Effects of a Herbal Formula on Biochemical Markers of Bone Turnover in ovariectomized rats

Objective: To investigate the effect and mechanism of a Chinese herbal formula MG in ameliorating estrogen deficiency bone loss. Methods: 10 month old female rats were randomly divided into three groups: ovariectomized (OVX), OVX treated with MG decoction (OVX M) and OVX treated with estrogen injection (OVX E). The urinary pyridinoline creatinine ratio (Pyd/Cr), deoxypyridinoline creatinine ratio (Dpd/Cr), and plasma alkaline phosphatase (ALP) were determined at the baseline, 1, 4, 8 and 12 postoperative weeks. Results: Our data showed an average increase of 93% in Pyd/Cr ( P rats, the increase of the OVX M group were prevented 39% in Pyd/Cr, 102% in Dpd/Cr and 19% in ALP respectively ( P <0.05). Conclusion: The herbal formula MG possesses a therapeutic effect on estrogen dependent bone loss by inhibiting the bone turnover as estrogen replacement.

＜正＞Objective:To investigate the effect of Fructus Psoraleae on the weight and leptin level in ovariectomized ***:Female ovariectomized SD rats,weighted（220±3） g,were randomly divided into three groups, ovariectomized group,sham operated group and Fructus Psoraleae *** ovariectomized operation,we studied the effects of Fructus Psoraleae on the weight,foodintake and water-intake,and measured the levels of leptin and estrogen in ***:（1） The weight of ovariectomized group was significantly higher than those of sham operated group and Fructus Psoraleae group（P＜0.001,P＜0.01 respectively）;（2） The leptin level in ovariectomized group was significantly higher than those in shamoperated group and Fructus Psoraleae group （P＜0.05,P＜0.01,respectively）.（3） The levels of estrogens in ovariectomized group and Fructus Psoraleae group were significantly lower than that in shamoperated group（all P＜0.01）.Conclusion:Fructus Psoraleae can reduce the weight by leptin which joins its receptor to control the appetite of ovariectomized rats.

＜正＞Prolactin-releasing peptide（PrRP） was isolated and identified as an endogenous hypothalamic ligand for the GPRIO（G protein-coupled receptor 10）.The anatomical distribution of PrRP-immunoreactive（PrRP-ir） neurons in the central nervous system supports the hypothesis that PrRP could be involved in the regulation of hypothalamus-pituitaryovary axis（HPOA）.The aim of this study was to examine the action of brain PrRP and to investigate the mechanism of PrRP’s effect by measuring of hypothalamic gonadotropin releasing hormone（GnRH） and pituitary luteinizing hormone （LH） releasing in ovariectomized rats in *** the present experiment,PrRP-31 could inhibit the super-release of GnRH directly but didn’t affect LH *** we have shown for the first time the co-localization of GnRH and GPRIO in hypothalamic preoptic area（POA）,which might provide the direct morphological evidence that PrRP might be partly responsible for the regulation of GnRH release via its receptor3/4GPR10 in rat *** results suggested that *** a regulatory neuropeptide,regulates the gonadotrophin secretion acting via a hypothalamic mechanism involving the release of GnRH.

A Preliminary Study of The Short Circuit Current(I_(sc)) Responses of Endometria from Estrum and Bilateral Ovariectomy Female rats

Objective To investigate the property of the endometrium ion transport in the normal estruation and ovariectomize rats. Methods The basic electrophysiological property of the endometrium was carried out by mean of the short circuit current （Isc） recording with the rats. Results The baseline Lsc and the transepithelial resistance （Rte） were different between the estruation and ovariectomized groups. Apical application of Na＋ channel blocker, amiloride （10 μmol/L）, or CFTR Cl- channel agonist, forskolin （10 μmol/L） did not significantly affect the Lc in the two groups. However, the 1sc in the model group was more sensitive to the CFTR Cl- channel blocker （glibenclamide, 1 mmol/L, apical） decreased about 27.0% （P〈0.05, n=6）, no remarkably changes in the estruation rats. While apical application CA CC CI-channel blocker, DIDS （4, 4 ＇-diisothiocyanostilbene- 2,2 ＇-disulfonic, 100 μmol/L）, the Isc was much more sensitive, percentage almost to 36.52% （P〈0.01） in the model group than that in the control Furthermore, basolateral application of bumetanide （100μmol/L）, a blocker of Na＋-K＋-2Cl- cotransport, didn＇t significantly reduce the Lc in the two groups. Conclusion The baseline Lsc and membrane resistance in the ovariectomize group is much higher than that in the normal estruation group, and the endometrial epithelium CI- secretion may be mediated by predominantly calcium-dependent CI- channels （CACC） and activating adenylate cyclase and apical cAMP-dependent Cl- channels （CFTR） in the model group with minor contributions in the control. Furthermore, the endometrial epithelium responses to different stimulants exists difference in the control and model group a likely mechanism for the ovary hormone.

Antihypertensive drugs may have an effect on osteoporosis(OP),bone mineral density(BMD),and bone *** present study was designed to investigate the preventive effects of captopril,an angiotensin-converting enzyme inhibitor(ACEI),on osteoporosis in aged ovariectomized(OVX) rats and its impact on the differentiation of cultured primiary *** 10-month-old Wistar rats were ovariectomized(OVX) and,2 months later,were treated with captopril or *** static and dynamic parameters in trabecular bone of the forth lumbar vertebrae(LV4) were examined with histomorphometrical analysis,the fifth lumbar vertebrae(LV5) were used to perform the compression *** osteoblasts were isolated from the calvaria of new born rats and treated with various concentrations of captopril for various *** characterization of osteoblastic origin was carried out with the staining of alkaline phosphatese(ALP).The effects of captopril on the content of ALP and the mRNA expression of collagenⅠwere determined with examination of para-nitro-pheneye phosphate and RT-PCR..Under the circumstances of estrogen deficiency-induced osteopenia,Captopril increased trabecular area(%***) up to 33%,improved the biomechanical status,increased the break stress and elastic modulus in OVX *** a lower dose,the agent increased the break load,break stress and elastic ***,Captopril promoted the expression of ALP and stimulated the anabolic metabolism of collagenⅠin cultured rat *** summary, Captopril,as one of the most widely used ACEI,has the potential of improving the biomechanical status in osteoporosis and promoting the bone formation in osteoblasts.
