Coexistence of nitric oxide synthase with γ-aminobutyric acid during seizure induced by kainic acid in SD rats

Objective: Functional significance of NO and central inhibitory neurotransmitter γ-aminobutyric(GABA) during seizures were investigated morphorlogically. Methods: A kainate-induced complex partialseizure model was used in our experiment. Twenty SD rats were randomly divided into KA 30, 60, 90, 200min and control groups. The brain sections were stained by NADPh (nicotinamide adenine dinucleotide phosphate ) diaphorase (Nd ) histochemically, and were further stained by GABA immunohistochemically.Results: Histological and immunohistochemical study revealed that in KA groups the number of Nd and GABA-positive double labelled neurons in CA3 region, CA3 region and dentate gyms was significantly reduced,compared with the control group. Conclusion: Nd coexisted with GABA in the brain. Reduction of GABA release led to relief of GABA-ergic inhibition and in the same way, reduction of NO release weakened its negative feedback modulation. Therefore neuronal synchronous paroxysmal discharges increased. GABA and NO,both having antiepileptic action, acted through different ways or different link in the same way. NO may involve in the effect of GABA-ergic neurons and play cooperative antiepileptic action with GABA.

Rasmussen's encephalitis (RE), which was first described by Rasmussen in 1958, is a rare, dispersed, and progressive neurological syndrome that is characterized by focal epilepsy, unilateral inflammation of the cerebral cortex, progressive hemiplegia and cognitive deterioration. The etiology of this syndrome remains under investigation, and it is hypothesized and widely accepted that RE is a T-cell-mediated autoimmune response. However, the antigenic epitopes and mechanisms are still unknown. The pathological characteristics of RE are cortical inflammation, neuronal loss, and gliosis that are confined to one cerebral hemisphere. Hemispherectomy remains the only cure for the seizures and cognitive deterioration associated with the disease, but this procedure results in inevitable functional loss in the brain. Compared with surgery, immunomodulatory treatments are expected to cause less neurological deficits, but with limited clinical effect.

Magnesium (Mg) is an essential cofactor for many enzymatic reactions, especially those involved in energy metabolism. The aim of the present study was to determine the CSF concentration of Mg in various neurological disorders (n = 72) and in healthy subjects (n = 75). The control group included 35 males and 40 females, aged 16-89 years (mean age 53 years) who were subjected to a lumbar puncture for diagnostic reasons. The CSF examination was normal mainly as concerns the macroscopically examination, the leukocyte count and the protein level. The determination of Mg was performed with xylidyl-blue photometry. Our normal CSF Mg mean value was 0.97 ± 0.08 mmol/l (range 0.6-1.4 mmol/l). In the group of patients (n = 11) with convulsive seizures a slightly but significantly lower Mg were revealed (0.92 ± 0.03 mmol/l;p = 0.001;paired two-tailed Student’s t-tests). No statistically significant change of CSF Mg levels was noted in patients suffering from alcohol withdrawal syndrome, multiple sclerosis or Bell’s palsy. Our results indi-cate that magnesium deficiency may play a role for seizure manifestation even in patients with a moderate low Mg without neurological signs. Low CSF magnesium is associated with epilepsy, further studies may determine the influ-ence of anti-epileptic drug therapy on CSF magnesium levels.

Supratentorial cortical ependymoma is a rare primary brain tumor in adults. Only 50 cases are reported in the literature including pediatric and adult patients. We report a 30-year-old male with sensory-type seizures who was found to have a right posterior frontal cortical lesion with large cystic component. The lesion was resected completely and histopathology and immunohistochemistry examinations confirmed anaplastic ependymoma WHO grade III. The patient received radiotherapy treatment post-surgery, and he was tumor free one year after treatment.
