The role and mechanism of macrophage activation induced by exosomes from high glucosetreated renal tubular epithelial cells

Objective To investigate the role and mechanism of macrophage activation induced by exosomes from high glucose-treated renal tubular epithelial cells.Methods(1)The supernatant of renal tubular epithelial cells which were cultured in normal glucose control group(5.5 mmol/L D-glucose)or high glucose group(30.0 mmol/L D-glucose)for 48 h were collected and ultracentrifuged to harvest exosomes.Exosomes were identified under transmission electron microscope and using Western blotting.(2)Exosomes were labeled with the green lipophilic fluorescent dye PKH67 and cultured with THP-1 macrophage to investigate whether HK2-derived exosomes could be internalized by THP-1 macrophage.The morphology was observed microscopically and the chemotaxis function of THP-1 macrophages was detected in Transwell chamber after co-cultured with exosomes.The expression of inducible nitric oxide synthase(i NOS),tumor necrosis factor-α(TNF-α),Interleukin-1β(IL-1β),monocyte chemoattractant protein-1(MCP-1)in cells and supernatants were separately detected by quantitative Real-time PCR(qRT-PCR)and enzyme linked immunosorbent assay(ELISA),and the expression of pc-Jun NH2-terminal kinase(p-JNK),mitogen-activated protein kinase p-p38(p-p38 MAPK)and nuclear factor κB p65(NF-κB p65)in THP-1 macrophages were detected by Western blot.Results(1)Vesicles harvested by ultracentrifugation ranged in size from 30 nm to 100 nm and expressed exosomal marker CD63,TSG101 but absence of calnexin which is a marker of endoplasmic reticulum suggested that the exosomes were not contaminated with cells.(2)Results from laser scanning confocal microscope showed that each group of exosomes could be internalized by THP-1 macrophages.Compared with normal glucose exosomes group,high glucose exosomes increased the expression of i NOS,TNF-α,IL-1β and MCP-1 in THP-1 macrophages(all P<0.01),moreover,p-JNK,p-P38 MAPK and NF-κB p65 proteins level also increased significantly(all P<0.01).Conclusion Exosomes from high glucose-treated HK2 cells can induce THP-1 macrophage activation a

The role of Chinese herb in the treatment of disease:Science Fiction or Fact?

The clinical efficacy and safety of proprietary Chinese herb has become a huge concern to the public. There are still many weak points in this field, which makes post-marketing reappraisal necessary. The editorial by Ran Tian et al. introduced a new concept, 'Precise proprietary Chinese medicine', with six detailed steps to facilitate the post-marketing reappraisal. The insight provided by the authors is unique and novel. Ganduqing Granule is an approved natural traditional Chinese medicine in China (approved No. 2000-009). Mei Wei et al. discovered that Ganduqing Granule could balance the metabolic disorders related to lipids and bile acids as well as relieve oxidative stress in patients with chronic hepatitis B, which will help conduct the post-marketing reappraisal of Ganduqing Granule.

Potential role of lung dendritic cells and Th17/regulatory T cells in patients with chronic obstructive pulmonary disease

Objective To explore the role of lung dendritic cells(DCs) and Th17 /regulatory T cells ( Treg) pathway inthe pathogenesis of chronic obstructive pulmonary disease(COPD). Methods COPD patients who received lobectomyfrom Sep. 2015 to Mar. 2016 in our hospital wereenrolled and classified into non-smoking non-COPDgroup,smoking without COPD group and COPD group.The expression of CD80,chemokine recepter-6 (CCR6),interleukin-17A ( IL-17A) and fork-head transcriptionfactor P3 (FoxP3) were detected by immunohistochemistry(IHC) in lung tissue. Mature DCs (mDCs),immatureDCs ( imDCs),Th17 cells and Treg cells in lungtissue were detected by flow cytometry ( FCM) and thecorrelation between Th17 /Treg cells with lung functionwas analyzed. Results (1) The expression of CD80 andFoxP3 in COPD group was decreased,while the expressionof CCR6 and IL-17A was increased (P < 0. 05).

Generally, ROS are regarded as host defense molecules that are released by neutrophils to destroy exogenous pathogens such as bacteria and to act as secondary messengers in signal transduction. However, increased production of ROS is also involved in inducing pathophysiological changes such as apoptosis, cell cycle disruption, and necrosis. For this reason, induction of ROS-mediated damage in cancer cells by pharmacological agents that either promote ROS generation or disable the cellular antioxidant system is considered a potential therapeutic strategy for preferentially killing cancer cells[1].

Analysis of the role“Nick Carraway”in The Great Gatsby
