Background:The immune system is one aspect of health that is affected by dietary selenium(Se)levels and selenoprotein *** is an important immune organ of the body,which is directly involved in cellular ***,there are limited reports on Se levels and spleen ***,this study established a Se-deficient pig model to investigate the mechanism of Se deficiency-induced splenic ***:Twenty-four pure line castrated male Yorkshire pigs(45 days old,12.50±1.32 kg,12 full-sibling pairs)were divided into two equal groups and fed Se-deficient diet(0.007 mg Se/kg)or Se-adequate diet(0.3 mg Se/kg)for 16 *** the end of the trial,blood and spleen were collected to assay for erythroid parameters,the osmotic fragility of erythrocytes,the spleen index,histology,terminal deoxynucleotidyl transferase nick-end labeling(TUNEL)staining,Se concentrations,the selenogenome,redox status,and signaling related inflammation and ***:Dietary Se deficiency decreased the erythroid parameters and increased the number of osmotically fragile erythrocytes(Pdeficiency decreased the Se concentration and selenoprotein expression in the spleen(Pdeficiency activated the NF-κB and HIF-1αtranscription factors,thus increasing pro-inflammatory cytokines(IL-1β,IL-6,IL-8,IL-17,and TNF-α),decreasing anti-inflammatory cytokines(IL-10,IL-13,and TGF-β)and increasing expression of the downstream genes COX-2 and iNOS(Pdeficiency induced apoptosis through the mitochondrial pathway,upregulated apoptotic genes(Caspase3,Caspase8,and Bak),and downregulated antiapoptotic genes(Bcl-2)(P<0.05)at the mRNA

selenium(Se) and selenoproteins play important roles in the immune system and Se deficiency will affect immune functions in *** Se can influence the expression levels of selenoproteins,however,little is known about the expression patterns of *** aim of the present study was to investigate the effect of dietary Se deficiency on the expressions of 23 selenoproteins in broiler chicken immune *** this study,one hundred and fifty broiler chicks(1 day old;Weiwei ***.,Harbin,China) were randomly divided into two groups(75 chickens per group).The chickens were maintained either on a diet supplemented with Se through the addition of 0.2 mg/kg Se(C group) via sodium selenite or on a Se-deficient granulated 76 diet(L group,from the Sedeficient region of Heilongjiang Province in China,containing 0.008 mg/kg Se) until the broiler chicks exhibited ED *** euthanasia,the samples from the immune tissues(including the spleen,thymus and bursa of Fabricius) were quickly collected,and the mRNA expression levels of 23 selenoproteins were examined by realtime quantitative PCR and analyzed using the way of principal *** results showed that Se deficiency decreased the mRNA levels of 23 selenoproteins respectively in the thymus,spleen and bursa of Fabricius tissues of broiler chickens in the L ***,we found that the mRNA levels of Diol in the thymus,Txnrd2 in the spleen,and Txnrd3 in the bursa of Fabricius decreased mostly among 23 selenoproteins(90.9%,83.3%,96.8%).And in addition,principal component analysis(PCA) suggested Gpxs,Txnrds,and Dios families may play crucial roles in chicken immune *** is well known Gpxs,Txnrds were great concerned with *** results indicated that Se deficiency mainly influenced the expressions of antioxidative selenoproteins(Gpxs,Txnrds) in chicken immune ***,antioxidative selenoproteins especially Gpxs may play a crucial role in chicken immune ***,it helps us f

[Background]selenium（Se） plays an important role in regulating various metabolic processes in the *** raised considerable expectations for the prevention of vascular disease,cancer,and other chronic ***,the exact mechanism still needs to be clarified.[Objective]Establishment the chicken model of Se deficiency disease,and enunciate the mechanisms how Se induces vein injury.[Methods]Broilers were divided into two groups,fed deficient Se diets and common diets respectively for 30 *** euthanasia,veins were quickly removed blotted and then rinsed with ice-cold sterile deionized water,frozen immediately in liquid nitrogen,and stored at -80 ℃ until *** antioxidant related factors（GSH,MDA and H2O2） were determined using detection kits,mRNA expression of 25 selenoproteins,the mRNA and protein levels of inflammatory cytokines and heat shock proteins were detected by RT-PCR and Western *** of the tissues（5μm thick） were cut and stained with hematoxylin and eosin（H & E） and examined under microscope,TUNEL assay was using the in situ cell death detection kit.[Results]It was observed that,in broiler vein,the deficient Se diets markedly decreased the antioxidant including the activity of GPx,and the content of GSH and *** 25 selenoproteins,the expressions of 20 selenoproteins in Se deficiency group were decreased compared to the Se common *** content of H2S,the protein expression of 3-MST,CSE,NO,iNOS and the inflammatory cytokines in Se deficiency group were increased compared to the Se common group,the expression of heat shock proteins were also present a similar *** analysis showed,in Se deficiency group,karyopyknosis and chromatin *** adipose around vessels,surrounded and infiltrated by massive inflammatory cells and *** percent of apoptotic cells was increased in the Se deficiency group compared to the Se common group.[Conclusion]Selenoproteins,inflammatory cytokines and HSPs,they

Effect of dietary selenium deficiency on mineral elements in myocardium of chicken

selenium(Se) deficiency induces myocardial necrosis in chickens,but the exact mechanism remains *** objective of this study was to investigate the effect of dietary Se deficiency on 26 mineral elements in myocardium of *** hundred and eighty 1-day-old chickens were randomly divided into two *** control-Se group(C group) was fed a medium-Se diet containing 0.2 mg Se/kg and the low-Se group(L group)was fed a Se-deficient basal diet containing 0.033 mg Se/*** the end of the 55 days period,myocardium was collected and the contents of 26 mineral elements lithium(Li),boron(B),natrum(Na),magnesium(Mg),aluminium(AI),silicium(Si),kalium(K),calcium(Ca),vanadium(V),chromium(Cr),manganese(Mn),ferrum(Fe),cobalt(Co),nickel(Ni),copper(Cu),zinc(Zn),arsenic(As),selenium(Se),molybdenum(Mo),cadmium(Cd),stannum(Sn),stibium(Sb),barium(Ba),hydrargyrum(Hg),thallium(Tl) and plumbum(Pb)were assayed by inductively coupled plasma mass spectrometry(ICP-MS).The results indicate that 10 element levels were significantly changed(Pdeficiency leads to a disorder of 10mineral(Li,Mn,Fe,Cu,Zn,Se,Sb,Ba,Hg and Pb) in myocardium of chicken.

Preparation of Organic Selenocystine Using Locally Isolated Bread Yeast

In contrast to the toxic inorganic forms of selenium to life at very low concentrations, the organic-selenium compounds are of considerable interest and several of them play essential roles in cell biochemistry and nutritional science. Se-yeast （selenium-enriched yeast） is a common form of selenium used to supplement dietary intake of this important trace mineral. In the present study, we tried to prepare an organic selenocystine using locally isolated bread yeast （Saccharomyces cerevisiae）. A novel locally prepared date extract media enriched by addition of 0.2% KH2PO4 （potassium phosphate）, 0.6% ammonium sulfate was adopted as alternative culture media. Differences concentrations of selenium salt （30, 60, 120 and 240 lag/mL） were added to the yeast culture media. While the best concentration of selenium added was 30Bg/mL, it achieved optimal conditions for the growth of yeast and the production of red yeast growth identical to the standard. The products （organic selenocystine） were analyzed by HPLC （High Performance Liquid Chromatography） and AAS （Atomic Absorption Spectrophotometer） comparing with authentic standard obtained from Sigma. Results confirmed the formation of similar selenocystine products.

Se deficiency affected the mRNA expression of inflammatory factors and twenty-five selenoprotein genes in kidney of broiler chicks

The aim of the present study was to investigate effects of Se deficiency(SeD) on the mRNA expression of inflammatory factors and 25 selenoprotein genes in kidney of broiler chicks.150 1-day-old broiler chicks were randomly assigned to two groups fed with a low-Se diet(SeD group,0.033 mg/kg Se)or adequate Se diet(C group,0.2 mg/kg Se).The levels of uric acid(UA)and creatinine(Cr)in the serum,the mRNA levels of 6 inflammatory factors and 25 selenoprotein genes in kidney were determined as the symptom of SeD *** results showed that the contents of UA and Cr in the serum were significantly increased in the SeD *** mRNA levels of inflammatory factors(NF-κB,iNOS,COX-2,TNF-α)were significantly increased in SeD ***,the mRNA levels of PTGEs and HO-1 were not significantly changed in SeD *** addition,25 selenoprotein transcripts displayed a ubiquitous expression in kidney of *** mRNA levels of 14 selenoprotein genes(Dio1,Dio2,GPx3,Sepp1,SelH,SelI,SelK,Sepn1,SelO,SelW,Sell5,SelT,SelU and SelS)were decreased and 9selenoprotein genes(GPx1,GPx2,GPx4,SelPb,TrxR1,TrxR2,TrxR3,SPS2 and SelM)were up-regulated in the SeD group,but the changes of Dio3 and Selx1 mRNA levels were not significant(P> 0.05).The results indicated that Se deficiency induced the abnormal renal *** change of the inflammatory factors mRNA played an important role in the abnormal renal functions induced by SeD.25 selenoprotein genes showed a different expression in SeD group.

Anti-inflammatory effect of dietary selenium in LPS-induced model of mastitis mouse

Mastitis is a major cause of economic losses to the dairy ***(Se),a biological trace essential element,benefits health and immunity of the mammary *** the present study,we investigate that effects of dietary selenium on LPS-induced models of mammary mastitis in *** female BALB/c mice were randomly divided into three groups and fed with daily rations that contain 0.087 mg/(kg·BW),0.15 mg/(kg·BW) and 1.5 mg/(kg·BW) sodium *** the end of experiment,an intramammary infusion of LPS was used to establish models of mastitis in *** analysis,enzymatic activity detection and expression of pro-inflammatory genes were conducted to study effects of dietary selenium on LPS-induced models of mammary mastitis in *** showed that selenium can affect the activity of glutathione peroxidase and mRNA expression of GPX1 and GPX4 of mammary gland in a dose dependent *** observation of mammary gland in models of mammary mastitis in mouse showed more infiltration of inflammatory *** expression of TNF-α,IL-1β,IL-6,COX-2 and iNOS of mammary gland of Se-deficient group were higher than Se-adequate *** results showed that Se can affect LPS-induced inflammatory responses of mammary gland.
