Decreased levels of phosphorylated synuclein in plasma are correlated with poststroke cognitive impairment

Poststro ke cognitive impairment is a major secondary effect of ischemic stroke in many patients;however,few options are available for the early diagnosis and treatment of this *** aims of this study were to(1)determine the specific relationship between hypoxic andα-synuclein during the occur of poststroke cognitive impairment and(2)assess whether the serum phosphorylatedα-synuclein level can be used as a biomarker for poststro ke cognitive *** found that the phosphorylatedα-synuclein level was significantly increased and showed pathological aggregation around the cerebral infa rct area in a mouse model of ischemic *** addition,neuronalα-synuclein phosphorylation and aggregation were observed in the brain tissue of mice subjected to chronic hypoxia,suggesting that hypoxia is the underlying cause ofα-synuclein-mediated pathology in the brains of mice with ischemic *** phosphorylatedα-synuclein levels in patients with ischemic stroke were significantly lower than those in healt hy subjects,and were positively correlated with cognition levels in patients with ischemic ***,a decrease in serum high-density lipoprotein levels in stroke patie nts was significantly correlated with a decrease in phosphorylatedα-synuclein *** ischemic stroke mice did not show significant cognitive impairment or disrupted lipid metabolism 14 days after injury,some of them exhibited decreased cognitive function and reduced phosphorylatedα-synuclein *** together,our results suggest that serum phosphorylatedα-synuclein is a potential biomarker for poststroke cognitive impairment.

＜正＞Objective synuclein is a protein associated with familial and sporadic PD.α-synuclein is hyperphosphorylated at 129 serine in PD patients and animal models,approximately 90%in Lewy bodies,while only 4%normally;and phosphorylated a-synuclein is the main form of *** the brain,α-synuclein is widely expressed,but it is especially neurotoxic to dopaminergic neurons in the substantia nigra pars compacta（SNpc）.This may be due to the fact that nigral dopaminergic neurons are particularly exposed to oxidative stress due to DA *** present there are a few commercial antibodies against phosphorylatedα-synuclein and are less efficient for PD studies,furthermore no laboratory test that correlates with PD risk is *** tests for PD are critically needed for developing treatment designed to slow or prevent progression of the *** biomarkers of PD are also attracted more and more attention, we prepare monoclonal antibodies against phosphorylatedα-synuclein at Serl29,and establish a new in vitro diagnosis of PD Detection Kit for study on the pathogenesis of PD in future as well as on validation of phosphorylatedα-synuclein as a biomarker for early diagnosis of *** To obtain the antiserum against phosphorylatedα-synuclein at Serl29, two Blb/c mice were immunized with 14 amino acids（EMPpSEEMPpSEE-GYQ）as hapten which coupled with *** carbodiimide（EDC） coupling chemical technique was used to form a coupling agent（peptide-mcKLH） by GL Biochem （Shanghai）.The B cells taken from spleens of the mice were fused with myeloma cell（SP2 Cell）.Results After sufficient screening and testing work by ELISA,western-blot and immunoblot,we obtained the ideal monoclonal antibody of phosphorylatedα-synuclein at *** Subsequently,our next work will focus on the establishment of a new in vitro diagnosis of PD Detection Kit,to make any contribution to PD research.
