Surface energy-mediated fibronectin adsorption and osteoblast responses on nanostructured diamond

Nanostructured diamond have potential applications in many biomedical related fields and demonstrated extraordinary capacity to influence cellular responses. Studying the surface property of nanodiamond and its influence to protein adsorption and subsequent cellular responses along with the mechanism behind such capacity becomes more important. Here the role of surface energy associated with nanostructured diamond in modulating fibronectin and osteoblast(OB, bone forming cells) responses was investigated. Nanocrystalline diamond(NCD) and submicron crystalline diamond(SMCD) films with controllable surface energy were prepared by microwave-enhanced plasma chemical vapor deposition(MPCVD) techniques. Fibronectin adsorption on the diamond films with varied surface energy values was measured via the enzyme-linked immunosorbent assay(ELISA) and the relationship between the surface energy and fibronectin adsorption was studied. The result indicated that fibronectin adsorption on nanostructured surfaces was closely related to both surface energy and material microstructures. The spreading and migration of OB aggregates(each containing 30–50 cells) on the NCD with varied surface energy values were also studied. The result indicates a correlation between the cell spreading and migration on nanodiamond and the surface energy of nanostructured surface.

Effects of additions of trace Sn on the bake-hardening responses of a pre-aged Al-0.85 Mg-0.85 Si(in wt%)alloy were investigated through mechanical tests,differential scanning calorimetry,electrical resistivity and transmission electron *** indicate that trace Sn additions reduced the number density of pre-aging clusters by inhibiting the formation of unstable counterpart during pre-aging treatment,leading to low strength and high supersaturation of solute *** a subsequent paint-bake treatment,the presence of highly supersaturated solute atoms and high concentrated free vacancies moderated the activation energy barrier ofβ’’phase and thus kinetically accelerated the formation ofβ’’.Consequently,the trace Sn additions enhanced the bake-hardening responses of the pre-aged alloys *** Sn-containing pre-aged Al-Mg-Si alloys with low strength and great bake-hardening responses hold promising potential for automotive body skin applications.

Dynamic responses of K-type and inverted-K-type jacket support structures for offshore wind-turbines

The jacket structure has become more popular as the offshore wind-turbine support structure. K-type and inverted-K-type jacket support structures have superior potential due to their fewer joints and lower cost of manufacture and installation. A numerical study was presented on the dynamic responses of K-type and inverted-K-type jacket support structures subjected to different kinds of dynamic load. The results show that the inverted-K-type jacket structure has higher natural frequencies than the K-type. The wave force spectrum response shows that the maximum displacement of the K-type jacket structure is larger than that of the inverted-K-type. The time-history responses under wind and wave-current load indicate that the inverted-K-type jacket structure shows smaller displacement and stress compared with the K-type, and presents different stress concentration phenomena. The dynamic responses reveal that the inverted-K-type of jacket support structure has greater stiffness and superior mechanical properties, and thus is more applicable in the offshore area with relatively deep water.

Common mycorrhizal networks activate salicylic acid defense responses of trifoliate orange(Poncirus trifoliata)

Citrus canker, caused by Xanthomonas axonopo-dis pv. citri ('Xac'), is an important quarantine disease in citrus crops. Arbuscular mycorrhizal fungi (AMF) form symbiotic interactions with host plants and further affect their disease resistance, possibly by modulating the activity of salicylic acid (SA), a key phytohormone in disease resistance. Common mycorrhizal networks (CMNs) can interconnect plants, but it is not yet clear whether CMNs promote resistance to citrus canker and, if so, whether SA signaling is involved in this process. To test this possibility, we used a two-chambered rootbox to establish CMNs between trifoliate orange (Poncirus trifoliata) seedlings in chambers inoculated (treated) or not (neighboring) with the AMF, Paraglomus occultum. A subset of the AMF-inoculated seedlings were also inoculated with Xac (+AMF+Xac). At 2 d post-inoculation (dpi), compared with the +AMF-Xac treatment, neighboring seedlings in +AMF+Xac treatment had lower expression levels of the SA biosynthetic genes, PtPAL, PtEPS1, and PtPBS3, but higher SA levels, which attributed to the upregulation of PtPAL and PtPBS3 in treated seedlings and the transfer of SA, via CMNs, to the neighboring seedlings. At 4 dpi, the pathogenesis-related (PR) protein genes, PtPR1, PtPR4, and PtPR5, and the transcriptional regulatory factor gene, PtNPR1, were activated in neighboring seedlings of+AMF+Xac treatment. At 9 dpi, root phenylalanine ammo-nia-lyase activity and total soluble phenol and lignin concentrations increased in neighboring seedlings of+AMF+Xac treatment, likely due to the linkage and signal transfer, via CMNs. These findings support the hypothesis that CMNs transfer the SA signal from infected to neighboring healthy seedlings, to activate defense responses and affording protection to neighboring plants against citrus canker infection.

Bark beetles Tomicus yunnanensis and *** are two important pests of Pinus yunnanensis and can cause massive death of pine *** this study,we examined several traits related to photosynthesis in *** and their relationship with antibiotic defense responses after joint attack by the two bark beetles at the shoot and the trunk *** shoots were attacked by the beetles,the abundance of chlorophylls,carotenoids,and the rates of net photosynthesis(Pn)and transpiration(E)decreased in needles,while the levels of superoxide dismutase and malondialdehyde remained unchanged in both needles and *** activity of peroxidases also remained unchanged in needles,but increased in *** activity of catalases increased in both needles and *** trunks were attacked by the bark beetles,chlorophyll abundance,Pn,E,and antioxidative enzyme activities all declined,and the declines were more pronounced than in the attacked shoots.A decrease in protein concentrations was also observed in needles and phloem from the attacked *** on shoots by the bark beetles suppressed host defense and provided a favorable environment for larval growth and development,resulting in long-term decline of pine growth *** results suggest that attacks on trunks by beetles caused more severe damage to host trees than attacks on shoots.

Effect of heavy load carriage on cardiorespiratory responses with varying gradients and modes of carriage

Background: The present study was undertaken to determine the effect of different uphill and downhill gradients on cardiorespiratory and metabolic responses of soldiers while carrying heavy military loads in two different ***: Eight physically fit male soldiers with a mean age 32.0±2.0 years, a mean height of 169.5±4.9 cm, and a mean weight of 63.8±8.4 kg volunteered for this study. Each volunteer completed treadmill walking trials at a speed of 3.5 km/h while carrying no external load, 31.4 kg load in a distributed mode(existing load carriage ensembles) and compact mode(new back pack) over 5 different downhill and uphill gradients(–5%, –10%, 0%, 5%, 10%) for 6 min at each gradient. During the walking trials, heart rate(HR), oxygen uptake(VO_2), respiratory frequency(RF) and energy expenditure(EE) were determined by the process of breath-by-breath gas analysis using a K4 b2 system. The average of the last 2 min data from each 6 min walking trial for each individual was subjected to statistical ***: All parameters(HR, VO_2, RF, and EE) gradually increased with the change in gradient from downhill to level to uphill. The distributed mode showed higher values compared to compact mode for all gradients, e.g., for VO_2, there was a 10.7, 7.4, 5.1, 28.2 and 18.7% increase in the distributed mode across the 5 different ***: It can be concluded from the present study that the compact mode of load carriage is more beneficial than the distributed mode in terms of cardiorespiratory responses while walking on downhill and uphill surfaces with a 31.4 kg load.

PXR variants:the impact on drug metabolism and therapeutic responses

The pregnane X receptor(PXR) plays an important and diverse role in mediating xenobiotic induction of drug-metabolizing enzymes and *** protein isoforms of PXR exist,and they have differential transcriptional activity upon target genes; transcript variants 3(PXR3) and 4(PXR4) do not induce target gene expression,whereas transcript variants 1(PXR1) and 2(PXR2) respond to agonist by activating target gene *** protein variants also display differences in protein–protein interactions; PXR1 interacts with p53,whereas PXR3 does ***,the transcript variants of PXR that encode these protein isoforms are differentially regulated by methylation and deletions in the respective promoters of the variants,and their expression differs in various human cancers and also in cancerous tissue compared to adjacent normal ***1 and PXR4 m RNA are downregulated by methylation in cancerous tissue and have divergent effects on cellular proliferation when ectopically *** detailed and comparative mechanistic studies are required to predict the effect of PXR transcript variant expression on carcinogenesis,therapeutic response,and the development of toxicity.

Effects of shear keys on nonlinear seismic responses of an arch-gravity dam

In the paper,the added tangential nonlinear springs are introduced to represent the effects of one-way and two-way shear keys. The added tangential stiffness coefficient is relative to the joint opening and the relative tangential movement and determined numerically by using a refined model of shear *** user subroutine of nonlinear spring elements has been implemented into ABAQUS *** seismic responses of an arch-gravity dam are mainly *** is shown by the results that a significant increase of about 17%-30%in joint opening can be observed for the dam with one-way shear keys;the two-way shear keys are beneficial to restrain the sliding displacement along both the radial and vertical directions,whereas the one-way shear keys mainly restrain the displacement along the radial direction;for the dam with shear keys,the maximum arch stress is increased,the cantilever stress is reduced,and the location of the maximum cantilever stress is also moved from the dam heel to the midheight of the cantilever.
