Searching for Potential Drug Targets in Two-component and Phosphorelay Signal-transduction Systems using Three-dimensional Cluster Analysis

Two-component signaling pathways include histidine kinases,response regulators,and sometimes histidine-containing phosphotransfer proteins.A response regulator protein of two-component system,skn7-homologous gene was identified in the insect pathogenic fungus Metarhizium robertsii and designated as *** study its potential function,the Mrskn7 gene deletion mutants were *** normal growth conditions(*** dextrose agar),the mutant lost the ability to sporulate and demonstrated hyphal autolysis at late growth *** the salt(KCl or NaCl) stresses,the mutant was partially rescued to produce spores,but the virulence was significantly decreased when compared to the wild *** expression analysis by real-time PCR showed that the expressions of autolysis-related genes like NagA,chitinase and protease genes were significantly up-regulated in △*** wall staining results revealed a serious defect in cell wall composition in Mrskn7 *** one-hybrid tests confirmed that Mrskn7 could function as a transcription factor to control the expression of genes involved in regulating cell wall biosynthesis,cell autolysis and sporulation in ***.

Two-component and phosphorelay signal transduction systems are central components in thevirulence and antimicrobial resistance responses of a number of bacterial and fungal pathogens;in somecases,these systems are essential for bacterial growth and ***,we analyze in detail the conservedsurface residue clusters in the phosphotransferase domain of histidine kinases and the regulatory domain ofresponse regulators by using complex structure-based three-dimensional cluster *** also investigatethe protein-protein interactions that these residue clusters participate *** Spo0B-Spo0F complex structurewas used as the reference structure,and the multiple aligned sequences of phosphotransferases and responseregulators were paired *** results show that a contiguous conserved residue cluster isformed around the active site,which crosses the interface of histidine kinases and response *** residue clusters of phosphotransferase and the regulatory domains are directly involved in thefunctional implementation of two-component signal transduction systems and are good targets for thedevelopment of novel antimicrobial agents.

Deletion of the Initial 45 Residues of ARR18 Induces Cytokinin response in Arabidopsis

The plant hormone cytokinin plays important roles in various aspects of plant growth and development. Cytokinin signaling is mediated by a multistep phosphorelay similar to bacterial two-component system. Type-B ARRs lie at the end of the cytokinin signaling, typically mediating tile output response. However, it is still unclear how type-B ARRs are regulated in response to cytokinin. Typical type-B ARR contains an N-terminal receiver domain and a C-terminal effector domain. In this study, we performed a genome-wild comparative analysis by overexpressing full length and C-terminal effector domain of seven representative type-B ARRs. Our results indicated that overexpression of C-terminal effector domain causes short primary roots and short hypocotyls without the addition of cytokinin, sug- gesting that the inhibitory role of the receiver domain in the activity of the effector domain is a common mechanism in type-B ARRs. To investigate how the receiver domain inhibits the activity of the effector domain, we performed a deletion analysis. We found that deletion of the initial 45 residues of ARR18 （the 45 residues from N-terminus） causes pleiotropic growth defects by directly inducing cytokinin responsive genes. Together, our results suggest that the initial 45 residues are critical for the inhibitory role of the receiver domain to the effector domain in ARRI8.

The multistep phosphorelay （MSP） is a central signaling pathway in plants integrating a wide spectrum of hormonal and environmental inputs and controlling numerous developmental adaptations. For the thor- ough comprehension of the molecular mechanisms underlying the MSP-mediated signal recognition and transduction, the detailed structural characterization of individual members of the pathway is critical. In this review we describe and discuss the recently known crystal and nuclear magnetic resonance structures of proteins acting in MSP signaling in higher plants, focusing particularly on cytokinin and ethylene signaling in Arabidopsis thaliana. We discuss the range of functional aspects of available structural infor- mation including determination of ligand specificity, activation of the receptor via its autophosphorylaUon, and downstream signal transduction through the phosphorelay. We compare the plant structures with their bacterial counterparts and show that although the overall similarity is high, the differences in structural de- tails are frequent and functionally important. Finally, we discuss emerging knowledge on molecular recog- nition mechanisms in the MSP, and mention the latest findings regarding structural determinants of signaling specificity in the Arabidopsis MSP that could serve as a general model of this pathway in all higher plants.

Due to the selective permeability of the cytomembrane,high-yield fumaric acid strains form a steep difference between intra-and extracellular *** biosensors cannot detect the real concentration change of extracellular fumaric *** overcome this limitation,a two-component biosensor(TCB)that could respond to extracellular fumaric acid was designed based on the DcuS-DcuR two-component *** two-component system consists of a histidine kinase(SK)and response *** is a transmembrane histidine kinase sensor that can detect concentration changes in extracellular *** improve the dynamic range of the constructed fumaric acid TCB,we optimized the expression ratio and expression intensity of dcuS and *** found that the optimum expression ratio of dcuS:dcuR was 46:*** this ratio,the higher was the expression level,the greater the dynamic *** addition,we modified the ATP-binding site on the DcuS,and the final dynamic range of the TCB reached ***,the obtained fumaric acid-responsive TCB with a high dynamic range is reported for the first time,providing a synthetic biology tool for high-throughput screening and dynamic metabolic regulation of fumaric acid cell factories.

Seed weight is usually associated with seed size and is one of the important agronomic traits that determine *** of seed weight control is limited,especially in soybean *** we show that Glycine max JASMONATEZIM DOMAIN 3(GmJAZ3),a gene identified through gene co-expression network analysis,regulates seed-related traits in *** of GmJAZ3 promotes seed size/weight and other organ sizes in stable transgenic soybean plants likely by increasing cell ***3 interacted with both *** response regulator 18a(GmRR18a)and GmMYC2a to inhibit their transcriptional activation of cytokinin oxidase gene *** CYTOKININ OXIDASE 3-4(GmCKX3-4),which usually affects seed ***,the GmRR18a binds to the promoter of GmMYC2a and activates GmMYC2a gene *** GmJAZ3-overexpressing soybean seeds,the protein contents were increased while the fatty acid contents were reduced compared to those in the control seeds,indicating that the GmJAZ3 affects seed size/weight and *** variation in JAZ3 promoter region was further analyzed and Hap3 promoter correlates with higher promoter activity,higher gene expression and higher seed *** Hap3 promoter may be selected and fixed during soybean ***3 orthologs from other plants/crops may also control seed size and *** together,our study reveals a novel molecular module GmJAZ3-GmRR18a/GmMYC2a-GmCKXs for seed size and weight control,providing promising targets during soybean molecular breeding for better seed traits.
