Multiplex RT-PCR-based detections of CEA, CK20 and EGFR in colorectal cancer patients

AIM: To develop a multiplex reverse transcription polymerase chain reaction (RT-PCR) method detecting cir-culating tumor cells in the peripheral blood of colorectal cancer (CRC) patients. METHODS: peripheral blood samples were collected from 88 CRC patients and 40 healthy individuals from the blood donors' clinic and subsequently analyzed by multiplex RT-RCR for the expression of carcinoembryonic antigen (CEA), cytokeratin 20 (CK20) and epidermal growth factor receptor (EGFR) mRNA. The analysis involved determining the detection rates of CEA, CK20 and EGFR transcripts vs disease stage and overall survival. Median follow-up period was 19 mo (range 8-28 mo). RESULTS: Rates of CEA, CK20 and EGFR detection in CRC patients were 95.5%, 78.4% and 19.3%, respectively. CEA transcripts were detected in 3 healthy volunteer samples (7.5%), whereas all control samples were tested negative for CK20 and EGFR transcripts. The increasing number of positive detections for CEA, CK20 and EGFR transcripts in each blood sample was positively correlated with Astler-Coller disease stage (P< 0.001) and preoperative serum levels of CEA (P=0.029) in CRC patients. Data analysis using Kaplan-Meier estimator documented signif icant differences in the overall survival of the different CRC patient groups as formed according to the increasing number of positivity for CEA, CK20 and EGFR transcripts. CONCLUSION: These data suggest that multiplex RTPCR assay can provide useful information concerning disease stage and overall survival of CRC patients.

鉴于国内还缺乏正常人外周血T细胞亚群的有关资料,我们利用单克隆抗体T11、T4和T8及免疫细胞化学技术对一组成年人外周血中T细胞及其亚群分布进行了研究。我国正常人外周血中T细胞、T4+细胞及T8+细胞在单个核细胞中的比例分别为65.89±8.31;43.38±7.48;25.03±6.41％。无明显性别差异,但50～69岁老年人前期T细胞及T4+细胞有所降低,而T8+细胞有增高趋势。同时我们还发现血中的T细胞及其亚群存在日间节律性变动,建议应在每天上午8～9时采血。

BACKGROUND An immediate hypersensitive immune response to Aspergillus fumigatus antigens is one of the main characteristic features of allergic bronchopulmonary aspergillosis(ABPA).As ABPA is an allergic respiratory disease,immunoglobulin E and peripheral-blood eosinophilia have been used as diagnostic ***,eosinophilia in bronchoalveolar lavage fluid(BALF)has not been considered in the diagnostic criteria for *** SUMMARY We present a case of ABPA in which the eosinophil count in peripheral blood was not increased,whereas the eosinophil percentage in BALF reached 60%.After antifungal and hormone therapy,imaging revealed very good resolution of lung *** The value of the eosinophil count in BALF for the diagnosis of ABPA is worthy of the clinician's attention,especially when the patient’s clinical features lack specificity and the diagnostic parameters are negative.

Abnormal expression of hepatomas and circulating telomerase and its clinical values

BACKGROUND: Hepatocellular carcinoma (HCC) is one of the most common malignant tumours worldwide. Te-lomerase is reactivated in various types of malignant tumors and may contribute to the development of HCC. To evaluate the role of telomerase in formation and development of HCC, we analyzed its expression status in different parts of HCC tissues and peripheral blood mono-nuclear cells (PBMCs) , and explored its clinical implications for diagnosis of HCC. METHODS: Total RNAs and telomerase were extracted from HCC and their non-cancerous tissues, and both relationships were analyzed between them. The expression of telomerase reverse transcriptase (hTERT) mRNA and telomerase activities in liver tissues and PBMCs were detected by RT-PCR and telomeric repeat amplification protocol (TRAP)-ELISA, respectively. The diagnostic values of telomerase in PBMCs were investigated in the diagnosis and differentiation of HCC. RESULTS: The specific activities of telomerase were 18.25 ±15.02 A/μg RNA in HCC tissues, and significantly higher than those in their non-cancerous tissues (8.16 ± 6.22 A/μg RNA, P <0. 05). But total RNA levels in HCC were 12.40 ±7.34 μg/mg wet liver and lower than 53.77 ± 52.02 μg/mg wet liver in their non-cancerous parts (P < 0.01). The different telomerase levels could be detected in PBMCs from patients with chronic liver diseases and control group. The enzyme activities were significantly higher in HCC than in any other groups (P <0.01). However, circulating telomerase levels were more obviously decreased in HCC patients after transcatheter arterial embolization treatment than before the treatment (P <0.01). The analysis of combined serum alpha-fetoprotein level and PBMCs telomerase was more sensitive and specific for the diagnosis of HCC. CONCLUSION: The abnormal expression of telomerase in HCC tissues and circulating PBMCs could be a useful marker to the diagnosis and prognosis of HCC.

Recent cancer research has demonstrated the existence of circulating tumor cells(CTCs)in cancer patient’s *** identified,CTC biomarkers will be invaluable tools for clinical diagnosis,prognosis and *** this review,we propose ex vivo culture as a rational strategy for large scale amplification of the limited numbers of CTCs from a patient sample,to derive enough CTCs for accurate and reproducible characterization of the biophysical,biochemical,gene expressional and behavioral properties of the harvested *** of tumor cell heterogeneity,it is important to amplify all the CTCs in a blood sample for a comprehensive understanding of their role in cancer *** analyzing critical steps and technical issues in ex vivo CTC culture,we developed a cost-effective and reproducible protocol directly culturing whole peripheral blood mononuclear cells,relying on an assumed survival advantage in CTCs and CTC-like cells over the normal cells to amplify this specified cluster of cancer cells.

Objective: To investigate the relationship between the activation markers of platelets and the lung cancer. Meth- ods: Based on international stages of lung cancer in 1997, lung cancer patients of 120 cases diagnosed by pathology as well as with operation indication were selected as the experimental group. During the process of experiment, 60 cases concluded as healthy in the physical examination were chosen as control group. The activation markers of platelets were detected by FCM method. The experimental result would be processed by SPSS 11.5. Results: The level of activation markers of platelets in peripheral blood of lung cancer patients was significantly higher than those healthy people (P peripheral blood of lung cancer patients on the seventh postoperative day was significantly lower than that before operation and on the first postoperative day (P peripheral blood of lung cancer patients was closely related to the size of the primary tumor, lymph node status and stages, but not to the grade of cell differentiation, type of tumor, age, sex of the patients (P > 0.05). Conclusion: Elevation of the level of activation markers of platelets in peripheral blood exists in lung cancer patients and the levels of activation marker of platelets plays an important role in tumor growth and lymphatic metastasis. The levels of activation markers of platelets maybe a predictor for prognosis.

Brain derived neurotrophic factor(BDNF) has been known to play an important role in various mental disorders or diseases such as Alzheimer’s disease(AD).The aim of our study was to assess whether BDNF promoter methylation in peripheral blood was able to predict the risk of AD.A total of 44 AD patients and 62 age- and gender-matched controls were recruited in the current case-control *** the bisulphite pyrosequencing technology,we evaluated four CpG sites in the promoter of the *** results showed that BDNF methylation was significantly higher in AD cases than in the controls(CpGl:p = 0.021;CpG2:p = 0.002;CpG3:p = 0.007;CpG4:p = 0.005;average methylation:p = 0.004).In addition,BDNF promoter methylation was shown to be significantly correlated with the levels of alkaline phosphatase(ALP),glucose,Lp(a),ApoE and ApoA in males(ALP:r =-0.308,p =0.042;glucose:r =-0.383,p= 0.010;Lp(a):r = 0.333,p = 0.027;ApoE:r =-0.345,p= 0.032;),ApoA levels in females(r = 0.362,p = 0.033),and C Reactive Protein(CPvP) levels in both genders(males:r =-0.373,p =0.016;females:r =-0.399,p=0.021).Our work suggested that peripheral BDNF promoter methylation might be a diagnostic marker of AD risk,although its underlying function remains to be elaborated in the future.
