The Asiatic hybrid lily cultivar Lilium dazzling(family Liliaceae)is a perennial herbaceous bulbous plant with bright yellow flowers of ornamental values.It also shows resistance to diseases and abiotic stress,making it an ideal parent for breeding studies.This study established a sterile culture system using scales of dazzling lily as explants to induce polyploidy.Adventitious buds growing to 1 cm were treated with different concentrations of colchicine(125,250 and 500μmol·L-1),pendimethalin or trifluralin(100,200 and 300μmol·L-1,respectively)for 12,24 and 36 h.Flow cytometry and chromosome counting were used to identify ploidy,and the phenotype of the polyploids was examined by stomatal observation and leaf index determination.With increasing mutagen concentration and exposure time,the mortality and mutagenic rate increased.The optimal treatment group was:300μmol·L-1 trifluralin for 12 h,which resulted in 15.55%mortality rate and 42.22%induction rate.The polyploid plants showed a significant increase in stomata length,leaf length and leaf width,with a decrease in stomatal density compared with the control plants.This study provided a basis for polyploid breeding.

Bread wheat(Triticum aestivum L.,AABBDD,2 n=6 x=42),which accounts for most of the cultivated wheat crop worldwide,is a typical allohexaploid with a genome derived from three diploid wild ancestors.Bread wheat arose and evolved via two sequential allopolyploidization events and was further polished through multiple steps of domestication.Today,cultivated allohexaploid bread wheat has numerous advantageous traits,including adaptive plasticity,favorable yield traits,and extended end-use quality,which have enabled its cultivation well beyond the ranges of its tetraploid and diploid progenitors to become a global staple food crop.In the past decade,rapid advances in wheat genomic research have considerably accelerated our understanding of the bases for the shaping of complex agronomic traits in this polyploid crop.Here,we summarize recent advances in characterizing major genetic factors underlying the origin,evolution,and improvement of polyploid wheats.We end with a brief discussion of the future prospects for the design of gene cloning strategies and modern wheat breeding.

Origin and diversification of the Tibetan polyploid cyprinids(schizothoracins) may help us to explore relationships between diversification of the cyprinids and the Tibetan Plateau uplift. Cyprininae phylogeny was analyzed using mitochondrial and nuclear DNA sequences to trace origins of polyploidy and diversifications of schizothoracins. Ancestral states reconstruction for ploidy levels indicated that the Cyprininae was diploid origin and the schizothoracin clades tetraploid origins. There were two diversification rate shifts along with diversification of the cyprinine fishes in response to the Tibetan uplift. The unusual diversification shifts were located to branches subtending the clades of Tibetan polyploid cyprinids. Our analyses suggested that(i) phylogeny of Cyprininae recovered two independent origins of the Tibetan polyploidy schizothoracins;(ii) diversifications of the schizothoracins were closely related to the Neogene uplift of the Tibetan plateau in the following ways: the relatively ancient Late Oligocene-Middle Miocene adaptive radiation may be associated with the uplift of the southern Tibet and Himalaya; the Middle Miocene-Early Pleistocene lineage-specific diversification broadly coincident with major phase of the Neogene Tibetan uplift; and the most recent Pleistocene diversification shift in Schizothorax closely coincident with the successive Kunlun-Huanghe and Gonghe movements of the Tibetan uplift and the glaciation-induced climate oscillations on the plateau.

Sequence-specific nucleases(SSN) that generate double-stranded DNA breaks(DSBs) in genes of interest are the key to site-specific genome editing in plants. Genome editing has developed into one method of reducing undesirable traits in crops by the induction of knockout mutations. Different SSN-mediated genome-editing systems, including LAGLIDADG homing endonucleases or meganucleases, zinc-finger nucleases, transcription activator-like effector nucleases and clustered regularly interspaced short palindromic repeats, are emerging as robust tools for introducing functional mutations in polyploid crops including citrus, wheat, cotton, soybean, rapeseed, potato, grapes, Camelina sativa,dandelion, and tobacco. The approach utilizes knowledge of biological mechanisms for targeted induction of DSBs and their error-prone repair, allowing highly specific changes at designated genome loci. In this review, we briefly describe genome-editing technologies and their application to genetic improvement of polyploid crops.

Studies on the Growth Habits and Characteristics of Two polyploid Indica-Japonica Hybrid Rice with Powerful Heterosis

Based on a series of polyploid indica-japonica hybrid plant lines obtained from a new breeding strategy of using double predominance of wide cross and polyploidization to breed super flee, two polyploid indica-japonica hybrids, PSR073 and PSR120 were studied in their growth periods to show the powerful heterosis in a larger scale and to study the characteristics of polyploid indica-japonica hybrids more elaborately. The leaf age, tiller growth, flowering habits, and agronomic traits of them were observed to analyze their growth habits and characteristics. The results showed that the.agronomic traits of PSR073 and PSR120, such as the plant height, panicle length, grain length, grain width, and 1000-grain weight, all acquired obvious predominance of polyploidy, and that the seed setting rate was more than 83%. No significant difference was observed between the two tetraploids and common diploids in the leaf age, tiller growth, and flowering habits. It was concluded that the characteristics of the two powerful heterosis polyploid hybrids were different from those of the polyploid rice reported earlier. Wide cross and polyploidization had no negative effects on their growth habits and characteristics; on the contrary these had powerful heterosis. This had provided theoretic and practical evidences for their application to agricultural production.

Cultivated potato is a clonally propagated autotetraploid species with a highly heterogeneous genome.Phased assemblies of six cultivars including two chromosome-scale phased genome assemblies revealed extensive allelic diversity,including altered coding and transcript sequences,preferential allele expression,and structural variation that collectively result in a highly complex transcriptome and predicted proteome,which are distributed across the homologous chromosomes.Wild species contribute to the extensive allelic diversity in tetraploid cultivars,demonstrating ancestral introgressions predating modern breeding efforts.As a clonally propagated autotetraploid that undergoes limited meiosis,dysfunctional and deleterious alleles are not purged in tetraploid potato.Nearly a quarter of the loci bore mutations are predicted to have a high negative impact on protein function,complicating breeder’s efforts to reduce genetic load.The StCDF1 locus controls maturity,and analysis of six tetraploid genomes revealed that 12 allelic variants of StCDF1 are correlated with maturity in a dosage-dependent manner.Knowledge of the complexity of the tetraploid potato genome with its rampant structural variation and embedded deleterious and dysfunctional alleles will be key not only to implementing precision breeding of tetraploid cultivars but also to the construction of homozygous,diploid potato germplasm containing favorable alleles to capitalize on heterosis in F1 hybrids.

Lectins are natural proteins in animals,plants,and microorganisms and can be divided into 12 families.These lectins play important roles in various environmental stresses.Some polyploid plants show tolerance to environmental stresses and to insect pests.However,the mechanism of stress tolerance is unclear.Tetraploid Robinia pseudoacacia(4×)under salt stress showed higher tolerance than diploid R.pseudoacacia(2×).As lectin can improve stress tolerance,it was questioned whether the stress resistance of polyploid plants was related to the lectin protein.In this study,salt resistance of lectin gene TRpL1 was verified by its over-expression in plants.In addition,salt resistance of lectin protein by E.coli strains was detected.The data revealed that the over-expression transgenic plants of TRpL1 showed better salt tolerance than control plants under salt stress,and the TRpL1-expressing strain also grew better in the medium with added NaCl.Therefore,tetraploid plants can resist salt stress through TRpL1 protein regulation.

is the most important ability for organisms to survive in diverse habitats. Animals have the option to escape from stressful environments, but plants do not. In plants, polyploids consist of about 30%-70% angiosperms and 95% ferns, of which some are important crops such as cotton and wheat. How polyploid plants adapt to various habitats has been a fundamental question remained largely unanswered. The tetra- ploid Shepherd＇s purse （Capsella bursa-pastoris） is one of the most successful plants on earth and has been distributed across the world, thus being an ideal model system for studying the adaptation of poly- ploids. We found that there are frequent introgressions from congeneric diploids to Shepherd＇s purse. Ecological niche modeling suggests that ecological differentiation is evident between the introgressed and non-introgressed C. bursa-pastoris, and the introgressions are a source of adaptation. This result links an evolutionary process to the adaptation of polyploids, and sheds light on the breeding strategy of poly- ploids as well. We conclude that frequent introgressions from congeneric diploids contributed to the acqui- sition of adequate genetic variations, thereby allowing C. bursa-pastoris to adapt to various habitats across the world. Our results highlight how a polypioid could have successfully established after it originated.

In the year 2018,the world witnessed the finale of the race to sequence the genome of the world’s most widely grown crop,the common wheat.Wheat has been known to bear a notoriously large and complicated genome of a polyploidy nature.A decade competition to sequence the wheat genome initiated with a single consortium of multiple countries,taking a conventional strategy similar to that for sequencing Arabidopsis and rice,became ferocious over time as both sequencing technologies and genome assembling methodologies advanced.At different stages,multiple versions of genome sequences of the same variety(e.g.,Chinese Spring)were produced by several groups with their special strategies.Finally,16 years after the rice genome was finished and 9 years after that of maize,the wheat research community now possesses its own reference genome.Armed with these genomics tools,wheat will reestablish itself as a model for polyploid plants in studying the mechanisms of polyploidy evolution,domestication,genetic and epigenetic regulation of homoeolog expression,as well as defining its genetic diversity and breeding on the genome level.The enhanced resolution of the wheat genome should also help accelerate development of wheat cultivars that are more tolerant to biotic and/or abiotic stresses with better quality and higher yield.
