The role of protein kinase c （PKc） activation in advanced glycation end products （AGEs）-induced epithelial-mesenchymal transition in renal proximal tubular epithelial cells was investigated. HKc cells were divided into three groups： normal group, AGE-BSA group （100 mg/L AGE-BSA） and AGE-BSA＋PKc inhibitor （10 μmol/L chelerythrine chloride） group. PKc activity was measured by PKc assay kit. The expression of Vimentin, and phosphorylated β-catenin was detected by using Western blotting, and the content of TGF-β1 was examined by ELISA method. The intracellular disposition of Vimentin was observed by fluorescence microscopy. As compared with normal group, PKc activity was increased significantly in AGE-BSA group. The expression of Vimentin, phosphorylated β-catenin, and TGF-β1 was enhanced significantly in AGE-BSA group. The expression of Vimentin, phosphorylated β-catenin, and TGF-β1 was significantly blocked by chelerythrine chloride. High expression of Vimentin, phosphorylated β-catenin, and TGF-β1 induced by AGE-BSA may be mediated via the activation of PKc signal transduction pathway.

This study examined the effect of GHRP-6, a known GHSs receptor agonist, on the phosphorylation of cAMP-responsive element-binding protein （cREB） and the tmderly mechanism. GH3 cells were cultured and subjected to different treatments as follows： GHRP-6, GHRP-6 plus GHRH, phorbol ester （PMA）, an activator of PKc, alone or in combination with GHRP-6, G66983, a general inhibitor of PKcs, in the presence or absence of GHRP-6, rottlerin, an inhibitor of PKcs, alone or plus GHRP-6. The cells were transiently transfected with PKcσ-specific siRNA and then treated with GHRP-6. GH level was measured by enzyme-linked immunosorbent assay （ELISA）. The expression of phosphor-cREB, PKcσ, PKc0 and phosphor-PKco was determined by Western blotting. The results showed that GHRP-6 stimulated GH secretion in both time- and dose-dependent manners and enhanced the effect of GHRH on GH secretion. GHRP-6 was also found to induce cREB phosphorylation. Moreover, GH secretion was enhanced by the PKc activator PMA and reduced by the PKc inhibitors （G66983, rottlerin） and knockdown of PKcσ. PKcσ could be activated by GHRP-6. It is concluded that PKc, especialiy PKcσ, mediates cREB phosphorylation and GHRP-6-induced GH secretion.

In the present study, we investigated the activation of protein kinase c （PKc） family in mouse embryonic fibroblast NIH3T3 cells using gadolinium chloride as a representative lanthanide ion. With live cell imaging system and confocal laser scanning microscopy, we found that the treatment of 50 μM GdcI3 promoted cell survival under the condition of serum-starvation. Moreover, better cell attachment and cytoskeleton reorganization were also observed. Additionally, Gdc13 treatment resulted in the phosphorylation of PKc family at different time points. Furthermore, bisindolylmaleimide （a PKcpan inhibitor） could efficiently reduce the level of phosphorylated PKcpan （βIISer660）, alleviating ERK activation induced by Gdc13. This finding indicated that the PKc activation was involved in Gdc13-induced MAPK/ERK signaling and thus might contribute to Gdclβ-indueed cell cycle progression and cell survival.

protein kinase c with a molecular weight of 82 kD has been purified to electrophoresis homogenous from rat brain through a series of chromatography cohmns including DE-52, Sepharose G-200 and phenyl-Sepharose. The enzyme possessed autophosphorylation activity. Yuanhuacin A inhibited theH-phorbol-12, 13-dibutyrate （H-PdBu） binding of PKc with an Icvalue of 1.48±0.287×10mol/L when the concentration ofH-PdBu was 1.5×10mol/L (K=1.2×10mol/L). Yuanhuacin A inhibited the PdBu-stimulated PKc activity in the catalysis of the phosphorylation of Histone Ⅲ-S with an Icof 2.82±0.37×10mol/L (PdBu=10mol/L), while it had no effect on the basal and ca-stimulated PKc activity in the same assay system. This result suggests that Yuanhuacin A is a selective antagonist of the phorbol ester receptor in protein kinase c.

protein kinase c (PKc) is a superfamily of Ser/Thr protein kinases that is distributed widely in eukaryotes. It plays key regulatory roles at multiple steps of oocyte meiotic maturation and fertilization. During the process of meiotic maturation, the activation of PKc in cumulus cells stimulates meiotic maturation, whereas the activation of PKc in oocytes results in the inhibition of germinal vesicle breakdown. PKc activity increases following the meiotic maturation, and decreases at the transition of metaphase/anaphase in meiosis Ⅰ, so as to facilitate the release of the first polar body and the entry of meiosis Ⅱ. In fertilization of mammalian oocytes, PKc may act as one of the downstream targets of ca 2+ to stimulate the cortical granule exocytosis, release the oocytes from Mil arrest and to induce pronucleus formation. To explore the role of nuclear factor-κB(NF-κB) in the signal pathway of protein kinase c (PKc) regulating the proliferation and apoptosis of T lymphocytes in asthma. T lymphocytes were isolated from the asthmatic model of guinea pigs and the asthmatic patients. Either the T cells stimulated with PMA alone or those stimulated with PMA together with pyrrolidine dithiocarbamate (PDTc) were incubated for 1 and 24?h. The proliferation of and the presence of NF-κB in the cells incubated for 1?h were observed by MTT and immunohistochemical staining, respectively. And the cells incubated for 24?h were observed for the apoptosis by TUNEL. All the assays were paralleled with controls, and all the data were analyzedstatistically with the software SAS. Summary: In order to explore the role of activator protein-1 (AP-1) in the transcription of interleukin-5 (IL-5) gene regulated by protein kinase c (PKc) signal in peripheral blood T lymphocytes from asthmatic patient, T lymphocytes were isolated and purified from peripheral blood of each asthmatic patient. The T lymphocytes were randomly divided into 4 groups: group A (blank control), group B (treated with PKc agonist phorbol 12-myristate 13-acetate (PMA)), Group c (treated with PMA and AP-1 cis-element decoy oligodeoxynucleotides (decoy ODNs)), and group D (treated with PMA and AP-1 mutant decoy ODNs). The ODNs were transfected into the T cells of group c and D by cation liposome respectively. Reverse transcription-polymerase chain reaction (RT-PcR) was employed to assess IL-5 mRNA expression, and electrophoretic mobility shift assays (EMSA) for the activation of AP-1. The results showed that the activation of AP-1 (88 003.58±1 626.57) and the expression of IL-5 mRNA (0.8300±0.0294) in T lymphocytes stimulated with PMA were significantly higher than these in blank control (20 888.47±1103.56 and 0.3050±0.0208, respectively, Pctively) of T lymphocytes stimulated with PMA and AP-1 decoy ODNs were significantly inhibited, as compared with group B (Pctively) in T lymphocytes stimulated with PMA and AP-1 mutant decoy ODNs did not exhibit significant changes, as compared with group B (P>0.05). The significant positive correlation was found between the activation of AP-1 and the expression of IL-5 mRNA (Pconcluded that AP-1 might participate in the signal transduction of PKc-triggered transcription of IL-5 gene in asthmatic T lymphocytes. This suggests the activation of PKc/AP-1 signal transduction cascade of T lymphocytes may play an important role in the pathogenesis of asthma.

To investigate the effect of cigarette smoke extract (cSE) on the role of protein kinase c (PKc) in the proliferation of passively sensitized human airway smooth muscle cells (HASMcs). After synchronization of cultured HASMcs, they were divided into a group A and Group B. The group A was treated with normal human serum and served as controls and the group B was treated with the serum of asthma patients. The group A was further divided into group of A_1, A_2 and A_3 and the group B was sub-divided into the group of B_1, B_2, B_3, B_4 and B_5. No other agents were added to the group A_1 and B_1. The cells of group A_2 and B_2 were stimulated with 5 % cSE for 24 h. HASMcs from group A_3 and B_3 were treated with PKc agonist PMA (10 nmol/L) and cSE (5 %) for 24 h. PKc inhibitor Ro-31-8220 (5 μmol/L) was added to the HASMcs of group B_4 for 24 h. The cells from group B_5 were stimulated with Ro-31-8220 (5 μmol/L) and cSE (5 %) for 24 h. The proliferation of HASMcs isolated from group A and B was examined by cell cycle analysis, MTT colorimetric assay and 3H-TdR incorporation test. The expression of PKc-α in each group was observed by Western blotting and RT-PcR, respectively. The results showed that the percentage of S phase, absorbance (A) value, the rate of 3H-TdR incorporation, the ratios of A value of PKc-α mRNA and the A value of PKc-α protein in HASMcs from group B_1, B_2 and B_3 were significantly increased compared to those of group A_1, A_2 and A_3 correspondingly and respectively (Pcs of group A_2 and B_2 stimulated with cSE and group A_3 and B_3 stimulated with cSE and PMA were also significantly enhanced when group A_1, A_2 and A_3 and group B_1, B_2 and B_3 compared to each other (Pctively).
