protein expression of LIFR in healthy control and neural tube defect embryos.Results of the animal experiment demonstrated that expression of LIFR protein and mRNA in the spinal cords of normal rat embryos increased with embryonic development.LIFR was significantly downregulated in the spinal cords of spina bifida aperta rats compared with healthy rats from embryonic days 11 to 17.Immunohistochemical staining showed that the expression of LIFR in placenta and spinal cord in spina bifida aperta rat embryos was decreased compared with that in control embryos at embryonic day 15.Results from human embryo specimens showed that LIFR mRNA expression was significantly down-regulated in spinal cords of human fetuses with neural tube defects compared with normal controls at a gestational age of 24 to 33 weeks.The results were consistent with the down-regulation of LIFR in the animal experiments.Our study revealed spatiotemporal changes in expression of LIFR during embryonic neurulation.Thus,LIFR might play a specific role in neural tube development.All animal and human experimental procedures were approved by the Medical Ethics Committee of Shengjing Hospital of China Medical University,China(approval No.2016PS106K)on February 25,2016.

Supplemental Feeding of Laying Hens with Wood Vinegar to Decrease the Ratio of n-6 to n-3 Fatty Acids in Eggs

A balanced ratio of fatty acids n-6 to n-3 in chicken eggs is important for health and to help prevent and manage obesity and other diseases.Traditionally,fish oil or flax seed has been utilized as feed additives to decrease the ratio of n-6 to n-3(n-6:n-3)fatty acids in eggs.The hull of spina date seed(HSDS)is a common agricultural waste product in China,from which wood vinegar(HSDSWV)may be derived.This study evaluated HSDSWV as a sup-plement in hen feeds to improve the quality of eggs and decrease the ratio of fatty acids n-6:n-3.HSDSWV was obtained via carbonization,and refined.Six concentrations(nil to 0.5%)of HSDSWV were prepared and fed to 6 hen groups,respectively,for 50 d.The fatty acids of the hen’s egg yolks were analyzed by gas chromatography/electron ionization-mass spectrometry(GC/EI-MS)in the selected ion monitoring(SIM)mode.The 0.2%HSDSWV resulted in the best egg yolk quality,with a lower percentage of linoleic acid(C18:2n6)and higher percentages of cis-5,8,11,14,17-eicosapentaenoic acid(C20:5n3)and cis-4,7,10,13,16,19-docosahexaenoic acid(C22:6n3),and thus a lower n-6:n-3 ratio compared with the other HSDSWV concentrations.In addition,the eggs contained higher levels of yolk fat and egg yolk than the controls did.In conclusion,to modify the fatty acid composition of hens’eggs and obtain a balanced ratio of n-6:n-3,0.2%HSDSWV may be considered suitable as a dietary supplement in hens’feed.

LL-37 induced cystitis and the receptor for advanced glycation end-products (RAGE) pathway

To elucidate pathways in bladder inflammation, we employed our physiologically relevant LL-37 induced cystitis model. Based on inflammatory studies involving other organ systems implicating the receptor for advanced glycation end-products (RAGE), we first hypothesized that RAGE is critically involved in LL-37 induced cystitis. We further hypothesized that?a common RAGE ligand high mobility group box 1 (HMGB1) is up-regulated in bladders challenged with LL-37. Finally, we hypothesized that NF-κB dependent inflammatory genes are activated in LL-37 induced cystitis. Testing our first hypothesis, C57Bl/6 mice were challenged with either saline (control) or 320 μM of LL-37 intravesically for 1 hr. After 12 or 24 hours, tissues were examined with immunohistochemistry (IHC) for RAGE, and both mRNA and protein isolation for respective qRT-PCR and Western Blot analysis. Our second hypothesis was tested by employing HMGB1 IHC. Testing our final hypothesis, qRT-PCR was performed investigating five genes: TNFα, IL-6, IL-1β, GM-CSF, COX-2. In control and LL-37 challenged tissues, IHC for RAGE revealed similar qualitative expression. Evaluation with qRT-PCR and Western Blot for RAGE revealed diminished expression at the mRNA and protein level within LL-37 challenged bladders. IHC for HMGB1 revealed a moderate qualitative increase within LL-37 challenged tissues. Finally, with the exception of TNFα, all NF-κB dependent inflammatory genes yielded substantial up-regulation. We have employed our LL-37 induced cystitis model to gain insight to wards a possible mechanistic pathway involved in bladder inflammation. This work provides data for future studies involving the inflammatory ligand HMGB1, RAGE, and receptor pathways that activate NF-κB.

Background: Neural tube defects have a considerable importance because they can be prevented by supplementing Folic acid & Vitamin B12 during periconceptional period and fortification of staple foods. In Morocco, the Ministry of Health launched a national program for fortification of flour with folic acid. Our goal should be to evaluate the prevalence of neural tube defects after fortification. Description: This is a retrospective descriptive at the National Reference Centre for Nutrition and Neonatology of the Children’s Hospital of Rabat over 4 years. Data were identified from the registry of congenital malformations held at the perinatology unit. Results: During the 4 years, 674 congenital malformations were identified. The neural tube defects NTDs account for 11.9%. Their annual prevalence decreased significantly from 21.78 in 2008 to 12.1 per 10,000 total births in 2011. The most common form was anencephaly (60%). Neural tube defects were isolated in 85% of cases and associated with other malformations in 15% of cases. 49.4% of infants with neural tube defects were female and 50.6% were male. Perinatal mortality in newborns with neural tube defects was 63.8% versus 25.2% in malformed newborns without neural tube defects. Conclusions: The neural tube defects seem to be common in our context. Permanent epidemiological surveillance is needed to determine the true prevalence at the national as well as its temporal trends level.
