Epstein-Barr virus DNA loads in the peripheral blood cells predict the survival of locoregionally-advanced nasopharyngeal carcinoma patients

Objective:Circulating cell-free Epstein-Barr virus(EBV)DNA has been shown to be a valuable biomarker for population screening and prognostic surveillance for nasopharyngeal carcinoma(NPC).Despite important insights into the biology of persistence,few studies have addressed the clinical significance of cell-based EBV-DNA loads in peripheral blood cells(PBCs).Methods:A prospective observational cohort study was conducted involving 1,063 newly diagnosed,locoregionally-advanced NPC patients at Sun Yat-sen University Cancer Center from 2005 to *** regression analysis was conducted to identify the association of PBC EBV DNA loads to overall survival(OS)and other prognostic *** nomograms were developed based on PBC EBV DNA loads to predict survival outcomes for NPC ***:After a median follow-up of 108 months,patients with higher PBC EBV-DNA loads had significantly worse OS[hazard ratio(HR)of medium,medium-high,and high *** were 1.50,1.52,and 1.85 respectively;Ptrend<0.001].Similar results were found for progression-free survival and distant metastasis-free *** concordance index of the prognostic nomogram for predicting OS in the training set and validation set were 0.70 and 0.66,*** data showed that the PBC EBV DNA load was an independent and robust survival biomarker,which remained significant even after adjusting for plasma EBV DNA loads in a subset of 205 patients of the cohort(HR:1.88;P=0.025).Importantly,a combination of PBC EBV DNA load and plasma EBV DNA load improved the predicted ***:The EBV-DNA load in PBCs may be an independent prognosis marker for NPC patients.

Developing fluorescence probes with multiple responses has vital significance but remains ***,for the first time,we present a mitochondrial DNA(mtDNA)-triggered pH response signalamplified fluorescent probe(QCY-DBT)for multiple cell *** probe exhibited a large stokes shift(229 nm),excellent DNA selectivity over RNA,and ultrasensitivity of detection limit(DL;74.0 ng/mL).Thus,QCY-DBT was successfully applied to analyze multiple human peripheral blood cells and visualize mtDNA in healthy and apoptotic *** the tumor acidic environment(pH 6.0–7.0),the absorbance of QCY-DBT at 436 nm increased,and the fluorescence signal(665 nm)was amplified by mtDNA,which enabled the direct observation of tumor *** study provides help in designing smart probes with multiple responses for efficient abnormal cell detection.

Induced pluripotent stem cells:origins, applications, and future perspectives

Embryonic stem(ES)cells are widely used for different purposes,including gene targeting,cell therapy,tissue repair,organ regeneration,and so ***,studies and applications of ES cells are hindered by ethical issues regarding cell *** circumvent ethical disputes,great efforts have been taken to generate ES cell-like cells,which are not derived from the inner cell mass of blastocyst-stage *** 2006,Yamanaka et *** reprogrammed mouse embryonic fibroblasts into ES cell-like cells called induced pluripotent stem(iPS)*** one year later,Yamanaka et *** Thomson et *** reprogrammed human somatic cells into iPS *** the first generation of iPS cells,they have now been derived from quite a few different kinds of cell *** particular,the use of peripheral blood facilitates research on iPS cells because of safety,easy availability,and plenty of cell *** iPS cells have been used for cell therapy,disease modeling,and drug *** this review,we describe the generations,applications,potential issues,and future perspectives of iPS cells.

Upregulation of Nrf2-regulated gene expression by tBHQ alleviates cyclophosphamide-induced hematotoxicity in mice

Hematological toxicity （bone marrow suppression） is the most common dose-limiting adverse effect of chemotherapies. The nuclear factor erythroid 2-related factor 2 （Nrf2） is a pivotal coordinator of cellular defensive responses against chemical insults in many tissues including bone marrow. In the present study, the effects of tert-butylhydroquinone （tBHQ） on the expression of Nrf2-regulated genes in peripheral blood cells and cyclophosphamide （CTX）-induced hematotoxicity in mice were investigated. CTX induced apoptosis of peripheral blood nucleated cells and leukopenia in mice, accompanied by mobilization of bone marrow hematopoietic cells, tBHQ treatment induced the expression of Nrf2-regulated genes such as heine oxygenase 1 （HO1） and glutamate-cysteine ligase catalytic subtmit （GCLC） in RAW264.7 mouse macrophage cells and peripheral blood cells both in vitro and in vivo. Interestingly, pretreatment with tBHQ alleviated CTX-induced mouse peripheral blood cell apoptosis and leukopenia in vivo, indicating possible involvement of Nrf2 in the protection against CTX-induced hematotoxicity. This study provides new information on the chemotherapy-induced hematotoxicity, and suggests Nrf2 could serve as a target for the development of chemoprotectants against hematotoxicity.
