Aim:The aim of the present study was to investigate the inhibitory effect ofpseudolaric acid b （PAb） on human breast cancer MCF-7 ***:3-（4,5-dimethylthiazol-2-yl）-2,5-diphenyltetrazolium bromide analysis,morphologicalchanges,acridine orange staining,and agarose gel electrophoresis were appliedto detect *** percentage of apoptotic and necrotic cells was calcu-lated by the lactate dehydrogenase activity-based cytotoxicity assay;senescenceassociated （SA）-β-galactosidase activity was detected to evaluate senescence;flow cytometric analysis of propidium iodide staining was carried out to investi-gate the distribution of cell cycle,and the protein expression was examined byWestern blot ***:During apoptosis,the half maximal inhibitoryconcentration IC50was 3.4 and 1.35 μmol/L at 36 and 48 h after PAb treatment,*** MCF-7 cells exposed to PAb showed typical characteristics ofapoptosis,including the morphological changes and DNA ***-7 cells treated with 4 μmol/L PAb for 36 h underwent apoptosis,but *** apoptosis induced by PAb was independent of the death *** senescent cells became larger and flatter,and the SA-β-galactosi-dase staining was *** induced obvious mitotic arrest and it precededapoptosis and *** expressions of p21 and p53 was upregulated withPAb treatment,and cyclin b 1 was upregulated and transported from the cyto-plasm to nuclei,and sustained stable ***:PAb induced mitoticarrest in the MCF-7 cells and inhibited proliferation through apoptosis *** apoptosis was independent of the death receptor pathway.

Objective:pseudolaric acid b(Pb)is a newly identified diterpenoid isolated from Tujinpi(Cortex pseudolaricis).In the present study,we aimed to explore the anti-inflammatory effects of Pb on atopic dermatitis(AD),as well as the molecular mechanisms underlying its ***:bALb/c mice treated with 2,4-dinitrofluorobenzene were orally administered with Pb(10 mg?kg-1?d-1).After evaluating the AD score,serum levels of IgE and the mRNA expression of NLRP3 inflammasome and IL-1βwere measured by ELISA and qRT-PCR ***:The results showed that Pb treatment significantly ameliorated the development of AD-like clinical symptoms and effectively suppressed the infiltration of inflammatory ***,Pb inhibited the expression of NLRP3 inflammasome and IL-1βin skin lesions,and downregulated serum IgE ***:The anti-inflammatory properties of Pb were demonstrated using the 2,4-dinitrofluorobenzene-induced mouse model of AD-like skin *** study highlighted the potential use of Pb as a novel therapeutic agent for the treatment of inflammation-associated skin diseases.

Effect of pseudolaric acid b on gastric cancer cells: Inhibition of proliferation and induction of apoptosis

AIM: To examine the effect of pseudolaric acid b on the growth of human gastric cancer cell line, AGS, and its possible mechanism of ***: Growth inhibition by pseudolaric acid b was analyzed using MTT assay. Apoptotic cells were detected using Hoechst 33258 staining, and confirmed by DNA fragmentation analysis. Western blot was used to detect the expression of apoptosis-regulated gene bcl-2, caspase 3, and cleavage of poly (ADP-ribose)polymerase-1 (PARP-1).RESULTS: pseudolaric acid b inhibited the growth of AGS cells in a time- and dose-dependent manner by arresting the cells at G2/M phase, which was accompanied with a decrease in the levels of *** cells treated with pseudolaric acid b showed typical characteristics of apoptosis including chromatin condensation and DNA fragmentation. Moreover,treatment of AGS cells with pseudolaric acid b was also associated with decreased levels of the anti-apoptotic protein bcl-2, activation of caspase-3, and proteolytic cleavage of ***: pseudolaric acid b can dramatically suppress the AGS cell growth by inducing apoptosis after G2/M phase arrest. These findings are consistent with the possibility that G2/M phase arrest is mediated by the down-regulation of cdc2 levels. The data also suggest that pseudolaric acid b can trigger apoptosis by decreasing bcl-2 levels and activating caspase-3 protease.

Considering that extensive immunosuppressive agents have adverse reaction on normal cells to restrain their clinical application, researchers are devoting to find some novel agents with less side-effects and more security from Traditional Chinese Medicine(TCM). Cortex pseudolaricis, a kind of traditional Chinese medicine, has been used to treat tinea or other skin diseases since long time ago. pseudolaric acid b(PAb) is the main effective diterpenoid of Cortex pseudolaricis. According to previous studies, PAb has been proved to markedly inhibit the growth and proliferation of many tumor cell lines by inducing mitotic catastrophe and apoptosis, and affect cell cycle, which makes it as a novel microtubule inhibitor. Moreover, it has been reported to exert favorable anti-inflammatory and immunosuppressive activities with less *** analyzing the correlational studies in recent decades, we have made a review on PAb and its derivatives, espe-cially the research development of its pharmacological *** though the experimental utility of PAb is prevailing accepted, the mechanism is not completely clear so that we have a long way to explore the potential mechanisms of PAb against tumor and immune-related diseases.
