mitochondrial atpase is an important ATP synthase consisting of two specific functional regions, FO and F1 units.[1] mitochondrial atpase activity mainly maintains all kinds of intracellular energy metabolism balance in living organisms, which is closely related to FO units, F1 units and the proton gradient.[2] Herein, based on dynamic invertible ICT, we have reported a cancer cell-targeted fluorescence probe（OPM） for mitochondrial atpase *** is designed as a nucleotide analogue, that is, an ATP-derivative, since mitochondrial atpase usually catalyzes the decomposition and synthesis of phosphoric anhydride derivatives in very short physiological processes. 1-Oxo-1 H-phenalene-2,3-dicarbonitrile, a molecule with a rigid polycyclic aromatic skeleton, was chosen to be the fluorophore, on which adenosine was introduced to form an intramolecular hydrogen bond with the 6-amino groupDue to its abundant response signals, OPM could real-time monitor mitochondrial atpase activity during the cancer apoptosis process, successfully.

Jatonik polyherbal mixture induced rat liver MMPT pore opening in normal Wistar rat:In vitro and in vivo studies

Objective:To assess acute toxicity,the in vitro and in vivo effects of methanol and ethyl acetate extracts(JME and JEE)of Jatonik polyherbal mixture on some mitochondria-related parameters and their effect on the activity of some liver ***:Acute toxicity of JME and JEE was determined using Lorke’s *** vitro and in vivo opening of the mitochondrial membrane permeability transition pore(MMPT pore)was spectrophotometrically *** of malondialdehyde(MDA)as an index of lipid peroxidation and the activity of mitochondrial atpase was evaluated in vitro and in vivo and the effect of JME and JEE on the activity of liver enzymes such as alkaline phosphatase(ALP),aspartate and alanine aminotransferase(AST and ALT)and gamma-glutamyl transferase(GGT)was also ***:JME had an LD_(50) of 3808 mg/kg b.w whereas JEE had an LD_(50) greater than 5000 mg/kg *** *** the rats have been fed with both extracts,a photomicrograph of a piece of liver tissue showed no apparent symptoms of *** the in vitro and in vivo studies,both extracts prompted intact mitochondria to open their MMPT *** compared to the control,lipid peroxide product release and atpase activity were significantly increased(Patpase activity,but an increased MDA ***:Jatonik extracts may be a promising target for drug development in diseases where there is dysregulation of apoptosis,however,further studies are needed to better clarify the molecular mechanism involved in these phenomena.

Protective effects and time course of Huangqi on early-stage free radical injury following brain trauma in rats

BACKGROUND： Huangqi （Astragalus mongholicus）, a Chinese herb, has already been included in the ＂Chinese Pharmacopoeia＂ for the treatment of ischemic cerebrovascular disease. Secondary injury following brain injury is associated with free radical production, and Huangqi possesses the ability to ameliorate free radical-mediated injury. OBJECTIVE： This study was designed to observe the correlation between anti-free-radical properties of Huangqi and early histological changes of brain tissues following traumatic brain injury. DESIGN, TIME AND SETTING： This study, a randomized, controlled, animal experiment, was performed from May 2006 to June 2007 at the Experimental Center of Science and Technology, School of Basic Science, Liaoning Medical University, Jinzhou City, Liaoning Province, China. MATERIALS： Healthy, adult, Sprague Dawley rats of either gender were included. Huangqi injection was purchased from Heilongjiang Provincial Zhenbaodao Pharmaceutical Co., Ltd., China （National License Medical Number： Z23020781）. Na^＋-K^＋-adenosine triphosphatase （atpase）, Ca^2＋-atpase, and Mg^2＋-atpase, as well as kits to measure superoxide dismutase （SOD） activity and malondialdehyde （MDA） content, were purchased from Nanjing Jiancheng Biological Reagent Company, China. METHODS： Seventy-two rats were randomly divided into three groups, with 24 rats in each group：（1） sham-operated group： rats were only exposed, but not injured; （2） model group： brain focal laceration rat models were established by free-falling. These groups were intraperitoneally injected with saline, once every 10 hours; （3） Huangqi group： rats were intraperitoneally injected with 4 mL/kg Huangqi （2 g/mL）, once every 10 hours, following brain focal laceration by free-falling. MAIN OUTCOME MEASURES： Ultrastructural changes in brain tissue were observed under an electron microscope 24 hours after injury. The water content of brain tissue was measured using the dry-wet weight method. In addition, the activity of ATP
