An amperometric hydrogen peroxide biosensor using a nanobiocomposite based on neutral red modified carbon nanotubes and co-immobilized glucose oxidase and horseradish peroxidase is reported. Modification of the nanobiocomposite electrode with neutral red resulted in a sensitive, low-cost and reliable H_2O_2 sensor. The use of carbon nanotubes, as the conductive part of the composite, facilitated fast electron transfer rates. The biosensor was characterized for the influence of p H, potential and temperature. A remarkable feature of the biosensor is the detection of H_2O_2 at low applied potentials where the noise level and interferences are minimal. The sensor has a fast steady-state measuring time of 10 s with a quick response(2 s). The biosensor showed a linear range from 15 n M to 45 m M of H_2O_2 and a detection limit of 5 n M. Nafion, which is used as a binder, makes the determination free from other electroactive substances. The repeatability, reproducibility,stability and analytical performance of the sensor are very good.

Adsorption of neutral red (NR) onto peanut husk in aqueous solutions was investigated at 295 K. Experiments were carried out as function of pH, adsorbent dosage, contact time, and initial concentration. The equilibrium adsorption data were analyzed by Langmuir, Freundlich, Temkin, Dubinin-Radushkevich, and Toth isotherm models. A voltammetric study of the interaction of neutral red(NR) with DNA at a gold electrode in a phosphate buffer solution is described. After adding DNA in an NR solution, the reduction peak current of NR decreases. The binding mechahisms of NR to DNA in different pH ranges are different. The reduction peak potential of NR in a pH 7.0 phosphate buffer solution in the presence of DNA shifts positively, indicating that the binding of NR to DNA is intercalation action, but at pH=6.0 the reduction peak potential of NR shifts negatively, indicating that the binding of NR to DNA is electrostatic action. The formed complexes are DNA-NR when [NR]/[DNA]0.35, respectively.

The binding characteristics of neutral red (NR) with DNA were investigated by fluorescence spectrometry. Chemometrics approach as singular value decomposition (SVD) was used to evaluate the number of spectral species in the drug-DNA binding process, and then the intrinsic binding constant of 1.6 104 in base pairs and the binding site number of 0.97 were obtained from the Scatchard plot.

[ Objective] The aim of this study was to provide the reference for the artificial breeding of aquatic animals. [ Method] Amoeba discoides and Trichodina were treated by neutral red solution with different concentrations to observe the physiological activities of organelles in their death processes. Effects of neutral red solution on the growth of common aquatic animals such as Paramecium caudatum, Euglena viridis and Brachionus plicatilis were analyzed, and the specific lethal mechanism of trace neutral red to A. discoides and Trichodina was also preliminarily studied. [ Re- suit] The neutral red solution at a concentration of 0.5 mg/L damaged the physiological function of contractile vacuole in Trichodina and also had the specific lethal effect on Tdchodina, but it had no effect on the growth and reproduction of non-parasitic protozoa and B. p/icatilis. neutral red so- lution with certain concentration led to disorder of the physiological functions of A. discoides, such as assimilation and rejection, which was a main factor that caused the death of A. discoides. [ Conclusion] With the advantages such as targeting, safety and easiness to be oxidized and decom- posed, neutral red is an ideal drug for treating the diseases caused by A. discoides and Trichodina, and its suitable concentration is 0. 5 mg/L.

Using lysosomal membrane stability of haemocytes in Ruditapes philippinarum as a biomarker of cellular stress to assess contamination by caffeine, ibuprofen, carbamazepine and novobiocin

In this work, for the first time, neutral red functionalized FeO@SiO was prepared, characterized and subsequently used for magnetic solid phase extraction of Hg(Ⅱ) from environmental water samples followed by inductively coupled plasma-mass spectrometry determination. TEM images shows uniform decoration of SiO on FeO surface(with a typical particle size of 220 nm). The FT-IR analysis indicates that neutral red was combined to the SiO surface by chemical bonds. Trace level of Hg(Ⅱ) can be extracted efficiently and selectively due to the electrostatic and coordinate interactions of Hg(Ⅱ) and neutral red. Parameters affecting quantitatively recovery of Hg(Ⅱ) were investigated, and the optimized values were 6.0, 25 mg, 60 min, and 5 mL of 1.0 molL HNO for pH value of the solution, amount of the extractant, equilibration time and volume, concentration of eluent, respectively. Under the optimized conditions, at Hg(Ⅱ) concentration as low as 2 ng m L, the recovery is > 85%, and the enrichment factor is as high as 100. The commonly encountered ions pose no interference on the extraction *** material can be regenerated and reused at least ten times without loss of adsorption capacity(84 mg g). In addition, magnetic separation greatly simplified extraction procedure and filtration or centrifugation process was avoided. Compared with other adsorbent, the material exhibits better selectivity, adsorption capacity, and can be used to extract Hg(Ⅱ) directly in neutral conditions. The material was successfully applied for the separation and pre-concentration of low level of Hg(Ⅱ) in environmental water samples with satisfactory results.

Although pharmaceuticals have beendetected in the environment only in the range from ng/L to μg/L, it has beendemonstrated that they can adversely affect the health status of aquatic organisms. Lysosomal membrane stability （LMS） has previously been applied as an indicator of cellularwell-being todetermine health status in bivalve mussels. The objective of this study is to evaluate LMS in Ruditapes philippinarum haemolymph using the neutral red retention assay （NRRA）. Clamswere exposed in laboratory conditions to caffeine （0.1, 5, 15, 50 μg/L）, ibuprofen （0.1, 5, 10, 50 μg/L）, carbamazepine and novobiocin （both at 0.1, 1, 10, 50 μg/L） for35days. Results show adose-dependent effect of the pharmaceuticals. The neutral red retention time measured at the end of the bioassaywas significantly reduced by 50% after exposure to environmental concentrations （p 〈 0.05）（caffeine = 15 μg/L; ibuprofen = 10 μg/L; carbamazepine = 1 μg/L and novobiocin = 1 μg/L）, compared to controls. Clams exposed to these pharmaceuticalswere considered to present adiminished health status （retention time 〈45 min）, significantlyworse than controls （96 min）（p 〈 0.05）. The predicted no environmental effect concentration （PNEC） results showed that these pharmaceuticals are very toxic at the environmental concentrations tested. Measurement of the alteration of LMS has been found to be a sensitive technique that enables evaluation of the health status of clams after exposure to pharmaceuticals under laboratory conditions, thus representing a robust Tier-1 screening biomarker.

Aims Exposure of Eucalyptus tree stems to the radiant heat of forest fires can kill cambial cells and their embedded regenerative meristems,thus preventing epicormic resprouting and recovery of the ***,there is no tissue-level method to quantify the viability of cambial cells in Eucalyptus following heat *** first aim of this study was to adapt and validate the tetrazolium reduction method of testing for cell viability in *** second aim was to apply the method to establish a threshold level of cambium cell viability in Eucalyptus obliqua to enable the identification of a critical *** The study used the tetrazolium reduction method to quantitatively determine phloem-cambium cell viability in *** sections of bark with underlying phloem and cambium were cut from mature *** and samples ranging in mass from 1 to 30 mg were exposed for 1 min to temperature treatments ranging from 20 to 85℃and kept for 20-22 h at room temperature in 0.8%2,3,5-triphenyl tetrazolium chloride(TTC)to test for cell *** 1,3,5-triphenyl tetrazolium formazan(TPF)formed was cold extracted with ethanol and quantified as absorbance at 485 *** Findings The TTC reduction method reliably quantified a decline in cell viability with rising temperature in tissue sections that included vascular cambium,and identified 60℃as the critical temperature for cambium-phloem cells of Eucalyptus *** viability,calculated as[TPF Treatment℃]/[TPF 20℃],decli ned by 90%between 20 and 85℃.The cell viability results con firmed that significant tissue necrosis occurred in Eucalyptus at temperatures between 50 and 70℃,after 1 min of in vitro tissue *** decline in cell viability with increasing temperature shown by the TTC method was consistent with an independently derived count of live cells following temperature treatment and neutral red staining.
