Background:μ-opioid receptor agonists(MORAs)are indispensable for analgesia in bladder cancer(BC)patients,both during surgery and for chronic pain *** MORAs affect BC progression and metastasis remains largely *** study focused on the effects of MORAs on the formation of circulating tumor cells(CTCs)in BC and aimed to provide potential therapeutic targets,which would retain the pain-relieving effects of MORAs in BC patients without sacrificing their long-term ***:Different preclinical models were used to identify the effects of MORAs on the progression of BC.A novel immunocapture microfluidic chip was utilized to analyze whether MORAs affected the number of CTCs in mouse models and clinical BC *** analyses,total transcriptome sequencing,and molecular biology methods were then used to investigate the underlying mechanisms in these models and in BC cell ***:Mouse models of hematogenous metastasis and in situ BC demonstrated that tumor metastasis was significantly increased after MORA treatment.A significant increase in the number of mesenchymal and/or epithelial CTCs was detected after MORA treatment in both the mouse models and clinical trial ***,MORAs facilitated the formation of CTCs by activating the MOR/PI3K/AKT/slug signaling pathway,hereby promoting the epithelialmesenchymal transition(EMT)of BC cells,as knockdown of MOR,slug or blockade of PI3K inhibited the EMT process and CTC ***:MORAs promoted BC metastasis by facilitating CTC *** EMT-CTC axis could be targeted for preventive measures during MORA treatment to inhibit the associated tumormetastasis or recurrence in BC patients.

Metastasis-associated drug resistance accounts for high mortality in ovarian cancer and remains to be a major barrier for effective treatment. In this study, SKOV3/T4, a metastatic subpopulation of ovarian cancer SKOV3 cells, was enriched to explore potential interventions against metastaticassociated drug resistance. Quantitative genomic and functional analyses were performed and found that slug was significantly increased in the SKOV3/T4 subpopulation and contributed to the high resistance of SKOV3/T4. Further studies showed that slug activated c-Met in a ligand-independent manner due to elevated levels of fibronectin and provoked integrin α V function, which was confirmed by the significant correlation of slug and p-Met levels in 121 ovarian cancer patient samples. Intriguingly,c-Met inhibitor(s) exhibited greatly enhanced anti-cancer effects in slug-positive ovarian cancer models both in vitro and in vivo. Additionally, IHC analyses revealed that slug levels were highly correlated with reduced survival of ovarian cancer patients. Taken together, this study not only uncovers the critical roles of slug in drug resistance in ovarian cancer but also highlights a promising therapeutic strategy by targeting the noncanonical activation of c-Met in slug-positive ovarian cancer patients with poor prognosis.

Process reinforcement research was carried out in Y-junction slug flow microreactors under variable conditions,using EHEHPA(2-ethylhexyl phosphonic acid mono-2-ethylhexyl)to extract yttrium(Ⅲ)from hydrochloric acid *** influence of pH and flow rate on the extraction-reaction efficiency,slug size,mass transfer performance was *** experimental results showed that maximum extraction-reaction efficiency of 91.85%can be achieved in the smallest microreactor(residence time is 11.25 s),which was significantly higher than traditional extraction requiring more than 10 *** slug length tended to decrease with the increase of pH and flow *** mass transfer coefficients for the slug flow microreactors were found to be in the range of 1.39×10^-1-1.642s^-1 that is several orders of magnitude higher as compared to traditional extractors,which testified the beneficial effects of Y-junction slug flow *** mass transfer reinforcement has prompted the use of microreactor as a suitable alternative to traditional extractors,which can advantageously improve the economic and environmental development of mineral processing.

slug catcher is an important and costly equipment in the up-stream assets,but is the less supported by research or commercial simulation software in terms of design and sizing *** purpose of this paper is to present a simulation model based on the open-source software for computational fluid-dynamics OpenFOAM®,which can assist during the preliminary sizing,made by the usual simplified methods,proving the fluid-dynamic behaviour and drive the process engineers to an optimum final design by checking all the expected production profiles of the related oil/gas field,for a comprehensive and safe operation all along the reservoir *** slug catcher main function is to ensure the separation of the phases within the acceptance specifications dictated by the process units of downstream CPF(Central Production Facility).The optimization of the slug catcher could allow the reduction of the equipment overall dimensions,that is essential to reduce the capital expenditure and make the installation more *** graphic output is a strong tool in the hand of engineers,following the fluid-dynamic response of the slug catcher in the gas-liquid separation section,in function of the flow pattern at the entrance and superficial velocities of the phases.

Posterior capsular opacification(PCO)is linked to the pathological process of lens epithelial cells,which includes proliferation,migration,and epithelial-mesenchymal transition(EMT).Our goal was to investigate whether long noncoding RNA(lncRNA)XIST contributes to EMT via targeting miR-124/slug axis in TGF-β2-induced SRA01/04 *** was induced by stimulating SRA01/04 cells with 10 ng/mL TGF-β2 for 24 h,and PCO microenvironment was *** expressions levels of lncRNA XIST,miR-124,and slug were measured by realtime polymerase chain reaction(RT-PCR)and western *** role and mechanism of lncRNA XIST in promoting EMT of TGF-β2-treated SRA01/04 cells were investigated by using cell transfection,cell counting kit-8(CCK-8),immunofluorescence staining,transwell assay,wound-healing assay,RT-PCR,western blot and dual-luciferase reporter *** expression of slug and lncRNA XIST was markedly increased,while miR-124 was downregulated in TGF-β2-treated SRA01/04 cells compared with the control *** of lncRNA XIST reduced EMT,migration,and cell viability in TGF-β2-induced SRA01/04 cells;moreover,it adversely modulated miR-124 and adjusted the expression of slug in SRA01/04 cells,while these effects were diminished by co-transfection with *** data demonstrated that lncRNA XIST functioned as a competitive endogenous RNA(ceRNA)of miR-124 to modulate the expression level of slug,thereby modulating EMT,migration,and cell viability in SRA01/04 *** conclusion,lncRNA XIST has a crucial role in promoting TGF-β2-induced EMT via modulating the miR-124/slug axis in SRA01/04 cells,and it may provide a novel therapeutic option for PCO treatment.

AIM To evaluate the immunohistochemical(IHC) expression of five biomarkers, commonly involved in epithelial mesenchymal/mesenchymal epithelial transition(EMT/MET), in gastrointestinal stromal tumors(GISTs). METHODS In 80 consecutive GISTs the IHC examinations were performed using the EMT-related antibodies E-cadherin,N-cadherin, slug, V-set and immunoglobulin domain containing 1(VSIG1) and CD44. RESULTS The positivity rate was 88.75% for slug, 83.75% for VSIG1, 36.25% for CD44 and 10% for N-cadherin. No correlation was noted between the examined markers and clinicopathological parameters. Nuclear positivity for slug and VSIG1 was observed in all cases with distant metastasis. The extra-gastrointestinal stromal tumors(e-GISTs) expressed nuclear positivity for VSIG1 and slug, with infrequent positivity for N-cadherin and CD44. The low overall survival was mainly dependent on VSIG1 negativity(P = 0.01) and nuclear positivity for slug and/or CD44. CONCLUSION GIST aggressivity may be induced by nuclear upregulation of slug and loss or cytoplasm-to-nuclear translocation of VSIG1. slug and VSIG1 may act as activated nuclear transcription factors. The CD44, but not N-cadherin, might also have an independent prognostic value in these tumors. The role of the EMT/MET-related transcription factors in the evolution of GISTs, should be revisited with a larger dataset. This is the first study exploring the IHC pattern of VSIG1 in GISTs.
