Phosphorus(P)is involved in various biochemical reactions in plant growth,so it is beneficial to plants growing in soils contaminated by metals,including cadmium(Cd).However,few studies have reported on the mechanistic roles of P in mitigating Cd toxicity to ryegrass root,and especially in alleviating the disruption of the mitochondrial function of living cells.In this study,the physiological and biochemical mechanisms associated with ryegrass growth under various Cd and P treatments were investigated using leaching and pot systems.The concentration of Cd in soil leachates showed a significantly positive relationship with redox potential(Pprotoplast.Moreover,exogenous P was found to play a positive role in mitochondrial function and Ca^(2+)variation in root cells under Cd stress,which provides novel insights into the mechanisms of exogenous P in alleviating plant Cd injury.

In this paper, we have utilized the principle of dielectrophoresis to determine the dielectrophoretic spectra of single levitated tobacco leaf protoplast at various suspending media conductivities (12.5 μS/cm-520 μS/cm) over wide frequencies (10 Hz—15 MHz). From the experimental data, the membrane capacitance of the single protoplast which was 0.40±0.03μF/cm^2 has been obtained and the dipole interaction of two protoplasts in electrofusion has also been analysed. When the external electric field E_c=1.5kV/cm was applied and the frequency was higher than 20 kHz, the dipole interaction pressure ~πd would be much larger than the membrane potential pressure ~πm, and the pressure ~πd could be the main reason for membraue pores. This conclusion will provide a new basis to improve the protocol of cell electrofusion.

Embryogenic calli were induced from the mature seeds of the hexaploid semi-winter wheat. The embryogenic cell line was established, and then the friable calli suitable for suspension were induced by adjusting the content of reduced N and changing the 2,4DAA concentration. protoplasts were isolated from the suspension cells and cultured(?)in KM8P and some other media. A great number of microcolonies were obtained. By altering media, the microcolonies were promoted to grow further and form compact or nodule-like calli. Intact plants were regenerated through organogenesis and embryogenesis on differentiation media.

protoplasts were isolated from embryogenic calli derived from immature embryos ofwheat (Triticum aestivum L. cv. Jinan 177). The protoplasts were cultured in NMB mediumsupplemented with 1mg/L 2,4- D and 500mg/l casein hydrolysate (CH). The regenerated cellsfrom protoplasts divided to form somatic embryos directly. The somatic embryos grown to1.5- 2 mm in size directly developed into complete plants on solid MB medium without hor-mones.

Compact calli derived from immature spikelet of a foxtail millet variety—Jigu 11cann’t be directly used for protoplast isolation because of its firm physical structure,and must beloosened with subculturing in M1,M2 and M3 media successively and altering these media compo-sitions.The loosened calli can be selected from the regulation and used for protoplast isolationsuccessfully.Rate of protoplast division in KM8P medium was 12.3—33.5%.Calli derivedthrough protoplast division are loose and cann’t be used directly for plan regeneration because ofits soft physical structure.When they were subcultured in N6—1,N6—2,N6—3 and N6—4 media,in which the media compositions were changed,the compact calli were obtained and 129 plantletswere regenerated from them.101 plants,which grew to maturity after transplanting the plantletsinto field,exhibited sterility in some degree.Most of the subsequent lines derived from the regen-erated plants were sterile and only two lines could get normal reproduction.

3´,5´-Cyclic adenosine monophosphate(cAMP)is an important metabolite that is specifically enriched in jujube.However,the effect of cAMP on jujube cellular responses has not been comprehensively studied.Here,we established jujube cell suspension cultures and investigated the calcium influx in response to cAMP treatment through protoplast isolation and fluorescence intensity.Firstly,cAMP treatment could promote jujube growth and increase the content of endogenous cAMP.Using transcriptome analysis with transgenic Arabidopsis plants overexpressing adenylate cyclase(ZjAC)as a positive control,we identified 60 calcium-related differential expressed genes(DEGs)that contributed to the calcium signaling and inter-or intra-cellular responses.Pharmacological treatments such as cAMP and the calcium ionophore A23187 could induce ZjAC expression,the accumulation of cAMP and calcium influx in jujube cells,while ethylene glycol tetraacetic acid(EGTA)or bithionol treatment inhibited these changes.Moreover,the calcium channels and transporters in calcium influx,such as the ZjCNGC2 channel and the mitogen activated protein(MAP)kinase pathway,could be activated by cAMP treatment.In summary,our findings demonstrated that cAMP biosynthesis is dependent on calcium influx and the amplifying effect between calcium and cAMP may be involved in intracellular signal induction,which might contribute to the growth and development of jujube.

[Objectives] This study was conducted to improve the genetic character of Epipremnum aureum and provide a new way for genetic breeding of E. aureum. [Methods] In the paper,protoplasts were isolated from tender leaves of E. aureum and Scindapsus aureus var. wilcoxii by using 1% cellulase and 1% peetinase. Then the protoplasts were purified and dynamically identified,and fusion of protoplasts of E. aureum and S. aureus was induced by PEG. [Results]A large quantity of protoplasts were obtained from tender leaves of E. aureum and S. aureus successfully,and the activity of protoplasts was over 90%. Fusion product was obtained by PEG induction,and the fusion rate was 16. 3%. [Conclusions]protoplasts were isolated from tender leaves of E. aureum and S. aureus successfully,and a stable fusion system was built by PEG induction.

Laser tweezers can manipulate small particles, such as cells and organdies. When coupling them with laser microbeam selective fusion of two tobacco protoplasts containing some chloroplast was achieved. Physical and biological variables that affect laser trapping and laser-induced fusion were also discussed. The results show that the effect of chloroplast content and distribution on the yield of cell fusion is remarkable.
