Integrity of both nuclear and mitochondrial DNA is vital for the survival of an organism. Exposure to genotoxic agents has the potential to damage DNA leading to significant health ***,the ability to detect DNA damage is critical to understanding the health of an *** methods for detecting DNA damage are limited by the amount of DNA and time required to complete an assay,and low *** study presents a novel,fast and precise real-time extra long-pcr(xl-pcr) assay for DNA damage *** introduced SYTO-82 to TaKaRa LA Taq hot start system as the fluorescence *** real time,xl-pcr system successfully quantified three xl fragments of both mtDNA and nuclear DNA from sea lamprey: mtDNA fragment LD-U11880(15030 bp,E=1.63,R=0.9998);nuclear DNA fragments SL-Contig452(11924 bp,E=1.62,R=0.9936) and SL-X14061(10547 bp,E=1.64,R=0.9985). Results of in vitro UV exposure indicated that mtDNA was a more sensitive detection target than the other two nuclear DNA fragments.A pcr efficiency-based,real-time xl-pcr data analysis method that can accurately assess DNA damage is also detailed *** will use this method to detect DNA damage in chemo-sterilized sea lampreys used as a quality control for the sterile-male-release technique(SMRT),an important sea lamprey control method applied in the Great Lakes of North America.
