Intelectins are a recently identified extracellular animal lectin as a glycan-binding receptors that recognize glycan epitopes on foreign pathogens and in the host *** present study reports a new type of serum lectin found in lamprey,Lampetra *** proteing agarose mixed with serum from lamprey was analyzed by SDS-PAGE,the lectin appeared as a single 35kD band.A 1005bp cDNA sequence obtained by RT-PCR from liver encoded a 335 amino acid secretory protein with homology to mammalian and aquatic organism *** expression and western blot demonstrated that the intelectin gene was expressed in the blood, intestine,kidney,heart,gill,liver,medullary,***-NPGBP exhibited calcium-dependent agglutination to human red blood *** the purified NPGBP from chromatography with proteing-conjugated agarose was analyzed by non-reducing SDS-PAGE,the intelectin appeared as～34kD,and～124kD bands,suggested that the intelectin forms monomers and *** gene expression was induced by in vivo bacterial stimulation,and the NPGBP showed agglutination activity against the pathogenic Gram-positive and Gram-negative bacterium,suggesting that the NPGBP plays an important role in self-defense against bacteria in the internal circulation system of *** adding different antibody,the specific binding between NPGBP and proteing is competitive inhibited,these shows that the binding sites of NPGBP with proteing is the same with Fc section of Ig,and hypothesized that NPGBP shared similar spatial structure with Fc section of Ig. Also,NPGBP can effectively promote the endothelial cells *** finding suggest that NPGBP plays a role in immune defense against bacterial and represent one of the few examples of characterization and functional analysis of an aquatic organisms intelectin protein.
