Association between the serotonin 1A receptor C（-1019）G polymorphism and major depressive disorder in the northern Han ethnic group in China

Background Recent studies have suggested that susceptibility to major depressive disorder （MDD） might be related to the serotonin 1A receptor （5-HTR1A） C（-1019）G polymorphism. In this study, we aimed to assess the association between 5-HTR1A C（-1019）G polymorphism and MDD in the Northern Han ethnic group of China. Methods The C（-1019）G of 5-HTR1A was detected with polymerase chain reaction （PCR） in 400 patients with MDD and 400 unrelated age- and sex-matched healthy control subjects. Association between the C（-1019）G and MDD was statistically analyzed. Results There was a statistically significant difference between MDD patients and controls in both the genotype distribution （X^2=10.913, df=2, ,P=0.004） and the allele frequency （X^2=10.379, df=1, P=0.001 ）, and a significant difference in the genotype distribution and the allele frequency was found both in the female subjects （Genotype distribution： X^2=15.406, df=2, P=0.000; allele frequency： X^2=15.552, df=1, P=0.000） and the late-onset subjects （Genotype distribution X^2=7.771, df=2, P=0.021 ; allele frequency： X^2=8.007, df=1, P=0.005） in the two groups.Conclusion These results suggest that 5-HTR1A C（-1019）G polymorphism is probably associated with MDD and it is likely to be the susceptible gene locus for the female and late-onset MDD.

＜正＞It is well known that after spinal cord lesion the body part below the lesion first develops an areflexia and then it is followed by a slow return of reflexes,which eventually becomes increased stretch reflexes（spasticity） and hyperreflexia weeks or months after the injury.The pathophysiology of the spasticity has been related to:（1） specific changes in cellular properties of motoneurons,（2） changes in reflex transmission and /or（3） morphological reorganization.serotonin （5-HT） is one of the important neurotransmittors that regulate the motor function.Earlier studies have shown that after complete spinal transection the spinal motoneurons below the lesion become supersensitive to systematic administration of serotonin（5-HT） precursor,such as 5-hydroxytryptophan（Shibuya and Anderson,1968）.However,the mechanism of this denervation supersensitivity is still largely unknown.Recent studies have shown that after chronic lesion the spinal motoneurons below the lesion exhibit a 30-fold supersensitivity to 5-HT and this is mainly mediated by the activation of 5- HT2A（5-HT2AR） and 2C receptors（Harvey et al.,2006）.To see if this supersensitivity is caused by 5-HT receptor upregulation we have in this study investigated the changes of 5HT2AR immunoreactivity（IR） following acute and chronic spinal transection at the sacral spinal cord in the rat.Five time points were examined after spinalization:2 hour,8 hour,24 hour,48 hour and 60 day.The 5-HT2AR-IR density was analyzed in the ventral horn motoneurons and their dendrites below the lesion in both spinalized and sham-operated animals.The results showed that lesion after 8 hours or earlier did not cause any changes of 5-HT2AR-IR.After 24 hours the expression of 5-HT2AR-IR was seen to be upregulated in the spinal motoneuron somata and their dendrites,with that in the spinalized rats 3-fold higher than in the shamoperated rats.After 48 hours the spinalized rats showed the same level of upregulation as the animals after 24 hour lesion. ＜正＞ The aim of the present study was to investigate whether the recreational drug （ + /-）-3,4-methylenedioxymetamphetamine （MDMA） would affect mRNA of serotonin and dopamine receptors. Male Sprague - Dawley rats received either 3×1 or 3×5 mg/kg/day （3 hours apart） every 7: th day during 4 weeks. Real time RT-PCR was used to determine the mRNA levels of serotonin 5HT1A, 5HT1B, 5HT2A, 5HT2C, 5HT3, 5HT6 receptors and dopamine D1, D2, D3 receptors in seven brain nuclei. Results could be highlighted by profound MDMA induced increase of the 5HT1B-receptor mRNA in the cortex, caudate putamen, nucleus ac-cumbens, and hypothalamus at the highest dose. In addition, an inverse correlation between MDMA induced pellet consumption and 5HT2C mRNA levels was observed in the hypothalamus. This study is concluded to provide evidence for a unique implication of serotonin rather than dopamine receptor mRNA levels, in response to repeated intermittent MDMA administration.

Association of serotonin receptors with Attention Deficit Hyperactivity Disorder： A Systematic Review and Meta-analysis

Attention deficit hyperactivity disorder （ADHD） is one of the most common mental disorders in childhood, with a high heritability about 60% to 90%. serotonin is a monoamine neurotransmitter. Numerous studies have reported the association between the serotonin receptor family （5-HTR） gene polymorphisms and ADHD, but the results are still controversial. In this study, we conducted a meta-analysis of the association between 5-HTRIB, 5-HTR2A, and 5-HTR2C genetic variants and ADHD. The results showed that the 861G allele of 5-HTRIB SNP rs6296 could significantly increase the risk of ADHD （OR= 1.09, 95% CI： 1.01-1.18）; the 5-HTR2C gene rs518147 （OR=1.69, 95% CI： 1.38-2.07） and rs3813929 （OR = 1.57, 95% CI： 1.25-1.97） were all associated with the risk of ADHD. In addition, we also carried on a case- control study to explore the relevance between potential candidate genes 5-HTR1A, 5-HTRIE, 5-HTR3A and ADHD. The results indicated that 5-HTRIA rs6295 genotype （CC＋CG vs. GG OR=Z00, 95% CI： 1.23-3.27） and allele （OR=1.77, 95% CI： 1.16-2.72） models were statistically significantly different between case group and control group. This study is the first comprehensive exploration and summary of the association between serotonin receptor family genetic variations and ADHD, and it also provides more evidence for the etiology of ADHD.

Objective We investigated the regulatory effect of 5-HT receptors on spontaneous epileptiform activity in cortical slices.Methods We performed whole-cell recording and extracellular recording from entorhinal-hippocampal slices maintained in vitro.Spontaneous epileptiform activities could be obtained in the presence of picrotoxin,a GABA receptor blocker.We then examined the role of 5-HT and its receptor agonists or antagonists in regulating these epileptiform events.Results Bath application of 5-HT reduced the occurrence frequency of epileptiform activity,and this inhibitory effect could be completely blocked by the pretreatment of 5-HT receptor antagonist WAY-100635.Further experiments in slices obtained from 5-HT knockout mice revealed that 5-HT prolonged the duration of individual epileptiform discharges,and this excitatory effect could be mimicked by 5-HT receptor agonist TCB-2 and blocked by the antagonist ketanserin.Conclusion Our results indicate that activation of 5-HT receptors mediates the inhibitory effect of 5-HT on epileptiform activities,while activation of 5-HT receptors promote the induction of long-lasting epileptic discharges.
