Flavonoids from traditional Chinese herbs for diabetes in rats:a network Meta-analysis

OBJECTIVE:In this Meta-analysis,we evaluated the hypoglycemic effect of 5 flavonoids found in traditional Chinese herbs(naringenin,kaempferol,puerarin,baicalein,and luteolin)on diabetic rats.METHODS:Four databases including Pub Med,Web of Science,Embase,and Cochrane Library,were searched from inception to May 2020.Only studies using diabetes model rats were included in the analysis.Blood glucose data from the last measurement were collected and analyzed.Pair-wise Meta-analyses were conducted using STATA v14.0 software and a Meta-analysis was conducted using STATA v14.0,ADDIS v1.16.6,and R v3.6.1.The quality of included studies was assessed with the SYRCLE risk of bias tool for animal studies,and publication bias was evaluated with a comparisonadjusted funnel plot.RESULTS:A total of 33 studies were included in the analysis,in which all 5 flavonoids showed a beneficial effect on blood glucose level of diabetic rats were included in the final analysis.The standardized mean differences(95%confidence intervals)were-4.92(-6.67,-3.17)for naringenin,-12(-18.74,-5.27)for kaempferol,-2.52(-3.77,-1.26)for puerarin,-3.04(-5.75,-0.34)for baicalein,and-1.94(-2.95,-0.92)for luteolin.The network Meta-analysis showed no statistically significant differences between the effect sizes of the flavonoids.CONCLUSION:The results of the Meta-analysis showed that naringenin,kaempferol,puerarin,baicalein,and luteolin all have clear hypoglycemic effects in rat diabetes models,highlighting their therapeutic potential for preventing and treating diabetes mellitus in clinical practice.

naringenin is the aglycone of naringin with anti-inflammatory, antioxidant, anti-tumor, anti-atherosclerotic, and antibacterial activities. Current research supports naringenin as a new generation of immune modulators, which is of great significance for preventing and treating T cell-mediated autoimmune diseases. This study aimed to detect the possible effects and molecular mechanisms of naringenin against T cell-mediated autoimmune diseases, including multiple sclerosis(MS), inflammatory bowel disease(IBD), and rheumatoid arthritis(RA), by network pharmacology. Methods Firstly, the target of naringenin was predicted by the Traditional Chinese Medicine Systems Pharmacology(TCMSP), Swiss Target Prediction, the Comparative Toxicogenomics Database(TCD), and the diseases target were studied by GeneGards and DisgeNet database. Then obtain target gene information in UniProt database, submit to String, DAVID database for PPI, GO, and KEGG analysis. The network was visualized by Cytoscape 3.72 software. Finally, the ligands of the core targets were determined from the PDB database, and the molecular docking with naringenin was performed using PyMol and AutoDock software. Results Dietary naringenin ameliorates EAE clinical symptoms via inhibiting T cell proliferation and T cell differentiation, and reducing the CNS cell infiltration by blocking chemokine-mediated migration of DCs and pathogenic T cells into the CNS. The predition of DAVID enrichment analysis, PPI protein networkinteraction analysis, and molecular docking found that a total of sixteen common genes were observed,among which the core gene was PTGS2, ESR1, CAT, CASP3, MAPK1, AKT1, and thepossible molecular mechanism is related to the signaling pathways of HIF-1, estrogen, TNF, and NF-kB. Conclusion This study provided a new strategy and possible molecular mechanism for naringenin in treating T cell-mediated autoimmune diseases through network pharmacological studies. Protective effect of naringenin on acetic acid-induced ulcerative colitis in rats

AIM:To evaluate the ameliorative effect of naringenin(NG)during ulcerative colitis(UC)in rats.METHODS:Rats were treated with three different doses(25,50 and 100 mg/kg per day)of NG and a single dose of mesalazine(MES,300 mg/kg per day)for seven days prior to ulcerative colitis induction by4%acetic acid(AA).Twenty four hours after AA rectal administration,animals were scarified and the colonic tissues were dissected.Colonic mucus content was estimated using Alcian blue dye binding technique.In colon tissues,levels of total glutathione sulphadryls(T-GSH),non-protein sulphadryls(NP-SH)and thiobarbituric acid reactive substances(TBARS)were evaluated.The activities of the antioxidant enzymes,catalase(CAT)and superoxide dismutase(SOD)were measured.Concentrations of nucleic acids(DNA and RNA)and total protein were also estimated in colon tissues.Colonic levels of tumor necrosis factor-(TNF-),interleukin-1(IL-1),interleukin-6(IL-6),prostaglandin E2(PGE2)and nitric oxide(NO)were estimated.In cross section of colitis tissue the histopathological changes were observed.RESULTS:Colonic mucus content was decreased in AA compared to controls(587.09±65.59 mg/kg vs941.78±68.41 mg/kg,Pnaringenin supplementation,significantly and dose dependently increased the colonic mucus content.The elevated TBARS levels were significantly decreased(39.35±5.86nmol/L,P<0.05;26.74±3.17 nmol/L,P<0.01 nmol/L and 17.74±2.

Chemical composition and antifungal potential of apricot,sour cherry,and cherry tree bio-products(resins)against food-borne molds

In this study,the chemical components of ethanol extracts from the resins of apricot(Prunus armeniaca L.),sour cherry(Prunus cerasus L.)and cherry(Prunus avium L.)trees were investigated.The extracts'antifungal activity,minimal inhibitory concentration(MIC)and minimum fungicidal concentration(MFC)values were determined on seven different food-borne mold species.naringenin was the most abundant phenolic compound in Prunus cerasus and Prunus avium resin extracts,while chlorogenic acid had the highest concentration in Prunus armeniaca resin extracts.The highest antifungal activity and the lowest MIC and MFC values were in the sour cherry tree resin's ethanol extracts(P<0.05).The highest antifungal activity was in the sour cherry tree resin's ethanol extract with a 19.24 mm zone diameter on Aspergillus flavus.In contrast,the lowest effect was on Cladosporium cladosporioides with an 8.00 mm zone diameter in apricot tree resin's ethanol extracts(P<0.05).The lowest MIC and MFC values were recorded using the sour cherry tree resin's ethanol extracts on Rhizopus nigricans(16.6μg/mL)and Aspergillus flavus(47.3μg/mL)respectively.Therefore,apricot,sour cherry,and cherry tree resins extracts can be used as natural antifungals against molds.

The use of bioactive compounds as alternative medicine in the treatment of some intoxication cases is growing in the last years.Flavonoids are one of these bioactive compounds that are plant-based dietary nutrients.naringenin is the most influential flavonoid wherein it is found in citrus fruits such as orange,grapefruit,and mandarin.A lot of studies showed the ability of naringenin to scavenge free radicals in different body tissues preventing oxidative stress toxicity and lipid peroxidation.Therefore,naringenin can protect many body organs and systems against toxic manifestations of many drugs and toxic agents.So,it prevents the liver,kidney,heart,testes,and neurological system from toxic effects of a wide range of toxicants.Moreover,it can also ameliorate manifestations of cytotoxicity and developmental toxicity based on its efficacy as an antioxidant.

Flavonoid scutellarin positively regulates root length through NUTCRACKER

Exploring approaches to regulate meristem is of special importance and broad interest.In this study,we found that the flavonoid scutellarin,which has a 6-hydroxyl and a 7-glucoside,increased root length through the transcription factor NUTCRACKER(NUC).This root lengthening disappeared in NUCknockout and reappeared in NUC-rescue plants.Scutellarin induced NUC expression and promoted the division of cortex/endodermal initials.In contrast,naringenin,which has same chemical backbone but without 6-hydroxyl and with 7-hydroxyl group,showed the opposite or no effects.Our results demonstrate that scutellarin promotes root length through NUC-mediated regulatory pathways and reveal that flavonoids with and without the 6-hydroxyl and 7-glucoside have positive and negative effects on meristem size,respectively。

Autoimmune disease is prevalent and the conventional therapies have limited efficacy and often come with significant side effects.Nutrition represents an alternative and complementary approach to improving the autoimmune disorders.naringenin,a flavonoid found in citrus fruits,has been shown to have anti-inflammatory and antioxidant properties.Using the experimental autoimmune encephalomyelitis(EAE) induced in mice,a rodent model of human multiple sclerosis,we determined effect of dietary naringenin(0.5%) on autoimmune disease.We found that narigenin reduced the incidence,delayed the onset,and attenuated symptoms of EAE,which were accompanied by the reduced immune cell infiltration and demyelination in spinal cord.Additionally,the pro-inflammatory CD4+ T cell subsets Th1,Th9,and Th17 cells together with their respective transcription factors T-bet,PU.1,and RORγt were reduced in both central nervous system(CNS) and lymph nodes of EAE mice fed naringenin while no difference was found in Th2 and CD4+CD25+FoxP3+regulatory T cell(Treg) populations in either CNS or lymph nodes between two groups.We further showed that pathologic T cell proliferation induced by ex vivo re-stimulation with MOG

＜正＞Purpose:Vascular smooth muscle cell（VSMC） proliferation and migration,which is triggered by various inflammatory stimuli,contributes importantly to the pathogenesis of atherosclerosis and restenosis.naringenin is a citrus flavonoid with both lipid-lowering and insulin-like properties.Here,we investigated whether naringenin affects TNF-a-induced VSMC proliferation and migration and if so,whether heme oxygenase-1（HO-1） is involved.Methods: Rat VSMCs were pretreated with naringenin followed by TNF-αstimulation.Cell proliferation rate was determined by MTT and direct cell counting assays.Migration was assessed by Transwell and scratch wound motility assays.Real-time RT-PCR and Western blot were applied to quantify mRNA and protein levels.Generation of reactive oxygen species（ROS） was measured using DCF-DA staining.Results:We found that TNF-a down-regulated HO-1 mRNA and protein levels,as well as its activity,which was reversed by naringenin pretreatment. naringenin inhibited TNF-α-induced VSMC proliferation and migration in a dose-dependent manner.Mechanistic study demonstrated that the effects of naringenin on VSMC proliferation and migration result from its capacity to prevent ERK/MAPK and Akt phosphorylation. naringenin also blocked ROS generation.More importantly,the specific HO-1 inhibitor ZnPP IX partially abolished the beneficial effects of naringenin on VSMCs.Conclusion:These results suggest that naringenin may serve as a novel drug in the treatment of these pathologies by inducing HO-1 expression/activity and subsequently decreasing VSMC proliferation and migration.

naringenin, a citrus fruit-derived phytochemical, has been reported to exert valuable pharmacological properties as antitumor, anti-inflammatory, and anti-oxidant agent. However, little is known about naringenin on immune responses. In this study, we conducted an investigation to evaluate naringenin as an anti-T lymphocyte proliferation agent using CD3/CD28-activated lymphocytes isolated from C57BL/6 mice. We found that administration of naringenin inhibited CD3/CD28-activated lymphocyte proliferation and cell division progression in a dose dependent manner, but the inhibition at a wide range of doses used in this study did not induce cytotoxicity. In addition, naringenin dose dependently decreased the CD3/CD28-activated T lymphocyte secreting T helper(Th)1 cytokines(interleukin(IL)-2 and interferon(IFN)-γ);whereas, naringenin could not change Th2 cytokine(IL-4). Further mechanistic study demonstrated that naringenin doses dependently suppressed T cell early and middle/late activation marker CD69 and CD25 expression, respectively. Finally, naringenin could suppress cell cycle progression which contributes to inhibiting T cell proliferation and cell division and was associated with reducing the degradation of P27kip1. These findings indicate that naringenin exhibits anti-inflammatory properties via inhibiting T cell proliferation and their inflammatory cytokine secretion that may mediate the interaction between T cells and autoimmune disorders, suggesting that naringenin is a potential food-derived compound with a benefit to those with abnormally over-activation T cell mediated response and chronic inflammation such as autoimmune and inflammatory diseases.

Disrupted balance in the lineages of CD4 T cell subsets, including pro-inflammatory T helper(Th) cells and anti-inflammatory regulatory T cells(Treg), is a primary pathogenic factor for developing autoimmunity. We have found that this immunomodulatory effects of naringenin on effector T cells and T-cell mediated experimental autoimmune encephalomyelitis. We therefore explored the effects of naringenin on the development of different effector CD4 T cells. Na?ve CD4 T cells were differentiated under respective Th1, Th2, Th17, and Treg polarizing conditions with naringenin. Percent populations of each differentiated CD4 T cell subsets were determined and the corresponding regulating pathways were investigated as underlying mechanisms. naringenin mainly inhibited CD4 T cell proliferation and differentiation to Th1 and Th17, but did not affect Th2 cells. Impeded Th1 polarization was associated with inhibition of its specific regulator proteins T-bet, p-STAT1 and p-STAT4 by naringenin. Likewise, Th17 regulator proteins RORγt, p-STAT3 and Ac-STAT3 were also inhibited by naringenin. In addition, naringenin15 promoted Treg polarization and also prevented IL-6-induced suppression of Treg development via down-regulation of p-Smad2/3 as well as inhibition of IL-6 signaling, and the latter was further supported by the in vivo results showing lower soluble IL-6 R but higher soluble gp130 levels in plasma of naringenin-fed compared to the control EAE mice. naringenin impacts CD4 T cell differentiation in a manner that would explain its beneficial effect in preventing/mitigating T cell-mediated autoimmunity.
