protein sources and starch-protein digestive dynamics manipulate growth performance in broiler chickens defined by an equilateraltriangle response surface design

A total of 360 male,off-sex Ross 308 chicks were offered 10 dietary treatments from 14 to 35 d post ehatch in an equilateral-triangle response surface design feeding study in order to confirm the importance of protein and amino acid digestive dynamics in broiler chickens.The 3 apical diets were nutritionally-equivalent containing either soybean meal,non-bound amino acids or whey protein concentrate as the major source of dietary protein and amino acids.Appropriate blends of the 3 apical diets comprised the balance of 7 diets and each dietary treatment was offered to 6 replicate cages with 6 birds per cage.Growth performance,nutrient utilisation,apparent protein and starch digestibility coefficients were determined in 4 small intestinal segments.The optimal weight gain(2,085 g/bird)and feed conversion ratios(FCR,1.397)were generated by Diet 50S50W which included a 50:50 blend of apical diets rich in whey protein concentrate and soybean meal.Broiler chickens offered Diet 50S50W also had the highest experimental and predicted jejunal digestibility(0.685 in proximal jejunum and 0.823 in distal jejunum).FCR was not correlated with apparent distal ileal digestibility coefficient(P>0.05)of protein but was correlated with apparent protein digestibility in proximal jejunum(r=-0.369,P=0.040)and distal jejunum(r=-0.316,P=0.015).Surplus dietary starch was correlated with increased fat pad weight(r=0.781,P?0.008).The findings confirmed the relevance of protein digestion rate,reflected by jejunal digestibility,on feed conversion of broiler chickens.A balance between protein-bound and non-bound crystalline or synthetic amino acids may be required for optimal growth and protein digestion.

protein crystallization plays a significant role in three-dimensional structural analysis and protein pu-rification.It is important to increase the crystallization efficiency,which is possible by adding hetero-geneous templates in crystallization systems.DNA is biologically compatible and artificially designable polymer,which is easy to extract.In this study,single-and double-stranded DNA of precise sequences were designed and used as templates to promote protein crystallization of lysozyme and catalase.In-fluence of DNA,single-stranded DNA with 10,20,40 bases and double-stranded DNA with 10,20,40 base pairs,were investigated.The success rate of obtaining crystals of lysozyme and catalase in equal period was significantly improved with the addition of DNA comparing without templates added.Double-stranded DNA led to higher nucleation rate than that with single-stranded DNA.The promotion of nucleation was more obvious at low concentration of protein solution and with longer chain DNA templates.Crystal number and crystallization rate was enhanced with addition of long double-stranded DNA templates.All the results confirm that DNA is an effective polymer additive to enhance protein crystallization,especially for the application of the scarce protein crystallization.

protein@PP-Zn nanocomplex assembled by coordination of zinc ions used for intracellular protein delivery

In recent years,intracellular delivery of protein drugs has attracted great attention,and polymer-based systems have been extensively exploited to develop efficient and safe carriers.However,efficient intracellular delivery of protein drugs remains a challenge because of the cell membrane barrier and endosome entrapment.Herein,we report a protein@PP-Zn nanocomplex,which consists of an imidazole-containing block polymer poly(ethylene glycol)-block-poly(β-amino ester)(PEG-b-PAE(Im),PP),zinc ions,and protein drugs,for efficient intracellular protein delivery.PEG-b-PAE(Im)could conjugate proteins via the bridging effect of zinc ions which simultaneously coordinate with imidazole groups on polymer and electron donor groups,such as imidazole and primary amine groups,on protein to improve the loading stability of proteins.Under a slightly acidic environment near cancer cells,the protonation of PAE(Im)backbone increases the positive charge density of the nanocomplex and promotes endocytosis.While under a more acidic environment in endosomes,further protonation of imidazole groups leads to the disintegration of the nanocomplex and the breakdown of endosomes because of the proton sponge effect.Finally,protein is released into the cytoplasm.With the assistance of the nanocomplex,proteins with different sizes and isoelectric points are effectively delivered into cells.This work provides a stable,efficient and universal strategy for intracellular protein delivery.

protein adsorption preferentially occurs and significantly affects the physicochemical reactions once the biodegradable magnesium alloys as bone replacements have been implanted. To date, interactions mechanisms between Mg implants and proteins remain unclear at a molecular level. Thereby, a combination of molecular dynamic(MD) simulations and experimental exploration is used to investigate the adsorption behavior and conformational change of bovine serum albumin(BSA), a representative protein of blood plasma, upon the surface of microarc oxidation(MAO) coated Mg alloy AZ31. The influences of absorbed proteins on the cytocompatibility of MAO coating are evaluated by virtue of cytotoxicity assay. Results indicate that the negatively charged O atoms(BSA) exhibit strong interaction with Mg^(2+) ions of Mg(OH)_(2), revealing that BSA molecules are ionically adsorbed on the AZ31 surface. Interestingly, MD simulation reveals that MAO coating demonstrates superior ability to capture BSA molecules during the process of adsorption owing to strong electric attraction between the negatively charged O atoms in BSA molecules with Mg atoms of MgO in MAO coating. Moreover, the α-helix part of absorbed BSA molecules on AZ31 substrate and MAO coating markedly decreases with an increase in β-sheet, β-turn and unordered contents, which is attributed to the reduction in the number of hydrogen bonds in BSA molecules. Furthermore, the adsorbed BSA molecules improve the cytocompatibility of MAO coating since the positively charged-NH_(3)^(+) group and β-sheet content of absorbed BSA molecules mediate the cell adhesion by interacting with the negatively charged cell membrane.

protein-modified SEI formation and evolution in Li metal batteries

Despite numerous reported lithium metal batteries(LMBs) with excellent cycling performance achieved in labs, transferring the high performing LMBs from lab-scale to industrial-production remains challenging. Therefore, via imitating the stand-still process in battery production, a conventional but important procedure, to investigate the formation and evolution of a solid electrolyte interface(SEI) is particularly important for LMBs. Our previous studies indicate that zein(corn protein)-modified carbonate-ester electrolyte(the most commercialized) effectively improves the performance of LMBs through guiding Li-ions and repairing cracked SEI. Herein, we investigate the formation and evolution of the protein-modified SEIs on Li anodes by imitating the stand-still temperature and duration. A simulation study on the protein denaturation in the electrolyte under different temperatures demonstrates a highly unfolded configuration at elevated temperatures. The experiments show that this heat-treated-zein(H-zein) modified SEI forms quickly and becomes stable after a stand-still process of less than 100 min. Moreover, the Hzein SEI exhibits excellent wetting behavior with the electrolyte due to the highly unfolded protein structures with more functional groups exposed. The Li|Li cell with the H-zein SEI achieves prolonged cycling performance(>360 h vs. ~260 h of the cell with the untreated-zein(U-zein) modified SEI). The Li Fe PO_(4)|Li cell with the H-zein SEI shows much stable long-term cycling performance of capacity retention(70% vs.42% of the cell with U-zein SEI) after 200 cycles. This study confirms that the appropriately treated protein is able to effectively improve the performance of LMBs, and will inspire future studies for the production process of LMBs toward their commercialization.

protein assembly is the structural basis for the collaboration between proteins to accomplish life activities due to its realization of the domain-limited and precise spatial arrangement of proteins.Therefore,artificial manipulation of protein selfassembly has profound implications in areas such as exploring the mysteries of life and developing biomaterials.In this review,we not only summarize the classical assembly strategies and the structures of exquisite protein assemblies,but also aim to generalize the flexible and controllable assembly tools and the“interaction”between the assembled protein-based materials and the external environment.On the basis of this,the application,challenges and further development of protein assembly in biology are reviewed.

Background:Nutrient regulation has been proven to be an effective way to improve the flesh quality in fish.As a necessary nutrient for fish growth,protein accounts for the highest proportion in the fish diet and is expensive.Although our team found that the effect of protein on the muscle hardness of grass carp was probably related to an increased collagen content,the mechanism for this effect has not been deeply explored.Moreover,few studies have explored the protein requirements of sub-adult grass crap(Ctenopharyngodon idella).Therefore,the effects of different dietary protein levels on the growth performance,nutritional value,muscle hardness,muscle fiber growth,collagen metabolism and related molecule expression in grass carp were investigated.Methods:A total of 450 healthy grass carp(721.16±1.98 g)were selected and assigned randomly to six experimen-tal groups with three replicates each(n=25/replicate),and were fed six diets with 15.91%,19.39%,22.10%,25.59%,28.53%and 31.42%protein for 60 d.Results:Appropriate levels of dietary protein increased the feed intake,percentage weight gain,specific growth rate,body composition,unsaturated fatty acid content in muscle,partial free amino acid content in muscle,and muscle hardness of grass carp.These protein levels also increased the muscle fiber density,the frequency of new muscle fibers,the contents of collagen and IGF-1,and the enzyme activities of prolyl 4-hydroxylases and lysyloxidase,and decreased the activity of matrix metalloproteinase-2.At the molecular level,the optimal dietary protein increased col-lagen type Iα1(Colα1),Colα2,PI3K,Akt,S6K1,La ribonucleoprotein domain family member 6a(LARP6a),TGF-β1,Smad2,Smad4,Smad3,tissue inhibitor of metalloproteinase-2,MyoD,Myf5,MyoG and MyHC relative mRNA levels.The levels of the myostatin-1 and myostatin-2 genes were downregulated,and the protein expression levels of p-Smad2,Smad2,Smad4,p-Akt,Akt,LARP6 and Smad3 were increased.Conclusions:The appropriate levels of dietary protein promoted the growth of sub-adult grass c

The influence of dietary crude protein concentrations,grain types and arginine:lysine ratios on the performance of broiler chickens

The objective of this study was to investigate the effects of dietary crude protein(CP)concentrations,grain types and arginine:lysine ratios on performance parameters of broiler chickens.The 2×2×2factorial array of dietary treatments harnessed two CP concentrations(210 and 170 g/kg),two feed grains(wheat and sorghum),and two arginine:lysine ratios(104 and 110).Each dietary treatment was offered to 7 replicates of 14 birds per floor pen,a total of 784 off-sex male,Ross 308 broilers,from 14 to 35 d post-hatch.The dietary CP reduction compromised weight gain by 10.0%(2078 versus 2310 g/bird)as a main effect and FCR by 7.51%(1.474 versus 1.371),subject to an interaction.In a three-way interaction(P=0.008),expanded arginine:lysine ratios improved FCR by 2.30%in 170 g/kg CP,sorghum-based diets but compromised FCR by 2.12%in corresponding wheat-based diets.Sorghum was the more suitable feed grain in reduced-CP diets as sorghum generated significant advantages in weight gain of 7.59%(2154versus 2002 g/kg)and FCR of 6.94%(1.421 versus 1.527)in birds offered 170 g/kg CP diets.Both dietary CP and feed grain generated significant and divergent impacts in apparent ileal digestibility coefficients for the majority of 16 assessed amino acids.Dietary CP reductions increased non-bound amino acid inclusions(NBAA)in wheat-based diets(48.96 versus 9.80 g/kg)to a greater extent than sorghum-based diets(35.3 versus 9.50 g/kg)and increasing dietary NBAA inclusions were linearly associated with compromised weight gain(r=-0.834;P<0.001)and FCR(r=0.862;P<0.001).Increasing ratios of free arginine to lysine plasma concentrations were linearly(r=-0.466;P=0.004)related to improvements in FCR.The implications of the observed outcomes are discussed and possible explanations are advanced.

The objective of this study was to develop equations for estimating ileal digestible crude protein(CP)and metabolizable energy(ME)contents of meat meal(MM)and meat and bone meal(MBM)as feed in-gredients for pigs based on in vitro assays.Test ingredients were 4 sources of MM and 3 sources of MBM.Ash and CP contents of the ingredients ranged from 3.8%to 33.1%and 46.8%to 82.9%(as-is basis),respectively.In vitro ileal disappearance(IVID)of CP was determined and ileal digestible CP content was calculated by multiplying CP content by IVID of CP.In vitro total tract disappearance(IVTTD)of dry matter(DM)was determined and ME was calculated using gross energy,CP contents,and IVTTD of DM.The IVID of CP and IVTTD of DM ranged from 77.2%to 88.7%and from 82.7%to 92.4%,respectively.Calculated ileal digestible CP and ME contents ranged from 37.8%to 73.5%DM and 2,405 to 3,905 kcal/kg DM,respectively.Ash contents were negatively correlated(P<0.001)with CP(r=-0.99),in vitro ileal digestible CP(r=-0.97),gross energy(r-1.00),in vitro digestible energy(r=-0.97),and adjusted ME(r=-0.97).The most fitting equations for ileal digestible CP and adjusted ME were:ileal digestible CP(%DM)=11.91-0.90×Ash(%DM)+0.74×IVID of CP(%)(R2=0.99)and adjusted ME(kcal/kg DM)=130.85-50.90×ash(%DM)+47.06×IVTTD of DM(%)(R2=0.99).To validate the accuracy of the prediction equations for ME,mean bias and linear bias were determined using a regression analysis.Calculated ME values of MM and MBM were in a good agreement with data obtained from animal ex-periments based on a statistically insignificant bias in the models.In conclusion,ME concentrations of MM and MBM as swine feed ingredients can be calculated using ash concentration and in vitro disap-pearance of dry matter.

A large number of chemokines,cytokines,other trophic factors and the extracellular matrix molecules form a favorable microenvironment for peripheral nerve regeneration.This microenvironment is one of the major factors for regenerative success.Therefore,it is important to investigate the key molecules and regulators affecting nerve regeneration after peripheral nerve injury.However,the identities of specific cytokines at various time points after sciatic nerve injury have not been determined.The study was performed by transecting the sciatic nerve to establish a model of peripheral nerve injury and to analyze,by protein microarray,the expression of different cytokines in the distal nerve after injury.Results showed a large number of cytokines were up-regulated at different time points post injury and several cytokines,e.g.,ciliary neurotrophic factor,were downregulated.The construction of a protein-protein interaction network was used to screen how the proteins interacted with differentially expressed cytokines.Kyoto Encyclopedia of Genes and Genomes pathway and Gene ontology analyses indicated that the differentially expressed cytokines were significantly associated with chemokine signaling pathways,Janus kinase/signal transducers and activators of transcription,phosphoinositide 3-kinase/protein kinase B,and notch signaling pathway.The cytokines involved in inflammation,immune response and cell chemotaxis were up-regulated initially and the cytokines involved in neuronal apoptotic processes,cell-cell adhesion,and cell proliferation were up-regulated at 28 days after injury.Western blot analysis showed that the expression and changes of hepatocyte growth factor,glial cell line-derived neurotrophic factor and ciliary neurotrophic factor were consistent with the results of protein microarray analysis.The results provide a comprehensive understanding of changes in cytokine expression and changes in these cytokines and classical signaling pathways and biological functions during Wallerian degeneration,as well as a bas
