Background:Antidepressants commonly used in clinical practice include selective 5-HT reuptake inhibitors(SSRIs),5-HT and NE reuptake inhibitors(SSNRIs),selective NE reuptake inhibitors(SNRIs),NE and specific 5-HT antidepressants(NaSSA),5-HT balanced antidepressants(SMAs),DA and NE reuptake inhibitors(NDRIs),*** with so many antidepressants,there needs to be good evidence to guide clinicians to make the best *** the past,there were several network meta-analysis studies comparing the efficacy of ***,limited by the methodology of network meta-analysis,these studies cannot analyze the entire time course of each drug,nor can they simultaneously examine the impact of multiple factors on drug *** addition,the differences in efficacy between drugs in these studies were presented as relative value(odds ratio).The information given in these is not intuitive because of the lack of absolute efficacy ***:A pharmacodynamic model was established to quantitatively compare the therapeutic effects of dozens of antidepressants,and to quantitatively evaluate the influencing factors of the therapeutic effects,which provided the necessary quantitative information for clinicians to make ***:Comprehensive literature search was conducted in public *** placebo controlled trials of antidepressants for the treatment of adult patients with major depression were included for *** change rate of the mental scale(HAMD and MADRS,etc.) scores compared with the baseline was the primary outcome,which can be quantitatively described by the E model.#12 E is the observed efficacy at the m time point of the j drug group in the i study,E and E are the theoretical maximum efficacy of placebo group in the i study and the j drug group in the i *** and ETare the time achieve 50% of the maximum efficacy for the placebo group in the i study and the j drug group in i study,respectively.η and ηare the inter-stu

Background:Chronic constipation is a common gastrointestinal dysfunction ***,people generally lack the awareness of diagnosis and treatment of chronic constipation,resulting in delays in the course of chronic ***,the drugs to treat constipation are mainly divided into osmotic laxatives,stimulant laxatives,chloride channel activators,guanylate cyclase C agonists,serotonin 5-hydroxytryptamine 4(5-HT) receptor agonists,ileal bile acid transporter inhibitors,***,the relevant quantitative information is scarce in the current constipation medication guidelines,and they do not clearly reflect the differences in the characteristics of therapeutic efficacy among various ***:In this study,we aimed to establish a pharmacodynamic model to quantitatively describe the time course of each drug used for chronic constipation in order to identify relevant impacting factors and to use pharmacodynamic parameters to directly reflect the characteristics of the therapeutic efficacy of each drug,especially the rebound ***:A comprehensive literature survey was conducted in the public databases,and search keywords included indications and drug *** randomized placebo controlled trials were included in the *** frequency of change from the baseline of spontaneous bowel movement(SBM) and complete spontaneous bowel movement(CSBM) was used as efficacy *** eliminate the effect of dietary and lifestyle heterogeneity on the therapeutic drug efficacy,we established models based on the difference in efficacy between a drug group and the corresponding placebo group(namely,‘true’ drug efficacy) to describe the time course of SBM and CSBM for each drug.#12 E is the observed efficacy at the time point j in the 1 drug group.E represents the theoretical maximal efficacy of the i drug group.τ is the rate of rebound in the i drug group;ε is the residual error in the i drug group at the time point j, and was weighted by t
