scorpions,through ruthless survival of the fittest,evolve the unique ability to quickly locate and hunt prey with slit receptors near the leg joints and a sharp sting on the multi-freedom *** by this fantastic creature,we herein report a dual-bionic strategy to fabricate microcrack-assisted wrinkle strain sensor with both high sensitivity and ***,laserinduced graphene(LIG)is transferred from polyimide film to Ecoflex and then coated with silver paste using the casting-andpeeling and prestretch-and-release *** shape-adaptive and long-range ordered geometry(e.g.,amplitude and wavelength)of dual-bionic structure is prestrain-tuned to optimize the superfast response time(~76 ms),high sensitivity(gauge factor=223.6),broad working range(70%–100%),and good reliability(>800 cycles)of scorpion-inspired strain sensor,outperforming many LIG-based materials and other bionic *** alternate reconnect/disconnect behaviors of slit-organlike microcracks in the mechanical weak areas initiate tremendous resistance changes,whereas the scorpion-tail-like wrinkles act as a“bridge”connecting the adjacent LIG resistor units,enabling reversible resilience and unimpeded electrical linkages over a wide strain *** with the self-developed miniaturized,flexible,and all-in-one wireless transmission system,a variety of scenarios such as large body movements,tiny pulse,and heartbeat are real-time monitored via bluetooth and displayed in the client-sides,revealing a huge promise in future wearable electronics.

scorpion venom heat-resistant peptide(SVHRP)is a component purified from Buthus martensii Karsch scorpion venom. Our previous studies have shown that SVHRP is neuroprotective in models of Alzheimer’s disease and Parkinson’s disease. The present study aimed to explore the potential neuroprotective effects of SVHRP on cerebral ischemia/reperfusion(I/R) injury, using a mouse model of middle cerebral artery occlusion/reperfusion(MCAO/R) and a cellular model of oxygen-glucose deprivation/reoxygenation(OGD/R). Our results showed that SVHRP treatment decreased the neurological deficit scores, edema formation, infarct volume and neuronal loss in the MCAO/R mice, and protected primary neurons against OGD/R insult. SVHRP pretreatment suppressed the alterations in protein levels of N-methyl-D-aspartate receptors(NMDARs) and phosphorylated p38 MAPK as well as some proinflammatory factors in both the animal and cellular models. These results suggest that SVHRP has neuroprotective effects against cerebral I/R injury, which might be associated with inhibition of the NMDA-MAPKmediated excitotoxicity.

Recently,micro-vibrational perception mechanisms of nocturnal arthropods such as scorpions and spiders are attracting increasingly more attention and *** relevant micro-vibrational receptors are exquisite,in terms of their comprehensive performance such as sensitivity,stability,high anti-interference,and ultralow-power *** this work,we find the Basitarsal Compound Slit Sensilla (BCSS) of scorpion (Heterometrus petersii) are composed of the crack-shaped slits as mechanosensory structure and can efficiently detect substrate-borne vibrational signal in complex natural *** study on microstructures and mechanical properties of tissue phases constituting the BCSS reveals that the strategy of tessellation is used to make crack-shaped slit amplify the tiny mechanical *** addition,the magnitude-frequency characteristics of electrophysiological signals caused by vibration stimulation with different frequencies indicate that the scorpion is sensitive to micro-vibrational signals at a certain frequency ***,the vibrational perception mechanism based on geometrical amplification and resonance is proposed to explain how scorpions detect the tiny biotic vibrational signal efficiently in noise *** finding not only promotes our further understanding of ultra-sensitive mechanism of the vibrational receptors,but also provides biological inspiration for the next generation of mechanosensor for a broad range of applications.

Amino Acid Sequence of an Excitatory Insect-selective Toxin (BmK IT) From Venom of the scorpion Buthus martensi Karsch

The insect-selective neurotoxin(BmK IT) of scorpion Buthus martensi Karsch was first reduced and S-alkylated, and then digested by TPCK-trypsin and Staphylococcus aureus V-8 Protease. The enzymatic peptides were purified on TLC-plastic sheet and submitted to determine their amino acid compositions and sequences. The sequence of the 70 amino acid residues of BmK IT was established with reference to the primary structure of AaH IT, another excitatory insect-selective toxin from the venom of North African scorpion Androctonus australis Hector. About 75% of the homologous sequence was found in the molecules of BmK IT and AaH IT. It is obvious that the results contribute toward better understanding of the molecular structure characteristics, structure/activity relationship of scorpion insect-selective toxins, and they can serve as the molecular basis for utilizing the toxins as a tool to clarify molecular mechanism involved in channel gating, and to infer the possibility of developing them as new selective bioinsecticides.

The scorpion Venom Peptide Smp76 Inhibits Viral Infection by Regulating Type-Ⅰ Interferon Response

Dengue virus(DENV) and Zika virus(ZIKV) have spread throughout many countries in the developing world and infect millions of people every year, causing severe harm to human health and the economy. Unfortunately, there are few effective vaccines and therapies available against these viruses. Therefore, the discovery of new antiviral agents is ***, a scorpion venom peptide(Smp76) characterized from Scorpio maurus palmatus was successfully expressed and purified in Escherichia coli BL21(DE3). The recombinant Smp76(rSmp76) was found to effectively inhibit DENV and ZIKV infections in a dose-dependent manner in both cultured cell lines and primary mouse macrophages. Interestingly,rSmp76 did not inactivate the viral particles directly but suppressed the established viral infection, similar to the effect of interferon(IFN)-b. Mechanistically, rSmp76 was revealed to upregulate the expression of IFN-b by activating interferon regulatory transcription factor 3(IRF3) phosphorylation, enhancing the type-Ⅰ IFN response and inhibiting viral *** mechanism is significantly different from traditional virucidal antimicrobial peptides(AMPs). Overall, the scorpion venom peptide Smp76 is a potential new antiviral agent with a unique mechanism involving type-Ⅰ IFN responses,demonstrating that natural AMPs can enhance immunity by functioning as immunomodulators.

Preliminary crystallographic studies of a neurotoxin with a high toxicity from Chinese scorpion Buthus martensii Karsch

scorpion toxins are a family of small neurotoxic proteins with high selectivity. Gener-ally, most of the toxins are a group of basic homologous polypeptides containing about60—80 amino acids except BmK Ⅳ whose pl is 5.3. It has been reported that theybind to various ion channels with high affinity and selectivity. In spite of the significantsequence similarity found among different toxins, they display various degrees of toxicityand specificity to different animal species. So far they form a good system to study thestructure-function relationship.

Crystallization and preliminary X-ray analysis of an anti-insect toxin BmK 16 from Buthus martensii Karsch scorpion

An excitatory anti-insect toxin from BmK scorpion, named BmK 16, has been purified and crystallized recently. Two crystal forms have been obtained by using ammonium sulfate as precipitant at different pH, one of which is single crystal. Diffraction data have been collected for the single crystal on a Mar Research 345 Image Plate. The analyses of the data show that the crystal can diffract to 0.28 nm resolution and belongs to the space group C2 with unit cell parameters a = 4.65 nm, b = 8.52 nm, c= 3.26 nm, α= γ= 90oooooooo, β= 110.5°. There are 2 molecules per asymmetric unit according to its unit cell volume and molecular weight.
