Propagation dynamics of off-axis noncanonical vortices in a collimated Gaussian beam

It is widely accepted that an off-axis noncanonical vortex moves across the free-space diffracting Gaussian beam without rotation. But our analysis shows that the vortex swirls a while before it approaches infinite. By neglecting the divergence of the host beam, we focus on this rotation characteristics of the vortices in linear homogeneous media. For the symmetrical host beam, it is found that the vortex moves along an elliptical trajectory, while the topological charge and the angular momentum of the vortex core relative to the beam axis are conserved. For the asymmetrical host beam, the vortex trajectory is rather complicated, since the noncanonical parameter varies as the light propagates, resulting in topological charge inversion. But we find that the vortices are always confined in a rectangular area, and the rotation direction is determined by the topological charge.

Strawberry Notch 1 (SBN01) promotes proliferation of spermatogonial stem cells via the noncanonical Wnt pathway in mice

While it is known that spermatogonial stem cells (SSCs) initiate the production of male germ cells, the mechanisms of SSC self-renewal, proliferation, and differentiation remain poorly understood. We have previously identified Strawberry Notch 1 (SBN01), a vertebrate strawberry notch family protein, in the proteome profile for mouse SSC maturation and differentiation, revealing SBN01 is associated with neonatal testicular development. To explore further the location and function of SBN01 in the testes, we performed Sbnol gene knockdown in mice to study the effects of SBN01 on neonatal testicular and SSC development. Our results revealed that SBN01 is required for neonatal testicular and SSC development in mice. Particularly, in vitro Sbnol gene knockdown with morpholino oligonucleotides caused a reduction of SSCs and inactivation of the noncanonical Wnt pathway, through Jun N-terminal kinases. Our study suggests SBN01 maintains SSCs by promoting the noncanonical Wnt pathway.

Background:noncanonical autophagy is generally described as a lysosomal degradation process that requires only a subset of the core autophagy-related proteins to form functional ***:Accumulating evidence implicates noncanonical autophagy pathways in expanding the versatility of the immune system via regulation of functions that include antigen presentation,dead cell clearance,inflammatory cytokine production,and immune cell *** this review,we use microtubuleassociated protein 1 light chain 3-associated phagocytosis(LAP)as an example of noncanonical autophagy,describing its distinctive molecular machinery and highlighting recent advances in its functioning in *** also discuss the direct and indirect evidence supporting the pathogenic significance of abnormal levels of LAP in systemic lupus erythematosus(SLE).Future Perspectives:A better understanding of the role of noncanonical autophagy in SLE may reveal crucial information about the disease pathology,providing direction for therapeutic developments and improved prognosis.
