The aim of this study was to estimate the inhibitory effects of the flavonoid glycosides on the four pathways in diabetic complication, and find a new and effective extract of chinese herbs for treatment of diabetic *** used vitro selecting model to investigate the inhibitory effects of the tested *** was shown, *** and Breviscapine have different degrees of inhibitory effects on aldose reductase, protein kinase c, advanced glycation end-products and malondialdehyde (MDA). It hinted that the two chinese herbal extracts may prevent diabetic complication by inhibiting four pathways.2Silybin has different degrees of inhibitory effects on protein kinase c, advanced glycation end products and MDA. It hinted that Silybin may be effectively to prevencet diabetic complication.3. Puerarin has different degrees of inhibitory effects on aldose redactase and advanced glycation end-products.

＜正＞Hypoxia-induced inhibition of Qo and Qi site of mitochondrial complex Ⅲunder hypoxia has received attention, but its downstream pathways remain unclear. In this paper, we used the Qo site inhibitor myxothiazol and the Qi site,inhibitor antimycin A to mimick the inhibition of the Qo and Qi site of mitochondrial complex Ⅲand studied the effects of the inhibition of these site on persistent sodium currents, transient sodium currents and neuron excitability in rat hippocampal cA1 cells with whole cell patch-clamp methods combined biochemical and pharmacological approaches. In pilocarpine induced status epilepticus(PISE) in metabotropic glutamate receptor 5(mGluR5) knock-out (mGluR5) and wild-type(mGluR5) mice,we showed that at 2 h during PISE,induced PKcbeta2 or PKcgamma immunopositive product was demonstrated at the border between the stratum oriens and alveus(O/A border) of cA1 area in mGluR5 ***,much less staining was observed in mGluR5 *** 1d after PISE,induced PLcbeta4, PKcbeta2 or PKcgamma in stratum pyramidale of c A1 area in mGluR5 but not mGluR5 mice was *** labeling showed co-localization of induced PKcbeta2 or PKcgamma with mGluR5 or induced PLcbeta4 in the stratum pyramidale of cA1 *** therefore suggests that induced PLcbeta4-PKcbeta2/PKcgamma at 1 d after PISE in pyramidal neurons or induced PKcbeta2 or PKcgamma at O/A border at 2 h during PISE may be involved in mGluR5-related signal transduction leading to delayed loss of pyramidal neurons in mGluR5 *** both mGluR5 and mGluR5 mice,significant reduction of PKcdelta and PKceta immunopositive interneurons in cAl area at early stages during PISE,and up-regulation of PKcalpha and PKcbetal in cA3b area at 1 d after PISE and of PKcepsilon in stratum lucidum of cA3 area at 1 week after PISE,and progressive reduction of PKcgamma in inner moleculare of dentate gyrus suggest that they may be related to the development of temporal lobe *** 2 months after PISE,induced PLcbeta4,PKcbeta2,PKcdelta,PKceta,PKczeta in glial cells in gliotic hippocampus may be linked to the occurrence of spontaneously recurrent seizures.

Our previous work has shown that lymphocytes synthesize catecholamines(cAs) and the lymphocytederived cAs accelerate lymphocyte ***,the mechanism underlying the cAs-accelerated lymphocyte apoptosis is not ***,we explored the involvement of adrenoreceptors(ARs) and types of second messengers coupled to ARs in the endogenous cAs-mediated modulation of lymphocyte *** of cleaved caspase-3 and number of apoptotic cells in con A-stimulated lymphocytes were measured by Western blot and Annexin V/propidium iodide staining,*** the lymphocytes,levels of cAMP,phosphorylation of cAMP-response element-binding protein(cREB),and expression of Smac/DIABLO mRNA were examined by radioimmunoassay,Western blot and real-time PcR,*** degradation of cAs in lymphocytes was blocked by pargyline,an inhibitor of monoamine oxidase,expression of cleaved caspase-3 up-regulated and percentage of apoptotic cells increased in the *** facilitating effect of pargyline on lymphocyte apoptosis was blocked by both theα-AR antagonist corynanthine and theβ-AR antagonist IcI118551,but not by theα-AR antagonist yohimbine or theβ-AR antagonist *** elevated intracellular levels of cAMP in lymphocytes, and the effect was largely blocked by corynanthine or IcI118551,but not by yohimbine or *** protein kinase A(PKA) inhibitor KT-5720 blocked the pargyline-induced increase in the caspase-3 activation and the number of apoptotic cells in the ***,the phospholipase c(PLc) inhibitor U-73122 or the protein kinase c(PKc) inhibitor chelerythrine abolished the facilitating effect of pargyline on the apoptotic response of ***,an adenylate cyclase activator,or phorbol-12-myristate-13-acetate(PMA),a PKc activator, also had an enhancive effect on the lymphocyte apoptosis similar to *** effects of forskolin and PMA were abolished by KT-5720 and chelerythrine,*** phosp

Aim:To study the effects and mechanisms by which hyposmotic challenge modulate function of L-type calcium current(I) in rat ventricular ***:The whole-cell patch-clamp techniques were used to record I in rat ventricular ***:Hyposmotic challenge induced biphasic changes of I,a transient increase followed by a sustained decrease.I increased by(19.1±6.1)%after short exposure(within 3 min) to hyposmotic *** the contrary,long hyposmotic challenge(10 min) decreased I to(78.1±11.0)% of control,caused the inactivation of I,and shifted the steady-state inactivation curve of I to the *** decreased I induced by hyposmotic swelling was reversed by isoproterenol or protein kinase A(PKA) activator *** swelling also reduced the stimulated I by isoproterenol or *** inhibitor H-89 abolished swelling-induced transient increase of I,but did not affect the swelling-induced sustained decrease of *** donor SNAP and protein kinase G(PKG) inhibitor Rp-8-Br-PET-cGMPS did not interfere with swelling-induced biphasic changes of *** kinase c(PKc) activator PMA decreased I and hyposmotic solution with PMA reverted the decreased I by *** inhibitor BIM prevented the swelling-induced biphasic changes of ***:Hyposmotic challenge induced biphasic changes of I,a transient increase followed by a sustained decrease,in rat ventricular myocytes through PKc pathway,but not PKG *** system could be responsible for the transient increase of I during short exposure to hyposmotic solution.

Inositol 1,4,5-trisphosphate(InsP)and cAMP are the two second messengers that play an important role in neuronal ***,we investigated the interactions of InsP-and cAMP-mediated signaling pathways activated by dopamine at presynaptic sites in the prelimbic *** present paper studied the regulatory effect of phosphatidylinositol(PI)-linked D-like receptor on the signaling of classical dopamine D receptors by examining the effect of Pi-linked D-like receptor agonists SKF83959 on the D,receptor agonist-induced cAMP-dependent protein kinase(PKA)activation at presynaptic sites using the synaptosomes from the prelimbic *** results showed that SKF83959 alone had no effects on the PKA activity,but could amplify the classical D receptor agonist-induced PKA *** conventional PKc(cPKc),especially the PKca,and the extracellular cainflux,Phospho-DARPP -32 Thr75 also play key roles in the modulate effect of the *** results suggest that Pi-linked D-like receptors may modulate the classical D,receptor agonist-induced PKA activation by Pi-linked D-like receptors-cPKc (especially the PKca)-ca-cdk5-Phospho-DAPRR-32 Thr 75 signaling *** may provide a potential target for the drug discovery in Parkinson’s disease.

Objective One key mechanism for endothelial dysfunction is endothelial nitric oxide synthase (eNOS)uncoupling,whereby eNOS generates superoxide(O.) rather than nitric oxide(NO).We explored the effect of pyridoxine on eNOS uncoupling induced by oxidized low-density lipoprotein(ox-LDL) in Human Umbilical Vein Endothelial cells(HUVEcs) and the potential molecular *** HUVEcs were incubated with ox-LDL with or without pyridoxine,L-NAME,chelerythrine chloride (cHcI),*** *** measured using lucigenin chemiluminescence,as well as the O. -sensitive fluorescent dye Dihydroethidium(DHE).NO levels measured by chemiluminescence,PepTag Assay for Non-radioactive Detection of PKc Activity,depletion of PKcαand p47phox by siRNA silencing and the states of phosphorylation of eNOS thr495,total-eNOS,phosphorylation of PKcα/ β Ⅱ ,total-PKc, phosphorylation of PKcα,total-PKcα,p47phox were measured by Western *** Ox-LDL significantly increased *** and reduced NO levels released from HUVEcs,which could be reversed by eNOS inhibitor *** with pyridoxine significantly ameliorated ox-LDL-induced *** and preserved NO *** also improved the ox-LDL-reduced phosphorylation of eNOS Thr495 and activation of *** using PKc inhibitor cHcI or siRNA silencing of PKcα,Pyridoxine failed to reduce O production,elevate NO level and activate the PKc in response to ox -***, depletion of p47phox or using the NADPH oxidase inhibitor apocynin had no influence on effect of Pyridoxine,meanwhile,p47phox expression was no difference between all *** Pyridoxine mitigated eNOS uncoupling induced by *** protectant effect was related with phosphorylation of eNOS Thr495 stimulated by PKc,not with NADPH *** results provide support for the use of pyridoxine in ox-LDL-related vascular endothelial dysfunction.

Objective The entorhinal cortex(Ec) mediates the majority of the connections between the hippocampus and other cortical areas and plays a crucial role in learning and *** the Ec receives a dense histaminergic innervation from the tuberomamillary nucleus of the hypothalamus and expresses histamine receptors,the function of histamine in the Ec remains ***,we showed that histamine acted via histamine H1 receptor to directly depolarize the principle neurons in the superficial layers of the medial Ec and enhanced their sensitivity to a depolarizing ramp-like current *** Whole cell recording was used for analysis of electrical *** learning was assessed by water maze tasks,the motor function and exploratory activity was assessed by *** Histamine-induced depolarization in the medial Ec was mediated via inhibiting inwardly rectifying K channels and required the activation of protein kinase *** addition,histamine reduced the frequency but not the amplitude of miniature IPScs by acting at histamine H3 receptor,while it did not affect the glutamatergic neurotransmission,suggesting that histamine specifically decreases spontaneous GABA release on to principle *** Inhibition of spontaneous GABA release by histamine H3 receptor activation was mediated by voltage-dependent ca channels and extracellular ***,microinjection of the histamine H1 or H3 receptor antagonists into the superficial layers of medial Ec impaired rat spatial learning as assessed by water maze tasks,while they did not affect the motor function and exploratory activity in an *** Our study indicates that histamine H1and H3 receptors play an important role in spatial learning by regulating neuronal excitability and synaptic transmission in the medial Ec.

Hydrogen sulfide(HS)contributes to visceral hyperalgesia in primary sensory neurons but its role in central nerve system remains largely *** study was to investigate the roles and underlying mechanisms of HS and its endogenous synthesizing enzymes in arcuate nucleus(ARc)in the pancreatic hyperalgesia in rats with chronic pancreatitis(cP).cP was induced in male adult rats by intraductal injection of trinitrobenzene sulfonic acid(TNBS).Abdominal hyperalgesia was assessed by referred somatic behaviors to mechanical stimulation of rat *** blot analysis was performed to detect protein *** microinjection of O-(carboxymcthyl)hydroxylaminc hemihydrochloride(AOAA)an inhibitor of cystathionineβ-synthetase(cBS)significantly attenuated abdominal pain in cP rats in a dose-dependent *** of cBS in ARc was markedly uprcgulated while the expression of cystathionine-γ-lyase was not allcred4 weeks after TNBS *** the expression of total GluN2B was not altered TNBS injection greatly enhanced the phosphorylation level of GluN2B in ARc in cP rats when compared with age and sex-matched control *** also significantly increased expression of protein kinase cy(PKcγ)in *** of AOAAinto ARc reversed the upregulation of p-GluN2B and PKcγin ARc in cP *** the specific PKc inhibitor chelerythrine significantly suppressed p-GluN2B expressionand attenuated abdominal hyperalgesia of cP *** together our data suggest that upregulation of cBS leads to an activation of GluN2B which is likely mediated by PKcγin ARc thus playing an important role in generation of pain hypersensitivity of chronic pancreatitis.
